Determination of Reduced Glutathione and Glutamic Acid in Brain Tissue by HPLC with Pre-column Derivatization Using O-PhthaldialdehydeAbstract:Objective To establish a high-performance liquid chromatography(HPLC)method for the detection of glutathione(GSH)and glutamate levels in brain tissue.Methods C18 column was used with a mobile phase of methanol-acetonitrile-water and 16 mM phosphate buffer(pH=8.2)under isothermal elution at 38 ℃.O-phthalaldehyde was used as a pre-column derivation agent,with a flow rate of 1 mL/min.The excitation wavelength was set to 338 nm and the emission wavelength to 425 nm to simultaneously detect GSH and glutamate levels.Results GSH and glutamate were baseline-separated,showing good linearity within the range of 0.25 to 4.00 μg/mL,with correlation coefficients of 0.9994 and 0.9996,respectively.The recovery rates were 99.71%and 99.27%.Conclusion The method is sensitive and reliable.The levels of GSH and glutamate in rat brain tissue were measured at(65.05±10.15)and(105.91±39.38)μg/g wet weight,respectively.
The Expression Relationship of HIF-1α,MMP-2,and MMP-14 in Bladder Urothelial CarcinomaAbstract:Objective To investigate the expression levels and clinical significance of hypoxia inducible factor-1α(HIF-1α),matrix metalloproteinase-2(MMP-2),and matrix metalloproteinase-14(MMP-14)in bladder and urinary tract epithelial cancer.Methods 57 cases of bladder urothelial carcinoma tissues(observation group)and 15 cases of non tumor mucosal tissues(control group)were selected.Protein expression was detected by immunohistochemistry(IHC),and semi quantitative scoring(staining intensity x positive area ratio)was used to analyze expression differences.The tissue morphology was evaluated by HE staining.Spearman correlation analysis was used to explore the relationship between HIF-1 α,MMP-2,and MMP-14.Results The semi quantitative scores of HIF-1 α,MMP-2,and MMP-14 in the observation group were significantly higher than those in the control group(all P<0.05),and were positively correlated with tumor grading,vascular invasion,maximum diameter,and proliferation index(P<0.05).Spearman analysis showed a positive correlation between HIF-1 α and MMP-2,MMP-14.HE staining showed significant heterogeneity in cancer tissue,and immunohistochemical images confirmed high protein expression.Conclusion The high expression of HIF-1 α,MMP-2,and MMP-14 may promote the progression of bladder urothelial carcinoma by regulating extracellular matrix degradation and tumor microenvironment,and these three factors can serve as potential biomarkers and therapeutic targets.
Efficacy of Capsular Tension Rings in Treating Acute Angle-closure Glaucoma Secondary to Abnormal Lens Suspensory LigamentsAbstract:Objective To analyze clinical efficacy of applying a capsular tension ring in the treatment of acute angle-closure glaucoma secondary to abnormalities of the lens suspensory ligament.Methods A prospective study was conducted at Yantai Affiliated Hospital of Binzhou Medical University from December 2023 to July 2024 involving 40 patients with acute angle-closure glaucoma secondary to abnormalities of the lens suspensory ligament.The patients were randomly divided into two groups,with 20 cases in each group.Group A underwent capsular tension ring implantation,while Group B did not.Both groups underwent phacoemulsification cataract extraction combined with intraocular lens implantation and anterior chamber angle microscopy-assisted angle separation.The intraocular pressure,best corrected visual acuity,the tilt and eccentricity of the intraocular lens,and the openness of the anterior chamber angle at 1 week,1 month,and 6 months after surgery in the two groups were compared.Results The intraocular pressure of both groups decreased after the operation,and the difference was statistically significant compared with that before the operation(P=0.001).There was no statistically significant difference between the two groups in the comparison(P>0.05);the visual acuity of both groups improved after the operation,and the difference was statistically significant compared with that before the operation(P<0.05),and the difference between the two groups in the comparison was also statistically significant(P<0.05);the eccentricity and tilt degree of the intraocular lenses in both groups were statistically significantly different(P<0.001,P<0.001);there was no statistically significant difference in the comparison of the trabecular iris angle(TIA250)between the two groups(P>0.05).Conclusion The combined use of phacoemulsification and aspiration for cataract removal along with anterior chamber angle separation in the treatment of acute angle-closure glaucoma secondary to abnormalities of the lens suspensory ligament can effectively control intraocular pressure,improve vision,and in combination with the implantation of a capsular tension ring,it can reduce the postoperative eccentricity and tilt of the intraocular lens,ensuring the effective position of the postoperative intraocular lens.
