Advances in the Mechanism of Action of Secretory Aspartic Proteases in Candida albicans InfectionAbstract:Candida albicans is an important opportunistic pathogenic fungus in the human body, usually colonizing the skin, oral mucosa, vagina and gastrointestinal tract of healthy humans asymptomatically. It is the most common clinical pathogen of fungal bloodstream infections.Immune dysfunction in the body triggers the invasion of Candida albicans against the physiological barrier, leading to skin, mucous membrane and deep infection. In recent years, with the widespread use of drugs like glucocorticoids and immunosuppressants, the incidence and mortality of Candida albicans infections have increased significantly, reaching 46%-75%of the death rate, and still rising year by year.Secretory aspartic proteases(Saps)are invasive enzymes with protein hydrolyzing activity. As a key pathogenic factor for mucosal colonization and systemic dissemination of Candida albicans,Saps play an important role in the occurrence, development and drug resistance mechanism of Candida albicans. This article aims to review the main mechanism of Saps in the process of Candida albicans infection,in order to provide theoretical basis and new ideas for the diagnosis and treatment of Candida albicans infectious and the development of new drugs.

Preliminary Study of Psoriasis-related Cuproptosis Genes Based on Bioinformatics MethodsAbstract:Objective To screen psoriasis-related cuproptosis genes, and to explore possible regulatory mechanisms using bioinformatics methods. Methods A psoriasis dataset GSE 30999 was downloaded from the Gene Expression Omnibus(GEO)database to screen differentially expressed genes(DEGs)between lesional and non-lesional skin tissues.Gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis were carried out to identify biological functions and enriched pathways of the DEGs,respectively.Immune infiltration analysis was used to obtain the distribution of immune cells in psoriasis tissues.DEGs were intersected with previously reported cuproptosis genes, thus screening out the cuproptosis-related differentially expressed genes(CR-DEGs).Molecular docking with acitretin was utilized to verify its binding activity with CR-DEGs.The single-cell dataset GSE 230842 was used for cell clustering analysis to clarify the distribution of CR-DEGs in key cell clusters. Results A total of 2,061 DEGs were obtained.GO analysis showed that DEGs were mainly enriched in iron ion binding and fatty acid metabolic process, while KEGG pathway enrichment analysis revealed that the DEGs were significantly enriched in cytokine-cytokine receptor interactions, and PPAR signalling pathways. The results of immune infiltration indicated that the immune cells, such as CD4+T cells and γδT cells were significantly elevated in psoriasis lesions(P<0.05).There were 19 cuproptosis genes, and two CR-DEGs,namely DLAT and SLC31A1,were screened by taking an intersection.Single-cell analysis showed that they were abundantly expressed in keratinocytes. The results of molecular docking showed that acitretin well bound with these two genes, especially with SLC31A1.Conclusion DLAT and SLC31A1,as psoriasis-associated cuproptosis genes, may affect the occurrence and development of psoriasis by regulating the proliferation and differentiation of keratinocyte and promoting neovascularization.Acitretin binds strongly to SLC31A1,which may be used for the treatment of psoriasis through the cuproptosis pathway.
Study on the Expression and Clinical Significance of ARMC7 in Cutaneous Melanomas:Based on Bioinformatics MethodsAbstract:Objective To examine the expression levels, clinical significance and potentials of armadillo repeat-containing protein 7(ARMC 7)in cutaneous melanomas based on bioinformatics methods. Methods The mRNA expressions,DNA methylation and clinical data of cutaneous melanomas were obtained from the cancer genome atlas(TCGA)and the gene expression omnibus(GEO).Additionally, protein expression datasets of ARMC 7 were sourced from the human protein atlas(HPA).Differential expressions of ARMC 7 between melanoma tissues and normal tissues were compared. The diagnostic performance of ARMC 7 expression levels in cutaneous melanomas was assessed using the receiver operating characteristic(ROC)curves.Survival differences between groups with different ARMC 7 expression levels were evaluated using the Kaplan-Meier curves, and the hazard ratio(HR)was calculated using Cox regression.Mutations of the ARMC 7 gene in cutaneous melanomas were analyzed using the cBioPortal database. The methylation levels of the ARMC 7 gene were analyzed using the XENA database. The expression of ARMC 7 and its correlation with immune cell infiltration were displayed in lollipop plots.Gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analyses were conducted to illustrate the underlying functional mechanisms. Results The mRNA expression levels of ARMC 7 in melanoma tissues were significantly higher than those in the normal skin tissues and nevus cells(P<0.001),showing a high diagnostic efficacy(area under the curve[AUC]=0.712).Cutaneous melanoma patients with high expression levels of ARMC 7 had significantly shorter overall survival and worse prognosis than those with low levels(P=0.026).cBioPortal data analysis showed a mutation rate of the ARMC 7 gene of 2%.XENA database analysis found that the methylation levels of 6 CpG sites in the promoter region of ARMC 7 were significantly correlated with the prognosis of cutaneous melanomas.ARMC 7 expression levels were negatively correlated with most immune cells, and only positively correlated with natural killer(NK)cells.Functional enrichment analysis of GO and KEGG suggested the enrichment of chemokine signaling pathways and ubiquitin proteolysis. Conclusion ARMC 7 is highly expressed in cutaneous melanomas and is closely associated with poor prognosis, serving as a potential diagnostic and prognostic biomarker for melanomas.

Observation on the efficacy of two percutaneous absorption methods in hypertrophic lesions of extremities in patients with moderate-to-severe plaque psoriasisAbstract:Objective To explore the therapeutic effects of ultrasonic drug penetration therapy and occlusion therapy on hypertrophic lesions of extremities in moderate-to-severe plaque psoriasis. Methods A total of 90 patients with moderate-to-severe plaque psoriasis admitted to a tertiary hospital from January 2023 to December 2024 were selected and divided into the occlusion group, superconducting group, and control group, with 30 cases in each group.Hypertrophic plaque lesions on the extremities were selected as the test sites. The control group received conventional treatment and nursing measures, including oral antihistamines, intravenous infusion of anti-allergic drugs, and topical glucocorticoid ointments. The occlusive dressing group received occlusive dressing therapy in addition to topical medication as the control group. The ultrasound-mediated drug delivery group received ultrasound-mediated drug delivery therapy in addition to the same topical medication as the control group.Treatment efficacy was evaluated using the Psoriasis Area and Severity Index(PASI),plaque thickness scores, and plaque resolution time. Results PASI assessment showed that all three groups demonstrated significant reductions in PASI scores post-treatment compared to baseline(all P<0.001).No significant inter-group differences in PASI scores were observed either pre-treatment or post-treatment(all P>0.05).For limb plaque thickness scores post-treatment,the ultrasound-mediated drug delivery group(0.43±0.50 points)showed significantly greater improvement in plaque thickness compared to the occlusive dressing group(0.96±0.61 points)(P=0.001).Analysis of plaque resolution time revealed that the ultrasound-mediated drug delivery group(4.20±1.51 days)achieved significantly faster resolution than the occlusive dressing group(5.46±2.20 days)(P<0.001).Conclusion Ultrasound-mediated drug delivery therapy is significantly superior to occlusive dressing therapy in improving the degree and speed of hypertrophic plaque resolution in limbs, effectively shortening treatment duration.
