Mitochondrial-Derived Vesicles:A Novel Mitochondrial Quality Control Mechanism and Its Key Role in Disease
[Journal Article]QUE Linfeng, LI Yanni, LIU Chenxi et al.-Translational Medicine Journal2026, No.03

Abstract:Mitochondria, serving as cellular powerhouses and metabolic control centers, play a crucial role in maintaining cellular homeostasis.Mitochondrial quality control(MQC)is a core defense mechanism for preserving mitochondrial health. It regulates the relative stability of mitochondrial morphology, number, and quality through mitophagy, fission, and fusion, thereby sustaining cellular homeostasis.Mitochondrial-derived vesicles(MDVs)represent a newly identified MQC mechanism involved in regulating mitochondrial oxidative stress, widely present across diverse cell types and tissues.Under oxidative stress conditions,MDVs are extensively induced. They selectively encapsulate damaged mitochondrial components and transport them to lysosomes for degradation, thereby mediating the early clearance of damaged materials and effectively preserving mitochondrial structural and functional integrity. This review systematically summarizes the biogenesis, cargo selection, and transport pathways of MDVs,with a focus on elucidating the physiological roles of MDVs and recent research advances in their involvement in various diseases.

Exploring the Mechanism of Fuzheng Huayu Formula in Intervening Angiogenesis in Endometriosis Based on Bioinformatics and Animal Experiments
[Journal Article]ZHAO Jiajing, HUANG Yuxiao, ZHANG Yuqing et al.-Translational Medicine Journal2026, No.02

Abstract:Objective Fuzheng Huayu(FZHY)formula has clear clinical efficacy in the treatment of endometriosis(EM).Previous studies confirmed that its mechanism may be related to the inhibition of angiogenesis. This study focused on this research hotspot to explore the mechanism of FZHY formula in treating EM.Methods The study was divided into two parts. The first part involved bioinformatics analysis. We obtained the drug targets of FZHY formula, the differentially expressed genes in EM,and angiogenesis-related targets, and identified the top 6"EM core targets".The expression matrices of normal and model group samples for these"EM core targets"were obtained from the GSE58178 microarray.A differential group comparison diagram was plotted,a TF-mRNA-miRNA network was constructed, and the core targets of action were screened. The second part consisted of animal experiment validation. An EM rat model was established by allogeneic endometrial transplantation.Model rats were divided into the model group, the Chinese medicine group(FZHY group),and the positive control group, with 6 rats in each group.A sham-operated group(n=6)was set as control. After 14 days of gavage treatment, samples were collected.Serum and endometrial tissues were examined.Enzyme-linked immunosorbent assay was used to detect serum levels of carbohydrate antigen 125(CA125)and vascular endothelial growth factor(VEGF).Reverse transcription quantitative real-time polymerase chain reaction was used to measure the mRNA expression of miR-199a-5p,hypoxia-inducible factor 1α(HIF-1α),VEGF and other related molecules in normal/ectopic endometrial tissues. Results Bioinformatics analysis indicated that in the key process of angiogenesis,FZHY formula may interfere with EM angiogenesis and slow the growth of ectopic lesions by downregulating HIF-1α,VCAM1,STAT3,and SERPINE1,and upregulating MMP2 and PDGFRB.Subsequently, through construction of the"TF-mRNA-miRNA network"and verification by animal experiments, compared with the model group, the Chinese medicine group showed significantly lower serum CA125 and VEGF levels, significantly reduced volume of ectopic lesions, increased expression of miR-199a-5p,and decreased expression of HIF-1α and VEGF[(9.99±1.01)U/mL vs(14.60±0.65)U/mL,P<0.05;(170.04±29.40)pg/mL vs(245.42±36.10)pg/mL,P<0.05;(6.41±2.11)mm³ vs(23.92±4.63)mm³,P<0.05;0.80±0.08 vs 0.02±0.01,P<0.05;1.28±0.06 vs 2.21±0.32,P<0.05;1.29±0.23 vs 3.79±0.80,P<0.05].Conclusion FZHY formula may intervene in the key process of angiogenesis by downregulating the expression of factors such as HIF-1α and VEGF,thereby shrinking EM ectopic lesions and delaying the further progression of EM.

[Journal Article]LIU Lili, AN Ping, ZHANG Jia et al.-Translational Medicine Journal2026, No.02