Exploration of the Effect of Combining Orthodontics and Oral Restoration in the Treatment of Congenital Missing TeethAbstract:Objective Exploring the clinical efficacy of combining orthodontics and oral restoration in the treatment of congenital missing teeth.Methods 120 cases of congenital tooth loss who visited our hospital from February 2021 to February 2023 were selected and randomly divided into two groups using computer random sampling method,with 60 cases in the observation group and 60 cases in the control group.The control group received routine oral restoration treatment,while the observation group received a combination of orthodontic and oral restoration treatment.The oral comfort,aesthetic satisfaction,bite force,chewing function,anxiety situation,treatment effect,adverse reactions,language function,and orthodontic comfort were compared between the two groups.Results The satisfaction scores of oral restoration comfort and aesthetics in the observation group before and after treatment were significantly higher than the control group(P<0.05);observation group showed significantly better oral comfort,bite force,and chewing function recovery compared to the control group,with a higher total effective rate(P<0.05);the anxiety level of the observation group was significantly better than that of the control group(P<0.05);after treatment,the speech function scores of the observation group and the control group were higher than before treatment,and the observation group was higher than the control group(P<0.05);the comfort level of orthodontic treatment correction comfort of the observation group was significantly better than that of the control group(P<0.05).Conclusion Timely orthodontic and oral restoration combined treatment for congenital edentulism can effectively improve the clinical oral restoration effect,reduce adverse reaction after restoration,improve the patient's aesthetic degree and comfort,improve the patient's bite force,chewing function and language function recovery effect,and reduce the impact of oral diseases on the patient's quality of life.
Treatment of Ankle Fractures Combined with Distal Tibiofibular Syndesmosis Instability Using Button PlateAbstract:Objective To investigate the clinical effectiveness of loop titanium plate and distal tibiofibular screw fixation in the treatment of ankle fractures combined with distal tibiofibular syndesmosis instability.Methods 70 patients with ankle fractures combined with distal tibiofibular syndesmosis instability treated at our hospital from July 2022 to July 2024 were selected as the study subjects.They were randomly divided into a control group(n=35)and an observation group(n=35).The control group was treated with distal tibiofibular screw fixation,while the observation group was treated with loop titanium plate fixation.Surgical indicators,joint function scores,and complication rates were observed and compared between the two groups.Results The surgical indicators and joint function in the observation group were superior to those in the control group(P<0.05).The complication rate in the observation group was 5.71%,significantly lower than that in the control group(20.00%)(P<0.05).Conclusion Loop titanium plate fixation is more effective than distal tibiofibular screw fixation.It helps improve surgical outcomes,prevent the continuous deterioration of joint function,and reduce the likelihood of complications.
The Effect of Phacoemulsification Combined with Goniosynechialysis on Angle-closure Glaucoma Complicated with CataractAbstract:Objective To explore and analyze the impact of phacoemulsification combined with primary posterior chamber intraocular lens implantation and goniosynechialysis on the treatment of angle-closure glaucoma combined with cataract.Methods Patients diagnosed with primary angle-closure glaucoma and cataract at the Department of Ophthalmology,Gao'an People's Hospital,Jiangxi Province,from January 2021 to December 2023 were selected as the study subjects.The research subjects were randomly grouped through computer programming.In the end,92 patients were enrolled.The observation group received phacoemulsification and aspiration of cataract combined with one-stage implantation of posterior chamber intraocular lens and angle separation surgery(n=46),while the control group received phacoemulsification and aspiration of cataract combined with one-stage implantation of posterior chamber intraocular lens and trabeculectomy(n=46).Statistical analysis was conducted to determine whether there were differences in indicators such as best corrected visual acuity(BCVA),intraocular pressure,angle opening status,and central anterior chamber depth one month after the operation.Results One month after the operation,both groups were re-examined:The best corrected visual acuity(BCVA)of the observation group was significantly higher than that of the control group(P<0.01).The intraocular pressure of both groups was significantly lower after the operation than before,but there was no statistically significant difference in the degree of pressure reduction between the two groups(P>0.05).The central anterior chamber depth of the observation group was significantly increased compared with that of the control group after the operation(P<0.05),and the angle opening of the observation group was significantly wider than that of the control group after the operation(P<0.05).The incidence of postoperative complications in the observation group was lower than that in the control group(P<0.05).Conclusion For patients with primary angle-closure glaucoma combined with cataract,treatment with phacoemulsification combined with primary intraocular lens implantation and goniosynechialysis yields clinical outcomes that align more closely with expectations.This approach significantly reduces the probability of complications during treatment,markedly lowers postoperative intraocular pressure,significantly deepens the central anterior chamber,further opens the angle,and improves BCVA.The clinical efficacy is definite,with high safety,making it worthy of further clinical promotion and application.
