Mitochondrial-Derived Vesicles:A Novel Mitochondrial Quality Control Mechanism and Its Key Role in Disease

Abstract:Mitochondria,serving as cellular powerhouses and metabolic control centers,play a crucial role in maintaining cellular homeostasis.Mitochondrial quality control(MQC)is a core defense mechanism for preserving mitochondrial health.It regulates the relative stability of mitochondrial morphology,number,and quality through mitophagy,fission,and fusion,thereby sustaining cellular homeostasis.Mitochondrial-derived vesicles(MDVs)represent a newly identified MQC mechanism involved in regulating mitochondrial oxidative stress,widely present across diverse cell types and tissues.Under oxidative stress conditions,MDVs are extensively induced.They selectively encapsulate damaged mitochondrial components and transport them to lysosomes for degradation,thereby mediating the early clearance of damaged materials and effectively preserving mitochondrial structural and functional integrity.This review systematically summarizes the biogenesis,cargo selection,and transport pathways of MDVs,with a focus on elucidating the physiological roles of MDVs and recent research advances in their involvement in various diseases.
