Study on LncRNA XIST Promoting Proliferation of Colorectal Cancer Cells by Competitively Binding to miR-497-5p and Targeting EIF5AAbstract:Objective To explore the role of lncRNA XIST in colon cancer and analyze whether it regulates the proliferation by competitively binding to microRNA-497-5p(miR-497-5p)targeting EIF5A,in order to provide ideas for exploring the mechanism of colon cancer.Methods Bioinformatics analysis was used to predict the expression trend of EIF5A in colon cancer and predict possible related target genes.Colon cancer SW480 cell line was selected,plasmids were constructed,and pc-DNA-XIST group,si-XIST group,miR-497-5p mimic group,miR-497-5p inhibitor group and si-EIF5A group were established respectively.Empty vector transfection(EV)group was also set up.Experimental methods such as CCK-8,qRT-PCR and dual luciferase reporter gene were applied,and rescue experiments were performed.Forty-seven patients undergoing colon cancer surgery were selected.Tumor tissues were collected as the observation group,and normal mucosal tissues were collected as the control group.The expression of LncRNA XIST and miR-497-5p was detected by qRT-PCR,and the expression of EIF5A was detected by immunohistochemistry.Results Bioinformatics analysis predicted that LncRNA XIST had a high expression trend in colon cancer.LncRNA XIST and miR-497-5p,miR-497-5p and EIF5A had binding base sequences.Compared with the EV group,the cell proliferation activity of the si-XIST group,miR-497-5p mimic group and si-EIF5A group was significantly decreased,and the cell proliferation activity of the pc-DNA-XIST group and miR-497-5p inhibitor group was significantly increased.The dual luciferase reporter gene experiment showed that LncRNA XIST and miR-497-5p,miR-497-5p and EIF5A had a targeting relationship.Rescue experiments showed that inhibition of miR-497-5p could partially reverse the inhibitory effects of silencing LncRNA XIST and silencing EIF5A on colon cancer cell proliferation.LncRNA XIST and EIF5A expression were increased in colon adenocarcinoma,while miR-497-5p expression was downregulated.Conclusion LncRNA XIST plays a promoting role in the formation and progression of colon cancer.LncRNA XIST promotes the proliferation of colon cancer cells by competitively binding to miR-497-5p and targeting EIF5A.
The Safety of IDegLira Versus IdegAsp in T2DM Patients with Poor Glycemic ControlAbstract:Objective To observe the clinical efficacy and safety of IDegLira and IdegAsp in patients with type 2 diabetes mellitus(T2DM)who have poor glycemic control.Methods A total of 100 patients with type 2 diabetes who visited the outpatient department or were hospitalized in the Endocrinology Department of the Third Affiliated Hospital of Jinzhou Medical University from December 2022 to December 2024 and had poor blood sugar control despite using one or more oral hypoglycemic drugs were selected.They were randomly divided into the experimental group and the control group,with 50 cases in each group.The experimental group was given IDegLira injection by subcutaneous injection,once a day.The initial dose was 10 U per day.The dosage of IDegLira was adjusted daily according to the changes in the patient's fasting blood glucose level.Each adjustment was 2 to 4 U.The control group was given IdegAsp,with an initial dose of 0.4~0.5 U/kg/d.The insulin dose was evenly distributed 1∶1 before breakfast and dinner.The dose of IdegAsp for the next morning's breakfast was adjusted based on the blood glucose level before dinner the previous day,and the dose for the next evening's dinner was adjusted based on the blood glucose level before breakfast on that day.Each adjustment was 2-4 U.The observation period was 12 weeks.Waist circumference,body weight,body mass index(BMI),blood pressure,fasting blood glucose,2-hour blood glucose,glycosylated hemoglobin,fasting serum C-peptide,serum 2-hour C-peptide,triglycerides,total cholesterol,high-density lipoprotein cholesterol,low-density lipoprotein cholesterol,visceral fat content,the proportion of patients with blood glucose control,and adverse reactions were measured before treatment and at the 12th week after treatment.Results(1)After treatment,the waist circumference,BMI,and visceral fat of the experimental group were significantly lower than those of the control group,with a statistically significant difference(P<0.05).(2)After treatment,fasting blood glucose(FBG),2 h PG,and HbA1c in both groups decreased significantly compared with those before treatment,and the differences were statistically significant(P<0.01);however,there was no statistically significant difference between the two groups(P>0.05).There was no statistically significant difference in the proportion of patients who achieved the blood glucose target(P>0.05).(3)After treatment,the body weight,BMI,systolic blood pressure,triglycerides,total cholesterol,low-density lipoprotein cholesterol,and visceral fat content of the experimental group were lower than those before treatment,with statistically significant differences(P<0.05),while its high-density lipoprotein cholesterol was higher than that before treatment,with a statistically significant difference(P<0.05).In the control group,the high-density lipoprotein cholesterol after treatment was higher than that before treatment,and the difference was statistically significant(P<0.05).(4)In the control group,3 cases of mild hypoglycemia occurred;in the experimental group,2 cases of gastrointestinal reactions occurred.There was no significant difference in the incidence of adverse reactions between the two groups(P>0.05).Conclusion Both IDegLira and IdegAsp can significantly reduce blood glucose levels in T2DM patients with poor glycemic control.IDegLira can reduce waist circumference,decrease body weight,lower blood pressure,improve blood lipids,and reduce visceral fat content.Additionally,it has a lower incidence of hypoglycemia and high safety,making it worthy of clinical promotion and application.
