The Expression Relationship of HIF-1α,MMP-2,and MMP-14 in Bladder Urothelial Carcinoma

Abstract:Objective To investigate the expression levels and clinical significance of hypoxia inducible factor-1α(HIF-1α),matrix metalloproteinase-2(MMP-2),and matrix metalloproteinase-14(MMP-14)in bladder and urinary tract epithelial cancer.Methods 57 cases of bladder urothelial carcinoma tissues(observation group)and 15 cases of non tumor mucosal tissues(control group)were selected.Protein expression was detected by immunohistochemistry(IHC),and semi quantitative scoring(staining intensity x positive area ratio)was used to analyze expression differences.The tissue morphology was evaluated by HE staining.Spearman correlation analysis was used to explore the relationship between HIF-1 α,MMP-2,and MMP-14.Results The semi quantitative scores of HIF-1 α,MMP-2,and MMP-14 in the observation group were significantly higher than those in the control group(all P<0.05),and were positively correlated with tumor grading,vascular invasion,maximum diameter,and proliferation index(P<0.05).Spearman analysis showed a positive correlation between HIF-1 α and MMP-2,MMP-14.HE staining showed significant heterogeneity in cancer tissue,and immunohistochemical images confirmed high protein expression.Conclusion The high expression of HIF-1 α,MMP-2,and MMP-14 may promote the progression of bladder urothelial carcinoma by regulating extracellular matrix degradation and tumor microenvironment,and these three factors can serve as potential biomarkers and therapeutic targets.
