Study on the Expression and Clinical Significance of ARMC7 in Cutaneous Melanomas:Based on Bioinformatics Methods

Abstract:Objective To examine the expression levels,clinical significance and potentials of armadillo repeat-containing protein 7(ARMC 7)in cutaneous melanomas based on bioinformatics methods.Methods The mRNA expressions,DNA methylation and clinical data of cutaneous melanomas were obtained from the cancer genome atlas(TCGA)and the gene expression omnibus(GEO).Additionally,protein expression datasets of ARMC 7 were sourced from the human protein atlas(HPA).Differential expressions of ARMC 7 between melanoma tissues and normal tissues were compared.The diagnostic performance of ARMC 7 expression levels in cutaneous melanomas was assessed using the receiver operating characteristic(ROC)curves.Survival differences between groups with different ARMC 7 expression levels were evaluated using the Kaplan-Meier curves,and the hazard ratio(HR)was calculated using Cox regression.Mutations of the ARMC 7 gene in cutaneous melanomas were analyzed using the cBioPortal database.The methylation levels of the ARMC 7 gene were analyzed using the XENA database.The expression of ARMC 7 and its correlation with immune cell infiltration were displayed in lollipop plots.Gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analyses were conducted to illustrate the underlying functional mechanisms.Results The mRNA expression levels of ARMC 7 in melanoma tissues were significantly higher than those in the normal skin tissues and nevus cells(P<0.001),showing a high diagnostic efficacy(area under the curve[AUC]=0.712).Cutaneous melanoma patients with high expression levels of ARMC 7 had significantly shorter overall survival and worse prognosis than those with low levels(P=0.026).cBioPortal data analysis showed a mutation rate of the ARMC 7 gene of 2%.XENA database analysis found that the methylation levels of 6 CpG sites in the promoter region of ARMC 7 were significantly correlated with the prognosis of cutaneous melanomas.ARMC 7 expression levels were negatively correlated with most immune cells,and only positively correlated with natural killer(NK)cells.Functional enrichment analysis of GO and KEGG suggested the enrichment of chemokine signaling pathways and ubiquitin proteolysis.Conclusion ARMC 7 is highly expressed in cutaneous melanomas and is closely associated with poor prognosis,serving as a potential diagnostic and prognostic biomarker for melanomas.