Study on the Mechanism of SIRT2 Regulating Aerobic Glycolysis and Growth Proliferation of Colorectal Cancer CellsAbstract:Objective To explore the biological function of the silent information regulator factor 2(SIRT2)in the metabolic reprogramming and proliferation regulation of colorectal cancer cells,as well as its impact on the glucose metabolism pathway,At the same time to study the protein expression levels of SIRT2 and hexokinase Ⅱ(HK Ⅱ)in colorectal cancer and their correlation.Methods Western Blot was used to analyze the protein expression level of SIRT2 in colorectal cancer cells.Based on the results,cell lines with high SIRT2 expression were selected for gene interference,while cell lines with low SIRT2 expression were used for overexpression studies.The glucose concentration in the culture medium was measured using the glucose oxidase method,and lactate levels were analyzed colorimetrically to study the metabolic state of the cells.Cell viability was detected by CCK-8 assay,and proliferative capacity was assessed by colony formation assay.Additionally,reverse transcription PCR(RT-PCR)was used to study the effect of SIRT2 interference or overexpression on HK Ⅱ mRNA expression levels,and Western Blot was performed to further validate the effect of SIRT2 regulation on HK Ⅱ protein expression.Immunohistochemistry was used to detect the protein expression levels of SIRT2 and HK Ⅱ in colorectal cancer tissues.Rescue experiments were conducted by co-transfecting SIRT2-siRNA and HK Ⅱplasmids,followed by measurements of glucose and lactate levels in the culture medium,as well as CCK-8 and colony formation assays for each group.Results After RNA interference,compared with the negative control group,the interference group exhibited suppressed glycolysis,characterized by increased glucose levels and decreased lactate production,along with significantly reduced cell proliferation activity and colony formation ability.Both HK Ⅱ mRNA and protein expression levels decreased.After overexpression experiments,compared with the control group,overexpression significantly enhanced aerobic glycolysis in colorectal cancer cells,manifested as increased glucose consumption and lactate accumulation in the culture medium,while also promoting cell growth,proliferation,and colony formation efficiency.Both HK Ⅱ mRNA and protein expression levels increased.Immunohistochemistry results showed that the expression levels of SIRT2 and HK Ⅱ in colorectal cancer tissues were significantly higher than those in paired adjacent tissues.In the rescue experiments,compared with the interference group,the co-transfection group showed significantly decreased glucose levels,significantly increased lactate levels,and markedly enhanced cell proliferation activity and colony formation rate.Conclusion SIRT2 promotes aerobic glycolysis and cell growth/proliferation in colorectal cancer cells,and the mechanism may be related to SIRT2 regulating HK Ⅱ gene expression.
Study on ICAM1 Influencing the Function of Drug-resistant Cells in Triple-negative Breast CancerAbstract:Objective To investigate the changes in cellular functions by which the ICAM1 gene regulates chemotherapeutic resistance to doxorubicin/paclitaxel in triple-negative breast cancer(TNBC).Methods TNBC samples without preoperative neoadjuvant chemotherapy and those with preoperative neoadjuvant chemotherapy were collected.Immunohistochemistry(IHC)and Western Blot(WB)were used to detect ICAM1 expression.qRT-PCR and WB were applied to detect ICAM1 expression in breast cancer cells.Chemoresistant cell lines were established by treating TNBC cells with combined doxorubicin and paclitaxel.Lentivirus vectors for human ICAM1 knockdown were constructed and infected into resistant cells.Cell proliferation was evaluated by CCK-8 and Edu assays.Cell migration and invasion were detected by Transwell assays.Western Blot was performed to detect the expression of ferroptosis-related proteins(GPX4,ACSIA,SLC7A11),breast cancer resistance protein ABCG2,and EMT-related proteins(E-cadherin,N-cadherin,Vimentin).Cellular ATP levels were measured using an ATP kit,and lipid peroxidation levels were detected by DCHF-DA probe.Results Lentivirus-mediated ICAM1 knockdown in MDA-MB-231 sensitive and resistant cell lines showed that ICAM1 promoted cell viability,proliferation,migration,and invasion;enhanced the expression of breast cancer resistance protein ABCG2;inhibited ferroptosis;and promoted the EMT process.Conclusion This study reveals that ICAM1 promotes the proliferation and invasion of breast cancer cells by regulating cellular metabolism,EMT process,and ferroptosis pathway.Meanwhile,ICAM1 contributes to chemoresistance by upregulating the expression of breast cancer resistance protein ABCG2.
Predictive Value of MAP in Surgical Risks of Robot-Assisted Partial Nephrectomy(RAPN)for T1a Renal TumorsAbstract:Objective This study aims to deeply explore the clinical practical value of the Mayo Adhesive Probability(MAP)score in the preoperative evaluation of laparoscopic partial nephrectomy(LPN).It breaks through the limitation of traditional studies that only focus on tumor-specific characteristics and emphasizes the crucial but often overlooked factor of perirenal fat adhesion.Methods Through an in-depth retrospective analysis of the detailed data of 116 patients who underwent LPN in the Department of Urology of the First Affiliated Hospital of Jinzhou Medical University,the patients were divided into a low-score group(0~3 points,76 cases in total)and a high-score group(4~5 points,40 cases in total)according to the MAP scoring criteria.The MAP score was innovatively correlated with multiple peri-operative indicators to precisely explore the internal relationships between the MAP score and key peri-operative indicators such as operation time and intraoperative blood loss.Results For the 116 patients,the average operation time was(140.51±42.31)minutes,the intraoperative blood loss was(176.73±83.57)milliliters,and the renal warm ischemia time was(23.55±5.36)minutes.Compared with the low-MAP-score group,patients in the high-score group had a higher body mass index(BMI),a longer operation time,and more intraoperative blood loss.However,there were no statistically significant differences in the intraoperative blood transfusion rate,the proportion of Clavien-Dindo grade Ⅱ surgical complications,and the serum creatinine level before discharge.The MAP score was significantly correlated with the operation time and blood loss of LPN and was an independent predictor of the operation time.Conclusion The MAP score can predict the operation time and blood loss of LPN,providing a new perspective and strong evidence for clinical accurate prediction of surgical conditions.This study also clearly indicates that large-sample and multi-center studies are needed for further verification,pointing out the direction for follow-up research.