Alisma Orientalis Extract and Its Compound Prescription Reduce Body Weight and Body Fat in Obese RatsAbstract:Objective To investigate the lipid-lowering effects of alisma orientale extract(AE)and its compound preparation(CP),which contains konjac flour,L-carnitine,and tea polyphenols,on obese rats fed with high-fat diet.Methods 80 rats were randomly divided into four groups:the blank group(fed with normal diet),the model group(fed with high-fat diet),the AE group,and the CP group(20 rats in each group).The intervention was carried out by intragastric administration for 4 weeks until 16 weeks.Body weight and food intake were monitored,and serum lipid metabolism indicators such as total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C),as well as liver function indicators such as alanine aminotransferase(ALT),aspartate aminotransferase(AST),were detected.To determine the oxidative stress level in liver tissue,the contents of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),and malondialdehyde(MDA)were measured.At the same time,to evaluate the inflammatory response,the levels of tumor necrosis factor-α(TNF-α),interleukin-1 β(IL-1β),and interleukin-6(IL-6)were measured.Results After the intervention,the AE group and the CP group could reduce the weight increase,liver weight and liver index compared with the model group(P<0.05),among which the CP group had a better effect.The serum levels of TG,TC,ALT,AST,and LDL-C in both intervention groups decreased by 17.30%to 28.10%,while HDL-C increased by 21.50%to 24.80%.The activities of SOD and GSH-Px in liver tissue increased by 34.70%to 41.20%,and the MDA content decreased by 39.40%to 45.10%.The levels of inflammatory factors TNF-α,IL-1β,and IL-6 decreased by 32.80%to 50.30%.Conclusion The extract of Alisma and its compound preparations can effectively inhibit lipid deposition in obese rats.The compound formulation has a more significant advantage in improving lipid metabolism disorders,reducing oxidative stress and inflammatory responses through a synergistic enhancement mechanism.
Curcumin Alleviates SCI Glial Scars by Regulating Mitochondrial Function and Promotes the Recovery of Motor Function in MiceAbstract:Objective This study aims to investigate whether curcumin attenuates the excessive activation of astrocytes after spinal cord injury(SCI)by regulating mitochondrial function,thereby inhibiting glial scar formation and promoting the recovery of motor function in mice.Methods A mouse model of spinal cord injury was established,and the animals were randomly divided into sham-operated group,spinal cord injury group,and curcumin-treated groups.Western Blot analysis was performed to detect the expression level of glial fibrillary acidic protein(GFAP).In vitro experiments on astrocytes were conducted using mitochondrial membrane potential probes to assess changes in membrane potential,and ATP content was measured to evaluate cellular energy metabolism.Behavioral analysis was carried out using the Basso Mouse Scale(BMS)scoring system to assess the recovery of motor function.Results Curcumin treatment significantly enhanced mitochondrial membrane potential and ATP synthesis levels in cells within the injured area and markedly suppressed the expression of the astrocyte marker GFAP.Furthermore,mice in the curcumin-treated group showed significant improvement in motor function recovery as indicated by BMS scores.Conclusion Curcumin promotes neurological repair and functional recovery after spinal cord injury by improving mitochondrial function and suppressing astrocyte activation and glial scar formation,suggesting a potential drug intervention strategy for the clinical treatment of SCI.
The Effect of Modified Endoscopic Mucosal Resection on Surgical Stress Response and Gastrointestinal Function in Elderly Patients with Gastrointestinal PolypsAbstract:Objective To explore the effect of modified endoscopic mucosal resection on surgical stress response and gastrointestinal function in elderly patients with gastrointestinal polyps.Methods A total of 120 elderly patients with gastrointestinal polyps admitted to our hospital from January 2022 to December 2023 were randomly selected for this study.The patients were divided into two groups using the random number table method.Among them,60 patients in the control group received conventional treatment,while 60 patients in the observation group underwent modified endoscopic submucosal dissection.The cortisol(Cor),norepinephrine(NE),creatine kinase(CK)levels,the one-time resection rate of polyps,and the amplitude of the ascending colon and transverse colon were compared between the two groups of patients regarding their surgical stress response indicators.Results The one-time resection rate of polyps in the observation group was 98.33%(59/60),significantly higher than that in the control group which was 85.00%(51/60).The difference was statistically significant(x2=5.125,P=0.014).After the operation,the amplitude of the ascending colon(123.15±13.46)μV and the amplitude of the transverse colon(6.14±0.75)per minute in the observation group were significantly lower than those in the control group[(137.24±14.57)μV,(7.28±0.83)times/minute],and the difference was statistically significant(t=5.237/6.392,P<0.001).The postoperative Cor(175.03±12.34)ng/mL,NE(44.19±5.48)ng/mL,and CK(179.58±21.34)U/L expression levels in the observation group were lower than those in the control group(213.51±12.41)mg/mL,(52.32±5.53)ng/mL,and(223.23±21.13)U/L,and the differences were statistically significant(t=9.648,5.427,8.075,P<0.001).Conclusion Modified endoscopic mucosal resection can effectively reduce surgical stress response,and improve postoperative gastrointestinal function in patients with gastrointestinal polyps.
