Optimization of Ultrasonic-assisted Enzymatic Extraction Process for Alkaloids from Metaplexis japonic and Its Anti-tumor Activity Analysis

Abstract:Objective To optimize the extraction process of alkaloids from the Metaplexis japonic and study its anti-tumor activity.Methods Using the Metaplexis japonic as the raw material,ultrasound-assisted enzymatic hydrolysis was used to extract alkaloids from Metaplexis japonic.The extraction rate of alkaloids from Metaplexis japonic was used as the indicator,and single factor experiments and response surface methodology were used to design optimization experiments to explore the optimal extraction process of alkaloids from Metaplexis japonic.Results The optimal process for extracting alkaloids from Metaplexis japonic was achieved with an enzyme concentration of 530 U/mL,enzymatic hydrolysis time of 63 min,enzymatic hydrolysis temperature of 51 ℃,and ultrasound power of 140 W.At this time,the extraction rate of alkaloids from Metaplexis japonic was 10.40%.The cell viability of MCF-7 decreases with the increase of the concentration and duration of action of alkaloids from Metaplexis japonic.Conclusion The above results indicate that ultrasound-assisted enzymatic hydrolysis can improve the extraction rate of alkaloids from Metaplexis japonic.The optimized extraction process conditions of this method are feasible and reliable.