Study on Optimization of Extraction Technology of Metaplexis japonic Total Flavonoids by Response Surface Methodology and Its Antioxidant ActivityAbstract:Objective To optimize the extraction process of total flavonoids extracted from Metaplexis japonic and study its antioxidant properties,providing a reference for improving its resource utilization.Methods Ultrasonic extraction method was used to extract total flavonoids from Metaplexis japonic.Through a single factor experimental design,the important factors affecting the extraction rate of total flavonoids from Metaplexis japonic,including ethanol volume fraction,solid-liquid ratio,time and ultrasonic power were evaluated.The Box Behnken center combination design was used to optimize the optimal parameter levels of the four significant influencing factors.Results The optimal extraction process for total flavonoids from Metaplexis japonic was 70.00%ethanol volume fraction,a solid-liquid ratio of 1∶15,time of 30 minutes and ultrasonic power of 195 W.Under these conditions,the extraction rate of total flavonoids from Metaplexis japonic was 10.78%.The antioxidant activity experiment showed that the highest scavenging rates of total flavonoids from Metaplexis japonic on DPPH and ABTS·+free radicals were 79.28%and 88.62%,respectively.Conclusion The optimized extraction process of total flavonoids from Metaplexis japonic is feasible;the antioxidant activity experiment proves that it has good antioxidant activity.
Optimization of Ultrasonic-assisted Enzymatic Extraction Process for Alkaloids from Metaplexis japonic and Its Anti-tumor Activity AnalysisAbstract:Objective To optimize the extraction process of alkaloids from the Metaplexis japonic and study its anti-tumor activity.Methods Using the Metaplexis japonic as the raw material,ultrasound-assisted enzymatic hydrolysis was used to extract alkaloids from Metaplexis japonic.The extraction rate of alkaloids from Metaplexis japonic was used as the indicator,and single factor experiments and response surface methodology were used to design optimization experiments to explore the optimal extraction process of alkaloids from Metaplexis japonic.Results The optimal process for extracting alkaloids from Metaplexis japonic was achieved with an enzyme concentration of 530 U/mL,enzymatic hydrolysis time of 63 min,enzymatic hydrolysis temperature of 51 ℃,and ultrasound power of 140 W.At this time,the extraction rate of alkaloids from Metaplexis japonic was 10.40%.The cell viability of MCF-7 decreases with the increase of the concentration and duration of action of alkaloids from Metaplexis japonic.Conclusion The above results indicate that ultrasound-assisted enzymatic hydrolysis can improve the extraction rate of alkaloids from Metaplexis japonic.The optimized extraction process conditions of this method are feasible and reliable.
Mechanistic Study of Xuefu Zhuyu Capsules in the Treatment of Acetaminophen-induced Liver injuryAbstract:Objective To explore the mechanism of Xuefu Zhuyu capsules(XFZYC)in the treatment of acetaminophen(APAP)-induced liver injury through network pharmacology,molecular docking and experimental verification.Methods The active components of Xuefu Zhuyu Capsule were screened through the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)and the herbal medicine database(HERB).Their corresponding targets were predicted and selected through the SwissADME and Swiss Target Prediction databases.The relevant targets of drug-induced liver injury were obtained from the Human Genome Database(GeneCards),the Online Mendelian Inheritance in Man(OMIM)database,and the Database of Gene-Disease Associations(DisGeNET).The intersection targets of drugs and diseases were imported into the STRING database to construct a protein-protein interaction(PPI)network model.Topological parameter analysis was conducted in Cytoscape 3.9.1.Gene Ontology(GO)analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis of the intersection targets were carried out using the database for annotation,visualization and integrated discovery(DAVID).Molecular docking verification was performed using the CB-Dock2 online molecular docking server.Animal experiments verified the prediction results of network pharmacology.A mouse model of drug-induced liver injury was established using APAP,and different doses of Xuefu Zhuyu Capsules were administered for intervention.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of liver tissue.Serum biochemical indicators such as alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP),and total bilirubin(TBIL)were measured by the microplate method.The levels of oxidative factors such as superoxide dismutase(SOD),glutathione(GSH),catalase(CAT),malondialdehyde(MDA),and inflammatory factors such as tumor necrosis factor-α(TNF-α),interleukin-1 β(IL-1β),and interleukin-6(IL-6)were detected by enzyme-linked immunosorbent assay(ELISA).The expression of key proteins in the phosphatidylinositol 3-kinase-protein kinase B(PI3K-AKT)and mitogen-activated protein kinase(MAPK)pathways was detected by Western Blot.Results A total of 158 active components,914 potential targets,1657 drug-induced liver injury disease targets,348 intersecting targets,and 67 core targets(including protein kinase B1(AKT1),interleukin 6(IL6),tumor necrosis factor(TNF),tumor protein p53(TP53),interleukin 1β(IL1β),epidermal growth factor receptor(EGFR),signal transducer and activator of transcription 3(STAT3),caspase 3(CASP3),B lymphocyte tumor-2(BCL2),and proto-oncogene tyrosine kinase(SRC))were identified through PPI network analysis.The KEGG enrichment analysis indicates that the effect of Xuefu Zhuyu Capsule is mainly achieved through multiple biological pathways,including cancer-related pathways,metabolic pathways,PI3K-AKT signaling pathway,and MAPK signaling pathway,etc.The molecular docking results indicate that the main active components in Xuefu Zhuyu Capsule,namely quercetin,kaempferol,baicalein,β-sitosterol,stigmasterol,isoquercitrin,naringenin,and luteolin,have good binding with the target proteins AKT1,IL6,TNF,TP53,IL1β,and EGFR.Animal experiments show that Xuefu Zhuyu Capsule reduces the levels of serum ALT,AST,ALP,TBIL and inflammatory factors TNF-α,IL-1β,and IL-6 in liver tissue,increases the levels of oxidative factors SOD,CAT,and GSH in liver tissue,and reduces the level of MDA.Western blot results show that Xuefu Zhuyu Capsule reduces the expression levels of phosphorylated phosphatidylinositol 3-kinase(p-PI3K),phosphorylated protein kinase B(p-AKT),phosphorylated c-Jun N-terminal kinase(p-JNK),phosphorylated extracellular signal-regulated kinase(p-ERK),and phosphorylated p38 mitogen-activated protein kinase(p-P38)in the PI3K-AKT and MAPK signaling pathways.Conclusion This study,by combining network pharmacology,molecular docking and animal experiments,confirmed that the Xuefu Zhuyu Capsule exerts an anti-liver injury effect induced by acetaminophen through multiple components,multiple targets and multiple pathways.The mechanism may be to inhibit the expression of proteins related to the PI3K-AKT and MAPK signaling pathways to regulate the oxidative stress and inflammatory response caused by acetaminophen stimulation.