Expression Characteristics of PDZRN3 in Endometrial Cancer and Its Bioinformatics EvaluationAbstract:Objective To evaluate the expression characteristics of E3 ubiquitin ligase(PDZ Domain Containing Ring Finger 3,PDZRN3)in endometrial cancer using bioinformatics.Methods The expression differences of PDZRN3 in endometrial cancer tissues and normal endometrial tissues were analyzed using TIMER2 and Xiantao Academic( on the clinical information of endometrial cancer patients from the TCGA database,baseline data tables were created using the stats package in R software.The clinical stages and pathological grades were analyzed using the stats,car,and ggplot2 packages.Kaplan-Meier survival curves were plotted using the survival and survminer packages.The COX proportional hazards model was analyzed using the survivalR package,and COX regression analysis tables were created.Diagnostic ROC curves and nomogram models were generated using the pROC and survival packages.In vitro,endometrial cancer cell lines AN3CA and Ishikawa were cultured,and the protein expression of PDZRN3 in these two cell lines was detected by Western Blot.Results TIMER2 database and Xiantao academic analysis showed that PDZRN3 was lowly expressed in endometrial cancer tissues(P<0.05).TCGA database indicated that the expression of PDZRN3 was significantly correlated with clinical stage,histological grade and histological type(P<0.05).Endometrial cancer patients with higher PDZRN3 expression had higher overall survival(OS),disease-specific survival(DSS)and progression-free interval(PFI),while those with lower PDZRN3 expression had lower OS,DSS and PFI.Both univariate and multivariate COX regression analyses demonstrated that the expression of PDZRN3,age,clinical stage,and pathological grade in endometrial cancer patients were risk factors affecting prognosis(P<0.05).The ROC curve results indicated that the expression of PDZRN3 could help distinguish tumor tissues from non-tumor tissues(AUC:0.865),and the nomogram model could be used to predict the 1-year,3-year,and 5-year survival probabilities of clinical patients.Western Blot results showed that PDZRN3 was highly expressed in the endometrial cancer cell line AN3CA(P<0.01),and was lowly expressed in the endometrial cancer cell line Ishikawa(P<0.01).Conclusion PDZRN3,as a highly promising biomarker,is expected to play a significant role in the stratification,prognosis assessment,and treatment intervention of patients with endometrial cancer.
The Expression and Significance of CEACAM6,VEGF,ANG2,and miR-146a-5p in Prostate AdenocarcinomaAbstract:Objective To investigate the expression levels of carcinoembryonic antigen associated cell adhesion molecular-6(CEACAM6),vascular endothelial growth factor(VEGF),angiopoietin 2(ANG2),and microRNA-146a-5p(miR-146a-5p)in prostate adenocarcinoma,and analyze their relationship with clinical pathological characteristics.Methods 77 patients diagnosed with prostate adenocarcinoma through pathology were selected as the research group,and their postoperative wax block tissues were collected;normal mucosal tissues at the edge of the tumor were retained as the control group.The immunohistochemical two-step method was used to detect CEACAM6,VEGF,and ANG2 in the tissues of both groups.RT-qPCR was applied to detect the expression level of miR-146a-5p in prostate cancer tissues,and the correlations between the related indicators and the clinical pathological characteristics of the patients(Gleason score,TNM stage,tumor grade,etc.)were analyzed.Results The positive expression rates of CEACAM6,VEGF and ANG2 proteins in the research group were significantly higher than those in the control group.The expressions of CEACAM6,VEGF and ANG2 were closely related to clinical pathological features such as Gleason score,TNM stage,tumor grade and intratumoral vascular tumor thrombus.The expression level of miR-146a-5p in the research group was significantly lower than that in the control group,and it was negatively correlated with the expressions of CEACAM6 and VEGF.Conclusion CEACAM6,VEGF and ANG2 are highly expressed in prostate adenocarcinoma,and are associated with the invasiveness,metastasis risk and poor prognosis of the tumor.miR-146a-5p may act as a tumor suppressor,influencing the progression of the tumor by regulating the expression of CEACAM6 and VEGF.