Abstract:Objective To explore the risk factors of catheter-related bloodstream infections(CRBSI)in neonates and predict the risk of CRBSI associated with central venous catheterization. Methods Clinical records of 386 neonates who underwent central venous catheterization at Tangshan Maternal and Child Health Hospital from January 2020 to December 2024 were collected, and the neonates were divided into the CRBSI group(37 cases)and the non-CRBSI group(349 cases)based on whether CRBSI occurred. The t-test and χ2 test were used to compare clinical data between the two groups.Multivariate logistic stepwise regression analyzed influencing factors for neonatal CRBSI,and a nomogram prediction model was constructed. Results Among 386 neonates with central venous catheters,37 developed CRBSI(9.59%),with 42 pathogenic strains isolated(predominantly gram-positive bacteria,57.14%).Risk factors for neonatal CRBSI included:birth weight<1.5 kg(OR=0.523,95%CI:0.381-0.716),low 5-minute Apgar score(OR=0.393,95%CI:0.263-0.586),≥2 puncture attempts(OR=2.401,95%CI:1.629-3.540),catheter indentation time≥2 weeks(OR=2.832,95%CI:1.865-4.299),and elevated procalcitonin levels(OR=2.479,95%CI:1.669-3.684).The nomogram model showed an AUC of 0.896(95%CI:0.844-0.948)for predicting CRBSI,with 67.53%specificity and 92.04%sensitivity.Conclusion The nomogram model based on risk factors for neonatal CRBSI demonstrates good predictive performance and can serve as an important tool for guiding clinical treatment.

Aloin may Promote the Proliferation of Human Gingival Fibroblasts by Upregulating EFNA1
[Journal Article]WU Tianzhen, ZHAO Hua, ZHOU Siwei et al.-Translational Medicine Journal2026, No.02

Abstract:Objective To preliminarily explore the targets and pathways through which aloin promotes the proliferation of human gingival fibroblasts. Methods A total of 10 cases of gingival tissue trimming from impacted tooth extraction surgeries in the Department of Oral and Maxillofacial Surgery at Hebei Medical University Hospital were randomly selected from January 2024 to January 2025.Human gingival fibroblasts were cultured using the tissue explant adherence method;third-passage cultured cells were selected and placed on glass slides for cell identification.Fibroblasts treated with 40 μmol/L aloin were used as the experimental group, and untreated fibroblasts served as the control group, with three samples in each group.Transcriptome sequencing was performed on both groups to identify differentially expressed genes and conduct KEGG pathway enrichment analysis. Results The sequencing results showed that the expression of ephrin-A1(EFNA1)in the experimental group(0.951 386,0.918 644,0.855 717)was significantly higher than that in the control group(0.207 686,0.274 180,0.238 854),and the difference was statistically significant(P<0.001).KEGG Enrichment analysis of differentially expressed genes revealed that, when ranked by P-value,these genes were significantly enriched in the phosphatidylinositol 3-kinase-protein kinase B pathway. Moreover, the EFNA1 gene is located upstream of this pathway and showed high expression. Conclusion Aloin may promote the proliferation of human gingival fibroblasts by upregulating EFNA1.

Correlation Between Serum CCL2 and CXCL8 Levels and In-Stent Restenosis after PCI in Patients with Coronary Heart Disease
[Journal Article]SUN Ning, GUAN Xin-Translational Medicine Journal2026, No.02

Abstract:Objective To investigate the correlation between serum levels of C-C motif chemokine ligand 2(CCL2)and C-X-C motif chemokine ligand 8(CXCL8)and in-stent restenosis(ISR)after percutaneous coronary intervention(PCI)in patients with coronary heart disease(CHD).Methods A total of 202 CHD patients who were admitted to Baoji People's Hospital and underwent PCI from July 2019 to July 2020 were enrolled.Based on the occurrence of ISR within 18 months after PCI,the patients were divided into an ISR group(32 cases)and a non-ISR group(170 cases).Clinical data were collected, and serum levels of CCL2 and CXCL8 were measured.Multivariate logistic regression analysis was used to identify independent influencing factors for ISR after PCI in CHD patients.Relative risk analysis was conducted to determine the impact of high versus low levels of CCL2 and CXCL8 on ISR.Receiver operating characteristic(ROC)curves were plotted to analyze the predictive value of CCL2 and CXCL8 levels for ISR.Results The proportion of diabetes,low-density lipoprotein cholesterol(LDL-C)levels,and serum levels of CCL2 and CXCL8 were significantly higher in the ISR group than in the non-ISR group(P<0.05).Multivariate logistic regression analysis showed that elevated LDL-C(OR=2.305,95%CI:1.202-4.419),elevated CCL2(OR=2.643,95%CI:1.114-6.273),and elevated CXCL8(OR=2.921,95%CI:1.236-6.906)were risk factors for ISR after PCI in CHD patients(P<0.05).Relative risk analysis showed that the risk of ISR in patients with high serum levels of CCL2 and CXCL8 was 2.136 times and 2.614 times higher, respectively, than that in patients with low levels.ROC curve analysis revealed that the area under the curve(AUC)for predicting postoperative ISR in CHD patients was 0.739 for CCL2 and 0.682 for CXCL8.The combined prediction of both markers yielded an AUC of 0.820,which was higher than that of either single indicator(Zcombined vs CCL2=2.031,P=0.042;Zcombined vs CXCL8=2.786,P=0.005).Conclusion Serum levels of CCL2 and CXCL8 were elevated in CHD patients with ISR.Both are independent risk factors for ISR after PCI in CHD patients and have good predictive value for ISR after PCI.