Relationship Between Liver Quality and Acute Rejection and Early Liver Function Recovery After Liver TransplantationAbstract:Objective To investigate the correlation between donor liver quality and the incidence of acute rejection as well as early postoperative liver function recovery in liver transplant recipients.Methods A retrospective analysis was conducted on the clinical data of 93 patients with decompensated liver cirrhosis who underwent liver transplantation at Zhengzhou People's Hospital from March 2022 to March 2025.The baseline data,surgical parameters,postoperative liver function indicators,and donor liver quality indicators of the patients were collected.The patients were divided into the rejection group(21 cases)and the non-rejection group(72 cases)based on whether acute rejection occurred within 14 days after surgery.The differences in donor and recipient-related indicators between the two groups were compared.Univariate and multivariate Cox regression analyses were used to screen independent influencing factors,and correlation analysis was conducted.Results The levels of alanine aminotransferase(ALT),alkaline phosphatase(ALP),gamma-glutamyl transferase(GGT),and total bilirubin(TB)on the third postoperative day,as well as the operation time,were all higher in the rejection group than in the non-rejection group.Meanwhile,the albumin(ALB)level on the third postoperative day in the rejection group was significantly lower than that in the non-rejection group(P<0.05);compared with the non-rejection group,the total ischemic time,cold ischemic time,graft-to-recipient weight ratio(GRWR),and the proportion of patients with fatty liver and hepatocyte edema were significantly higher in the rejection group(P<0.05);multivariate analysis of multiple logistic regression showed that ALB on the third day after surgery was a protective factor,while ALT on the third day after surgery,operation time,cold ischemia time of donor liver,and donor liver cell edema were risk factors for acute rejection in patients(P<0.05);the Pearson correlation analysis indicated that on the third day after surgery,ALB was significantly negatively correlated with the operation time,the cold ischemia time of the donor liver,the degree of cell edema of the donor liver,and GRWR;while ALT was significantly positively correlated with these indicators.The operation time was positively correlated with the cold ischemia time,cell edema,and GRWR,and the cell edema of the donor liver was also positively correlated with the cold ischemia time.Conclusion The quality of the donor liver has a significant impact on acute rejection reactions after liver transplantation and the early recovery of liver function.Optimizing the assessment of the donor liver,controlling the cold ischemia time,reducing liver histological damage,shortening the operation time,and closely monitoring liver function early on can help reduce the risk of acute rejection,improve the success rate of transplantation,and enhance the prognosis of patients.
The Impact of Soothing the Liver and Relieving Depression Method on Appetite-regulating Hormones in Patients with ObesityAbstract:Objective To explore the impact of the liver-soothing and depression-relieving therapy on the appetite-regulating hormones of obese patients based on the brain-gut axis.Methods From October 2022 to October 2024,97 obese patients who received medical treatment at Zhengzhou People's Hospital were selected as the subjects of this study.Using the random number table method,the patients were divided into the treatment group and the control group,with 49 and 48 cases respectively.The control group received conventional exercise and dietary intervention,while the treatment group received conventional intervention in addition to taking traditional Chinese medicine prescriptions for soothing the liver and relieving depression.The morphological indicators,clinical treatment effects,TCM syndrome scores,appetite-regulating hormone levels,and brain-gut axis-related indicators of the two groups were compared.Results After treatment,the body mass index(BMI)(23.82±2.68)kg/m2,weight(63.71±6.32)kg,waist circumference(88.33±5.07)cm,and hip circumference(91.61±4.73)cm of the treatment group were significantly lower than those of the control group(P<0.05);the therapeutic group(95.92%)showed significantly better efficacy than the control group(P<0.05);in terms of the integrated Chinese medicine syndrome scores,the treatment group(4.69±2.47)scored lower than the control group(P<0.05);the levels of leptin(3.27±0.18),ghrelin(3.48±0.28),and peptide YY(PYY)(2.17±0.28)in the treatment group were higher than those in the control group(P<0.05);the Substance P(SP)level in the treatment group(75.25±5.65)pg/mL was lower than that in the control group.Moreover,the neuropeptide Y(NPY)level(206.27±20.98)g/mL and the 5-hydroxytryptamine(5-HT)level(48.35±5.60)pg/mL in this group were higher than those in the control group(P<0.05).Conclusion The liver-soothing and depression-relieving therapy combined with conventional interventions effectively improves anthropometric parameters,modulates appetite-regulating hormones and brain-gut axis-related neurotransmitters,and significantly enhances clinical outcomes in obese patients,demonstrating the potential of integrated Chinese and Western medicine approaches for obesity management.