Analysis of the Influencing Factors of Anxiety in Patients with Parkinson's DiseaseAbstract:Objective To analyze the clinical characteristics and influencing factors of anxiety in patients with Parkinson's disease(PD)and provide evidence for improving their quality of life.Methods A total of 115 PD patients were recruited from the Parkinson's Disease Specialty Clinic of the First Affiliated Hospital of Jinzhou Medical University between September 2023 and September 2024.Demographic data(e.g.,sex,age,disease duration),motor/non-motor symptom assessments(via standardized scales),and inflammatory markers were collected.Patients were divided into anxiety and non-anxiety groups based on Hamilton Anxiety Rating Scale(HAMA)scores,and the anxiety group was further stratified into three severity levels:possible anxiety,probable anxiety,and marked anxiety.Univariate and multivariate Logistic regression analyses were performed to identify risk factors,and the predictive value was evaluated using receiver operating characteristic(ROC)curve analysis.Results PD-A patients exhibited higher scores on the MDS-UPDRS-Ⅲ,HAMD,PSQI,SCOPA-AUT,CCS,and FS-14 scales,as well as elevated levels of CRP and IBI,but lower levels of IL-6.MDS-UPDRS-Ⅲ score(OR=1.176,P=0.006),HAMD score(OR=1.181,P=0.015),IL-6 level(OR=0.904,P=0.014),and IBI level(OR=1.017,P=0.044)were identified as independent predictors of PD-A.The combined predictive model demonstrated good performance,with an AUC of 0.833.Additionally,MDS-UPDRS-Ⅲscore(OR=1.439,P<0.001)was an independent risk factor for the severity of PD-A.Conclusion PD-A patients experience more severe motor and non-motor symptoms,along with higher levels of systemic inflammation.Motor dysfunction,depressive symptoms,IL-6,and IBI levels are independent predictors of PD-A,and the combined predictive model shows high clinical value.Motor dysfunction is a key factor influencing the severity of PD-A.
The Effects of a Single Bout of Acute Aerobic Exercise on Executive Inhibition and Prefrontal Cortex Mechanisms in Individuals with Different Types of PainAbstract:Objective To explore the effect of a single acute aerobic exercise on the inhibitory function of different types of pain and the underlying neural mechanism,in order to provide references for promoting the management and intervention methods of different types of pain.Methods A total of 29 subjects,including those with chronic pain and healthy individuals,were selected.Acute pain was induced in the healthy subjects.Behavioral and functional near-infrared spectroscopy(fNIRS)techniques were used to investigate the differences in the performance of executive inhibition among different types of pain sufferers before and after the exercise intervention.Results The behavioral results showed that the reaction time of the chronic pain group in completing the executive inhibition task was significantly higher than that of the acute pain group.After the exercise intervention,the reaction time of both the chronic pain group and the acute pain group in completing the executive inhibition task significantly shortened,and the accuracy rate significantly improved.The fNIRS results indicated that after the exercise intervention,the oxygenated hemoglobin content in the right dorsolateral prefrontal cortex(DLPFC)and the right prefrontal eye field(FEF)of the acute pain group was significantly higher than that before the exercise,and the oxygenated hemoglobin content in the left DLPFC of the acute pain group was significantly higher than that of the chronic pain group.Conclusion A single acute aerobic exercise has a dual effect of behavioral and neural improvement on the inhibitory function in the acute pain group,while it only shows compensatory behavioral improvement in the chronic pain group.This result provides a neurobehavioral basis for clinical pain management,that is,during the acute pain period,acute aerobic exercise intervention can effectively improve the inhibitory function and alleviate the damage to the PFC neural activity,thereby delaying the chronic progression of pain.