Experimental Study on Lupeol Alleviating Airway Inflammation and Mucus Hypersecretion in Mice with Chronic Asthma via Th2 CytokinesAbstract:Objective To explore the effects of lupeol(LUP)on airway inflammation and mucus hypersecretion in asthmatic mouse models.Methods Adult male mice were selected and randomly divided into 4 groups(n=8)using a random number table,including the Control group,the Ovalbumin(OVA)group(OVA group),the OVA+LUP-low-dose group(OVA+L-LUP group),and the OVA+LUP-high-dose group(OVA+H-LUP group).The mouse asthma model was established by using OVA aerosolization.After 75 days of intervention,the mice were sacrificed and the bronchoalveolar lavage fluid(BALF)and lung tissue specimens were obtained.The pathological changes and mucus secretion in lung tissue were analyzed by hematoxylin-eosin(HE)staining and periodic acid schiff(PAS)staining;the levels of interleukin 4(IL-4),interleukin 5(IL-5),and interleukin 13(IL-13)in BALF were detected by enzyme-linked immunosorbent assay(ELISA);the mRNA levels of mucin 5AC(MUC5AC),thymic stromal lymphopoietin(TSLP),IL-4,IL-5 and IL-13 in lung tissue were detected by real-time fluorescence quantitative PCR(Quantitative Real-time PCR,qPCR).Results After 75 days of OVA aerosol intervention,HE staining revealed significant inflammatory cell infiltration in the airways and surrounding tissues of asthmatic mice,while PAS staining showed a marked increase in goblet cells and mucus in the airways of asthmatic mice;the number of lymphocytes,and the number of eosinophils in BALF were all significantly increased(P<0.05);the levels of IL-4,IL-5,and IL-13 in BALF were significantly increased;the mRNA levels of MUC5AC,TSLP,IL-4,IL-5,and IL-13 in lung tissue were significantly increased(P<0.05).Compared with the OVA group,all indicators in the OVA+L-LUP group and the OVA+H-LUP group were lower(P<0.05).At the same time,compared with the OVA+L-LUP group,all indicators in the OVA+H-LUP group were lower(P<0.05).Conclusion In conclusion,lupeol can alleviate the airway inflammatory response in mice with chronic asthma induced by OVA by down-regulating the synthesis and release of Th2 inflammatory factors.This study has laid a theoretical foundation for the application of lupeol in the clinical treatment of asthma.
Relationship Between Blastocyst Trophoblast Cell Score and Single Blastocyst Transfer-assisted Live Birth RateAbstract:Objective To investigate the relationship between blastocyst trophoblast cell score and single blastocyst transfer assisted live birth rate.Methods A retrospective selection was made of 350 patients who underwent single blastocyst transfer at the Second Affiliated Hospital of Zhengzhou University from April 2022 to April 2024 as the research subjects.The live birth rate of the patients was recorded;the patients were divided into the live birth group and the non-live birth group based on whether they gave birth,and the general data of the two groups were compared;Binary Logistic Regression analysis was used to analyze the influencing factors of live birth through single blastocyst transfer in assisted pregnancy,and forest plots were used to analyze the effect values of each influencing factor;the predictive efficacy was verified using the ROC curve;and the relationship between the blastocyst trophectoderm cell score and the live birth rate after single blastocyst transfer was analyzed through Pearson correlation analysis.Results The live birth rate of 350 patients during childbirth was 46.00%.General data showed that the levels of anti-müllerian hormone(AMH),antral follicle count(AFC),trophoblast grading(TE),endometrial thickness,number of metaphase Ⅱ oocytes(MⅡ),and the proportion of fresh cycle transplantation in the live birth group were significantly higher than those in the non-live birth group.Meanwhile,the proportion of fresh cycle transplantation,basal estradiol(E2),follicle-stimulating hormone(FSH)levels,and patient age were significantly lower(P<0.05);the multivariate analysis of binary Logistic regression showed that the advanced age of the patient during pregnancy(OR=1.364,95%CI:1.113-1.615)was a risk factor for non-viability of pregnancy after single blastocyst transfer,while high levels of AMH(OR=0.595,95%CI:0.344-0.846),TE grade(OR=0.367,95%CI:0.116-0.618),and recovery cycle(OR=0.431,95%CI:0.218-0.682)were protective factors(P<0.05).The ROC curve indicated that the independent influencing factors had better predictive efficacy for the live birth of single blastocyst transfer assisted pregnancy;the cutoff values of patient age and AMH were 31.502 years and 5.341 ng/mL respectively(P<0.05);and the Pearson correlation analysis showed that the score of blastocyst trophoblast cells was positively correlated with the live birth rate of single blastocyst transfer assisted pregnancy.Conclusion When single blastocyst transfer is used for assisted pregnancy and leads to live birth,the AMH level,TE grade,and the proportion of the recovery cycle are relatively high.The patient's age(>31.502 years old)is an independent risk factor for single blastocyst transfer-assisted pregnancy that fails to result in live birth.However,AMH(>5.341 ng/mL),TE grade,and the recovery cycle are protective factors,and they have a good predictive effect on the live birth rate of single blastocyst transfer-assisted pregnancy,providing a theoretical reference for improving the live birth rate of single blastocyst transfer-assisted pregnancy in clinical practice.
Study on the Preparation Process of Total Flavonoids Liposomes from Suaeda salsa by Ethanol Injection Method and Its Anti-tumor ActivityAbstract:Objective To optimize the preparation process of total flavonoids liposomes from Suaeda salsa and analyze their anti-tumor activity.Methods Ethanol injection was used to prepare liposomes of total flavonoids of Suaeda salsa.Single factor experiments were conducted to investigate the factors of lipid drug ratio,membrane material ratio,injection rate,and buffer PBS temperature.The encapsulation efficiency of liposomes was used as the index,and the preparation process was optimized by Box Behnken response surface methodology,the antitumor activity of total flavonoids liposomes from Suaeda salsa was evaluated using the MTT method.Results The optimal preparation formula for the total flavonoid liposomes of Suaeda salsa was a lipid drug ratio of 9.6∶1,a membrane material ratio of 7.6∶1,an injection rate of 0.8 mL/min,and a buffer PBS temperature of 50 ℃,the encapsulation efficiency of 82.03%.In vitro release experiments showed a cumulative release rate of 66.42%within 24 hours.The total flavonoid liposomes of Suaeda salsa had inhibitory effects on the proliferation of both rectal cancer HCT116 and HT29 cells.Conclusion The liposomes of total flavonoids of Suaeda salsa prepared by ethanol injection method are reliable and convenient,and the optimized liposomes have good sustained release and anti-tumor activity.
Correlation Between Serum Apelin-13 Level and Sleep Disorder in Patients with Parkinson's DiseaseAbstract:Objective To investigate the serum levels of Apelin-13 in Parkinson's disease(PD)patients and analyze their correlation with sleep disorders.Methods A total of 95 PD patients hospitalized in the Department of Neurology of Pingdingshan Hospital of Traditional Chinese Medicine between January 2022 and January 2024 were selected.Serum Apelin-13 levels were measured,and sleep disturbances were assessed using the Parkinson's Disease Sleep Scale(PDSS).Based on Pittsburgh Sleep Quality Index(PSQI)scores,patients were divided into a sleep disorder group(n=53)and a normal sleep group(n=42).Polysomnography was used to evaluate the sleep architecture characteristics of the subjects.Serum Apelin-13 levels were compared between the two groups,and the relationship between serum Apelin-13 levels and sleep disturbances was further analyzed.The predictive value was assessed using ROC curve analysis.Results No significant differences were observed in baseline characteristics between the sleep disorder group and the normal sleep group(all P>0.05).The PDSS assessment revealed that PDSS-2(81.13%)and PDSS-8(75.47%)were the most prevalent symptoms,while PDSS-7(16.98%)and PDSS-9(15.09%)were the least frequent.Compared to the normal group,the sleep disorder group exhibited significantly reduced sleep efficiency(53.96±3.97)%,prolonged sleep latency(63.05±5.18)min,shortened REM latency(31.96±1.91)min,and increased awakenings(5.97±1.03)times(P<0.05).Serum Apelin-13 levels were significantly higher in the sleep disorder group(596.82±177.29 pg/mL)than in the normal group(P<0.05).Binary Logistic Regression identified elevated Apelin-13(OR=1.594,95%CI:1.207~1.981)as an independent risk factor for sleep disturbances in PD patients(P<O.05).ROC curve analysis demonstrated moderate predictive value for Apelin-13(AUC=0.729).Conclusion PD patients with sleep disturbances exhibit significantly elevated serum Apelin-13 levels,which serves as an independent risk factor for sleep disorders.These findings suggest Apelin-13's potential as a biomarker and highlight the therapeutic promise of developing APJ receptor-targeted drugs to improve sleep quality in PD patients.
The Relationship Between Plasma IL-6 Levels and Anticoagulant Protein Levels in Sepsis PatientsAbstract:Objective To explore the relationship between plasma IL-6 levels and anticoagulant protein levels in sepsis patients.Methods A total of 83 patients with sepsis who were treated in the Intensive Care Units(ICU)of Anyang People's Hospital from January 2024 to May 2025 were selected.According to the interleukin-6(IL-6)level(with 32.1 pg/mL as the critical value),the patients were divided into the low IL-6 level group(n=28)and the high IL-6 level group(n=55),and the differences in anticoagulant protein levels between the two groups were analyzed.Based on the death status within 30 days after the diagnosis of sepsis,the patients were divided into the death group(n=18)and the survival group(n=65).The relationship between IL-6 levels and anticoagulant protein levels was compared between the two groups.Results The procalcitonin(PCT)(79.12±29.81)ng/mL,C-reactiveprotein(CRP)(173.33±63.37)mg/L,sequential organ failure assessment(SOFA)score(9.05±3.81)points,and APACHE Ⅱ score(19.46±5.18)points in the high IL-6 level group were significantly higher than those in the low IL-6 level group(P<0.05).In this study,a total of 18 cases died,with a mortality rate of 21.60%.The survival rate of the high IL-6 level group(70.00%)was significantly lower than that of the low IL-6 level group(90.91%)(P<0.05).The levels of Protein C(PC)(37.53±13.27)%,Prtein S(PS)(69.06±13.28)%,and antithrombin Ⅲ(AT-Ⅲ)(56.15±12.19)%,in the high IL-6 level group were significantly lower than those in the low IL-6 level group.In the sepsis death group,the levels of PC(34.37±9.19)%,PS(68.38±8.47)%,and AT-Ⅲ(49.65±10.45)%,in the high IL-6 level group were significantly lower than those in the low IL-6 level group(P<0.05).The high IL-6 level group in the sepsis survival group had significantly lower levels of PC(42.35±15.18)%,PS(72.42±10.35)%,and AT-Ⅲ(60.46±9.33)%compared to the low IL-6 level group(P<0.05).Pearson correlation analysis revealed that IL-6 was significantly negatively correlated with PC(r=-0.623,P<0.001),AT-Ⅲ(r=-0.598,P<0.001),and PS(r=-0.112,P=0.025).Conclusion The elevated level of IL-6 in patients with sepsis is a factor influencing anticoagulant proteins(PC,AT-Ⅲ and PS).It can serve as a key early warning indicator for the deterioration and prognosis of sepsis-induced coagulopathy,and is beneficial for monitoring the changes in the condition of sepsis patients in clinical practice.