The Effect of Percutaneous Acupoint Electrical Stimulation Therapy Combined with PCIA on Gastrointestinal Function Recovery and Pain After Radical Gastrectomy for Gastric CancerAbstract:Objective To explore the effect of transcutaneous electrical acupoint stimulation therapy combined with patient controlled intravenous analgesia(PCIA)on the recovery of gastrointestinal function and pain after radical gastrectomy for gastric cancer.Methods A total of 80 patients who underwent gastric cancer surgery and were admitted to the Anesthesiology Department of Anyang People's Hospital from March 2022 to February 2025 were included in the study.They were randomly and equally divided into the control group and the study group(40 cases in each group).Both groups received standardized perioperative management,with the control group receiving PCIA alone while the study group combined PCIA with transcutaneous electrical acupoint stimulation.Evaluations included analgesic and sedative effects,pain mediators,inflammatory markers,lymphocyte subsets,time to first flatus,defecation,bowel sound recovery,hospitalizationduration,gastrointestinal motility,and adverse reactions.Results The assessment of pain intensity at 4,24 and 48 hours after the operation showed that the visual analogue scale(VAS)of the study group was significantly lower than that of the control group,while the Ramsay score was significantly higher than that of the control group(P<0.05);four hours after the operation,the levels of 5-hydroxytryptamine(5-HT)in the study group(1.36±0.31)μmol/L and the level of prostaglandin E2(PGE2)(36.19±3.27)ng/mL were significantly lower than those in the control group(P<0.05);four hours after the operation,the levels of interleukin(IL)-6(156.85±6.18)pg/mL and IL-1 β(122.82±11.09)pg/mL in the study group were significantly lower than those in the control group(P<0.05);four hours after the operation,the percentages of CD3 positive(75.19±13.11)%,CD4 positive(52.61±8.43)%,and CD4 positive/CD8 positive(3.55±0.86)in the study group were significantly higher than those in the control group,while the percentage of CD8 positive(20.61±4.18)was significantly lower than that in the control group(P<0.05);the recovery time of bowel sounds(15.23±2.42)hours,the time of anal exhaust(25.48±2.61)hours,the defecation time(42.28±9.43)hours,and the hospital stay(7.53±0.23)days in the study group were significantly shorter than those in the control group(P<0.05);four hours after the operation,the levels of motilin(MOT),gastrin(GAS),substance P(SP),and cholecystokinin(CCK)in the study group were higher than those in the control group,while the levels of vasoactive intestinal peptide(VIP),somatostatin(SS),and neurotensin(NT)were lower than those in the control group(P<0.05);in the assessment of adverse reactions,the incidence of postoperative nausea and vomiting in the study group(12.50%)was significantly lower than that in the control group(52.50%)(P<0.05).Conclusion The combination of transcutaneous electrical acupoint stimulation with PCIA significantly enhances postoperative analgesia and sedation in radical gastrectomy patients,while effectively reducing pain mediators and inflammatory factors.This integrated approach demonstrates immunomodulatory benefits,accelerates gastrointestinal motility recovery,shortens postoperative rehabilitation time,and decreases the incidence of adverse reactions such as nausea and vomiting.The combined intervention shows substantial clinical value in promoting enhanced recovery after gastric cancer surgery.
Application of Metagenomic Next-generation Sequencing in the Diagnosis of Pulmonary InfectionsAbstract:Objective To investigate the value of metagenomic next-generation sequencing(mNGS)technology in the diagnosis of pulmonary infectious diseases.Methods A retrospective collection of 76 samples from patients with suspected pulmonary infections in the south of Anhui Province between November 2023 and February 2024 was conducted.Samples were tested using both mNGS technology and traditional methods.Clinical data were collected,and the results were analyzed.Results Among the 76 patients with suspected pulmonary infections,69 cases were confirmed pulmonary infections.The sensitivity,specificity,and positive rate of mNGS technology were 81.16%,42.86%,and 78.95%respectively,while those of the conventional method were 43.48%,71.42%,and 42.11%respectively.A comparison between the two methods revealed statistically significant differences in detection sensitivity,specificity,and positive rate(x2=30.645,15.993,28.643,P<0.05).Among the 76 patient samples,mNGS detected a total of 42 pathogens,while traditional methods detected 18 pathogens.The detection rates of bacteria,fungi,and viruses were higher with mNGS than with other methods(x2 values were 12.737,7.611,and 7.134 respectively,P<0.05).Although the detection rates of Mycobacterium,Mycoplasma,and Chlamydia by mNGS were higher than those by traditional methods,the differences were not statistically significant(P>0.05).Conclusion mNGS technology has higher sensitivity and detection rates compared to traditional methods,but its specificity is lower.It can complement traditional pathogen detection methods by identifying pathogens that may be missed.
Application of Motivational Psychological Intervention Based on Behavior Change Theory in Patients with Cerebral Small Vessel DiseaseAbstract:Objective To evaluate the efficacy of a motivational psychological intervention based on behavior change theory in patients with cerebral small vessel disease(CSVD).Methods 80 CSVD inpatients meeting inclusion/exclusion criteria were enrolled from the Zhengzhou People's Hospital(October 2022-October 2024)and randomly allocated via lottery to either a control group(n=40,routine care)or an intervention group(n=40,routine care plus behavior change theory-based intervention).Outcomes included step width,gait speed,stride length,compliance,Short Physical Performance Battery(SPPB)scores,Tinetti scores,cognitive function,Self-Rating Anxiety Scale(SAS)scores,and Self-Rating Depression Scale(SDS)scores.Results After the intervention,the observation group showed significantly better improvements in step width,walking speed,and stride length compared to the control group(P<0.05).Additionally,the observation group had significantly higher scores in physical activity,exercise monitoring,and proactive advice-seeking(P<0.05).The observation group also demonstrated significantly higher scores in standing balance,3-meter walk,and five-time sit-to-stand tests,as well as overall scores(P<0.05).According to the Tinetti assessment,the observation group had significantly higher scores in gait,balance,and total scores after the intervention(P<0.05).Moreover,the observation group exhibited significantly improved cognitive function(P<0.05).In terms of emotional status,the observation group showed significantly lower SAS(anxiety)and SDS(depression)scores compared to the control group after the intervention(P<0.05).Conclusion The behavior change theory-based intervention significantly improves gait,balance,compliance,cognitive function,and emotional status in CSVD patients,offering an effective rehabilitation strategy.
The Investigation on the Effect of β-caryophyllene on Ferroptosis and Neuroprotection Mechanisms in Rats with Vascular DementiaAbstract:Objective To verify the effect of β-stilbene on ferroptosis in brain tissues of rats with vascular dementia(VaD)and explore its neuroprotective mechanism.Methods During the experimental operation,the muscles,fascia and other tissues of the rats were carefully dissected to fully expose the bilateral common carotid arteries.Subsequently,only suturing was performed on them,which was used as the sham operation group.In another group of rats,the bilateral common carotid arteries were also exposed,but ligation was used,and thus the VaD model of SD rats was successfully established.After the model preparation was completed,the rats with successfully prepared models were selected and randomly divided into the model group and the β-caryophyllene administration group.Seven days after the successful establishment of the model,rats in the β-caryophyllene administration group were injected withβ-caryophyllene(408 mg/kg)through the tail vein.After continuous administration for two weeks,rats in each group were taken for the Morris water maze test.The next day,some rats were randomly selected from each group,and their brain tissues were taken for hematoxylin-eosin(HE)staining.Strictly follow the operation steps in the kit manual to complete the determination of Fe2+,malondialdehyde(MDA),glutataione(GSH),and superoxide dismutase(SOD)contents in brain tissue.Using the Western Blot method,the protein contents of glutathioneperoxidase 4(GPX4)and acyl-coA synthetase long-chain family member 4(ACSIA)in each group were detected.Results The results of the Morris water maze experiment indicated that during the period from the 2nd to the 5th day after β-caryophyllene administration treatment,the escape latency of rats was shortened,the stay time in the target area was prolonged,and the number of times they crossed the platform increased.Compared with the sham operation group,the levels of Fe2+and MDA in the brain tissue of rats in the model group were significantly increased,the content of GSH decreased,the activity of SOD decreased,the number of hippocampal cells decreased and nuclear contraction occurred.Compared with the model group,the levels of Fe2+and MDA in the brain tissue of rats in the β-caryophyllene administration group decreased,the content of GSH increased,the activity of SOD enhanced,the number of hippocampal cells increased,and the tissue morphology tended to be normal.The Western Blot results showed that the content of GPX4 protein in the hippocampal tissue of the model group was significantly lower than that of the sham operation group,while the content of ACSL4 protein was higher.Compared with the model group,the expression of GPX4 was upregulated and the expression of ACSL4 was downregulated in the hippocampal tissue of the β-caryophyllene group.Conclusion β-caryophyllene may protect cerebral neurons by inhibiting ferroptosis of cerebral neurons in VaD rats,thereby improving learning and spatial memory abilities of VaD rats.
Expression of Tubulin and mRNA in Non-small Cell Lung CancerAbstract:Objective The expression levels of microtubule-associated protein 2(MAP2)and β-tubulin at both the protein and mRNA levels in non-small cell lung cancer tissues and their adjacent tissues.Methods In this study,immunohistochemical techniques were employed to systematically examine the expression levels of MAP2 and β-tubulin in 252 non-small cell lung cancer tissue specimens and 68 corresponding adjacent tissue specimens.Real-time PCR was used to detect the mRNA levels of MAP2 and β-tubulin in 32 non-small cell lung cancer tissues and 32 adjacent tissues.Results The results of immunohistochemical detection showed that the positive expression rates of MAP2 and β-tubulin in the non-small cell lung cancer group were significantly higher than those in the adjacent tissue group,and the difference between the two groups was statistically significant(P<0.01).The results of Real Time PCR indicated that the mRNA levels of MAP2 and β-tubulin in the non-small cell lung cancer group were higher than those in the adjacent tissue group,and the difference between the two was statistically significant(P<0.05).Conclusion The expression levels of MAP2 and β-tubulin in non-small cell lung cancer tissues were significantly higher than those in adjacent tissues.This suggests that both MAP2 and β-tubulin are specifically present in lung tumor tissues.These expression differences provide meaningful indicators for the molecular diagnosis of non-small cell lung cancer and may potentially serve as new targets for anti-tumor treatment.