The Application of Nasal Humidified High-flow Pre-oxygenation During the Induction Period of General Anesthesia in PatientsAbstract:Objective To observe the effect of nasal humidification high-flow pre-oxygenation technique in the induction period of general anesthesia patients.Methods Fifty patients scheduled for general anesthesia surgeries from November 2024 to April 2025 were selected.There were 29 males and 21 females,aged 18 to 60 years old,whose ASA classification was Ⅰ or Ⅱ,Mallampatti classification was Ⅰ or Ⅱ,and BMI ranged from 18 to 28 kg/m2.They were randomly divided into two groups:Group A(mask pre-oxygenation)and Group B(nasal humidification high-flow pre-oxygenation),with 25 patients in each group.The induction methods of the anesthetic drugs were the same for both groups.Before induction,Group A received pre-oxygenation with a mask for 5 minutes,while Group B was pre-oxygenated with an oxygen flow of 50 L/min through a nasal catheter for 5 minutes after warming and humidification.The MAP,HR,and blood gas analysis(PaO2,PaCO2,FiO2/PaO2)were recorded at each time point:at the time of entering the operating room(T0),5 minutes after pre-oxygenation(T1),before tracheal intubation(T2),and after tracheal intubation(T3).The safe asphyxia time of the patients in both groups,PetCO2 immediately after tracheal intubation,whether the patients in both groups experienced a sharp drop in SpO2 requiring mask-assisted pressurized oxygenation,and the adverse reactions of the patients were recorded.Results At T3,the MAP and HR of group A were significantly higher than those at T0 and T2(P<0.05),and compared with group B,the differences were statistically significant(P<0.05);at T1,T2,and T3,the PaO2 of both groups increased(P<0.05).At T1,the PaO2 of group B was significantly higher than that of group A(P<0.05),and the increase value of PaO2(T0-1)was significantly higher than that of group A(P<0.05);at T2,the increase value of PaO2(T0-2)was significantly higher than that of group A(P<0.05).The safe asphyxia time of group B was significantly longer than that of group A(P<0.05).In group A,6 cases(24.00%)required mask-assisted pressurized oxygen supply,and there was a statistically significant difference compared with group B(P<0.05).Conclusion During the induction period of anesthesia,high-flow nasal humidification pre-oxygenation can increase PaO2,prolong the safe asphyxia time,stabilize hemodynamic indicators,and have fewer adverse reactions.It can be safely applied.
Correlation Analysis of Systemic Immune-inflammation Index with Coronary Lesions in Elderly Patients with Coronary Heart Disease Complicated with Type 2 Diabetes MellitusAbstract:Objective To investigate the correlation between systemic immune inflammation index(SII)and the severity of coronary artery lesions in patients aged≥65 years with coronary artery disease(CAD)and type 2 diabetes mellitus(T2DM).Methods A total of 122 patients admitted to the Department of Cardiovascular Medicine at the Dalian No.3 People's Hospital from January 2023 to August 2024 were enrolled.Spearman correlation analysis was used to assess the relationship between SII/100 and modified Gensini scores.Patients were divided into two groups based on modified Gensini scores:mild stenosis group(≤22 points)and moderate-to-severe stenosis group(>22 points).Clinical characteristics were compared between the two groups.Multivariate analysis was performed using logistic regression,and the model was evaluated using ROC curves and calibration curves.Additionally,RCS analysis was employed to further examine the linear relationship between SII/100 and coronary lesion severity.Results The SII/100,total cholesterol,low-density lipoprotein,alanine aminotransferase(ALT),aspartate aminotransferase(AST),and fibrinogen levels in the moderate-severe lesion group were significantly higher than those in the mild lesion group(P<0.005),while the serum cystatin C level was lower than that in the mild lesion group(P<0.005).Multivariate logistic regression analysis showed that SII/100(OR=1.835,95%CI:1.285-2.620,P<0.001)and serum cystatin C(OR=0.155,95%CI:0.026-0.935,P=0.042)were independent factors influencing coronary lesion severity.The ROC curve of the multivariate logistic regression model showed an AUC of 0.793(95%CI:0.735-0.906),and the calibration curve demonstrated a good fit.SII/100 was significantly positively correlated with modified Gensini scores(r=0.360,P<0.05).After adjusting for confounding factors such as age,BMI,CAD history,smoking,alcohol consumption,hypertension,LDL,ALT,AST,fibrinogen,and serum cystatin,SII/100 remained independently and positively associated with coronary lesion severity(OR:1.96,95%CI:1.300-2.970,P<0.001),with a linear relationship and with an inflection point at 5.11.Conclusion SII was closely associated with the severity of coronary artery lesions in elderly patients(≥65 years)with CAD and T2DM.Early monitoring of SII levels in this population may provide valuable insights for the prevention and management of coronary artery disease.
The Influence of Serum CTRP15,BACH1 and TRAIL Levels on Cardiac Function and Prognosis in Patients with Acute Myocardial InfarctionAbstract:Objective To explore the evaluation value of serum levels of C1q tumor necrosis factor-related protein 15(CTRP15),BTB-CNC homolog 1(BACH1),and tumor necrosis factor-related apoptosis-inducing ligand(TRAIL)for the cardiac function and prognosis of patients with acute myocardial infarction(AMI).Methods A total of 151 AMI patients admitted to the Sixth Ward of the Department of Cardiovascular Medicine of the Second Affiliated Hospital of Zhengzhou University from September 2022 to June 2024 were selected as the AMI group.They were divided into the good prognosis group and the poor prognosis group based on the prognosis.Another 100 healthy individuals undergoing physical examinations were selected as the control group.Color Doppler ultrasound was used to detect the cardiac function of the subjects.Enzyme-linked immunosorbnent assay(ELISA)method was used to detect the levels of serum CTRP15,BACH1,and TRAIL.The data and serum CTRP15,BACH1,and TRAIL levels of the AMI group and the control group were compared.The cardiac function parameters of the AMI group and the control group were compared.The serum CTRP15,BACH1,and TRAIL levels of patients with different cardiac function grades were compared.Pearson correlation analysis was used to analyze the correlation between serum CTRP15,BACH1,and TRAIL levels and cardiac function parameters.The incidence of poor prognosis in AMI patients was recorded.The data and serum CTRP15,BACH1,and TRAIL levels of patients with different prognoses were compared.The influencing factors of poor prognosis in AMI patients were analyzed using multivariate Logistic regression.The predictive value of serum CTRP15,BACH1,and TRAIL levels for poor prognosis in AMI patients was analyzed using receiver operating characteristic(ROC)curve analysis.Results The serum BACH1 level of patients in the AMI group was higher than that of the control group,while the serum levels of CTRP15 and TRAIL were lower than those of the control group(P<0.05).The LVEF level of patients in the AMI group was lower than that of the control group,while left ventricular end diastolic dimension(LVDD)and left ventricular posterior wall thickness(LVPWT)levels were higher than those of the control group(P<0.05).The higher the cardiac function grade of AMI patients,the higher the serum BACH1 level,and the lower the serum CTRP15 and TRAIL levels(P<0.05).The levels of serum CTRP15,BACH1 and TRAIL are closely related to the cardiac function parameters of AMI patients(P<0.05).The incidence of poor prognosis in AMI patients is 31.79%.The serum BACH1 level in the poor prognosis group is higher than that in the good prognosis group,while the serum CTRP15 and TRAIL levels are lower than those in the good prognosis group(P<0.05).The levels of serum CTRP15,BACH1 and TRAIL are influencing factors for poor prognosis in AMI patients(P<0.05).The joint of serum CTRP15,BACH1,and TRAIL levels had a higher value in predicting poor prognosis in AMI patients than individual predictions(ZCTRP15-joint prediction,ZBACH1-joint prediction,ZTRAIL-joint prediction=2.671,3.202,2.246;PCTRP15-joint prediction,PBACH1-joint prediction,PTRAIL-joint prediction=0.008,0.001,0.025).Conclusion Serum BACH1 is elevated in AMI patients,while serum CTRP15 and TRAIL are reduced.The levels of the three factors are closely related to the cardiac function and prognosis of AMI patients,and the joint of the three factors has a high value in predicting poor prognosis of AMI patients.
The Role of Vedolizumab in Combination Therapy for Treating Ulcerative Colitis with Non-responseAbstract:Objective To identify the precise molecular targets for vedolizumab(VDZ)treatment of ulcerative colitis(UC)that lead to non-response through clinical multivariate analysis and machine learning,and to explore combined medication strategies.Methods A retrospective study was conducted to analyze the risk factors related to the clinical response of patients with moderate to severe ulcerative colitis(UC)who received VDZ treatment,and a Logistic regression analysis was constructed.At the same time,based on the comprehensive dataset of gene expression,differential gene expression analysis,weighted gene co-expression network analysis,protein interaction network,and least absolute shrinkage and selection operator regression analysis were used to screen the key genes for non-response.Combined with immune infiltration analysis and the Enrichr website,potential combined therapeutic drugs were explored.Results Clinical multivariate analysis revealed that a Mayo endoscopy score of≥2 points was an independent risk factor for clinical response(OR=5.835,95%CI:1.052-32.361,P=0.044).Bioinformatics analysis and machine learning identified interferon regulatory factor 4(IRF4)as the core hub gene driving the failure to respond.Functional enrichment analysis indicated that it was mainly enriched in key pathways such as the differentiation of type 17 helper T cells(Th17)and the interleukin 17(IL-17)signaling pathway.Immune infiltration analysis showed that M1 macrophages and neutrophils were significantly enriched in patients with failure to respond,suggesting the activation of alternative inflammatory pathways.Based on these mechanisms,JAK inhibitors and anti-interleukin 23(IL-23)antibodies were selected as potential combined treatment strategies to reverse the failure to respond in VDZ.Conclusion A Mayo endoscopy score of≥2 points is an independent risk factor for the clinical response in patients undergoing VDZ treatment.For UC patients with severe endoscopic inflammation,the failure to respond to VDZ may be related to the activation of the alternative inflammatory pathway mediated by the IRF4 gene.The combined use of JAK inhibitors or anti-IL-23 antibodies may be an effective strategy to overcome this challenge.