Association Between Iron Overload and Metabolic Dysfunction-Associated Steatotic Liver Disease:Based on NHANES and Transcriptomic DataAbstract:Objective By integrating epidemiological and transcriptomic data,this study investigates the relationship between iron overload and metabolic dysfunction-associated steatotic liver disease(MASLD),explores the underlying molecular mechanisms linking iron overload to the onset and progression of MASLD,and provides a scientific basis for clinical targeted diagnosis,treatment,or interventions.Methods Utilizing data from the NHANES 2017-2020,multivariable Logistic regression analysis was employed to assess the relationship between iron overload and the risk of MASLD.Multivariable linear regression was used to evaluate the association between iron overload and four types of liver enzymes.Subsequently,GEO dataset(GSE126848)and FerrDb V2 data were analyzed.Weighted Gene Co-expression Network Analysis(WGCNA)was applied to identify modules associated with MASLD.Protein-Protein Interaction(PPI)network was constructed to identify key genes.Results This study ultimately included 2010 participants:769 in the MASLD group and 1241 in the non-MASLD group.The results of multivariate Logistic regression analysis showed that after adjusting for age,gender,ethnicity,smoking status,and BMI,iron overload was associated with the risk of MASLD(OR=1.315,95%CI:1.041-1.661,P=0.022).Under the same adjusted variables,multiple linear regression analysis was used and it was found that iron overload was correlated with elevated levels of ALT,AST and GGT(P<0.001).A total of 2,077 MASLD-related genes were obtained from the GSE126848 dataset of the GEO database,and 484 genes related to iron overload were finally determined from the FerrDb database.After analysis by the WGCNA and PPI networks,five potential core genes simultaneously related to iron overload and MASLD were finally obtained:TGFB1,HMOX1,HSPA5,CDKN1A and CTSB.Conclusion This study discovered the correlation between iron overload and MASLD as well as liver enzyme levels,analyzed and identified potential key genes related to iron overload and MASLD,and explored the potential functional mechanisms between iron metabolism and MASLD at the molecular level.
Effect of Risperidone Combined with Aerobic Endurance Training on Cognitive Function,Psychological Status,and Sleep Quality in Patients with Schizophrenia Sleep DisordersAbstract:Objective To explore the effects of risperidone combined with aerobic endurance training on cognitive function,psychological status,and sleep quality in patients with schizophrenia sleep disorders.Methods A total of 102 research subjects were patients with schizophrenia accompanied by sleep disorders who were hospitalized and diagnosed in Luoyang NO.5 People's Hospital from March 2022 to March 2024.They were divided into the observation group(n=51)and the control group(n=51)by the random number table method.The control group was treated with risperidone,and the observation group received aerobic endurance training on the basis of the control group.The intervention lasted for 4 weeks.The brief psychiatric rating scale(BPRS)was used to evaluate mental symptoms in the past week,the Wisconsin card sorting test(WCST)was used to evaluate cognitive function,the self-rating anxiety scale(SAS)and self-rating depression scale(SDS)were used to evaluate mental state in the past week,and the Pittsburgh sleep quality index(PSQI)was used to evaluate sleep quality.Results After 4 weeks of intervention,significant reductions were observed in all factor scores and the BPRS total score for both groups(P<0.05),with the observation group demonstrating significantly lower scores than the control group(P<0.05).Concurrently,cognitive performance improved:both groups showed significant increases in the number of categories completed and correct responses,alongside significant decreases in errors and perseverative errors(P<0.05),with the observation group outperforming the control group on all these measures(P<0.05).Furthermore,significant reductions were also seen in SAS,SDS,and PSQI scores for both groups(P<0.05),again with significantly lower scores in the observation group compared to the control group(P<0.05).Conclusion For patients with schizophrenia accompanied by sleep disorders,risperidone combined with aerobic endurance training has significant effects in alleviating clinical symptoms,reducing cognitive impairment,combating anxiety and depression,and improving sleep quality,and has clinical application value.
