[Journal Article]WANG Zhuxian, CHEN Qiuyu, ZHOU Sijie et al.-Journal of Diagnosis and Therapy on Dermato-venereology2026, No.01

Abstract:Objective To investigate the therapeutic effects and mechanisms of blue light com-bined with gel for skin photoaging in mice. Methods Twenty-five male C57/BL6 mice were ran-domly divided into five groups:blank control, model, blue light, gel, and blue light+gel(com-bination)groups,with five mice per group.Except for the blank control group,a UVA and UVB synergistic irradiation method was used to establish a photoaging model.Mice were irradiated with 1 minimal erythema dose(MED)from week 1 to week 4,followed by 2 MEDs in weeks 5 and 6.Concurrently,starting from the third week, blue light combined with gel therapy was administered on the last day of each week. The control group received no treatment. The gel group received on-ly gel treatment for 6 minutes. The blue light group received only blue light irradiation for 6 mi-nutes.The combined group received gel treatment, followed immediately by blue light therapy. After 6 weeks, dorsal skin samples were collected for hematoxylin and eosin(HE),and Masson's trichrome staining. Both cutaneous and serum levels of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),and tumor necrosis factor(TNF-α)were measured using ELISA.Results After 6 weeks:The mouse skin exhibited desquamation, dryness, roughness, lesions, leathery texture, and hyperpigmentation.Either the blue light or gel group showed no significant improve-ments in skin desquamation, dryness, roughness,or leathery texture compared with the model group. Moreover, expression levels of SOD,GSH-Px,and collagen were not significantly upregu-lated compared with the model group(P>0.05).However,the combined group showed signifi-cant improvement in wrinkles, dryness, desquamation, and leathery texture. The expression levels of SOD and GSH-Px in the skin were significantly upregulated to levels comparable to normal(P<0.05),accompanied by a significant increase in collagen(P<0.001).TNF-αexpression in the skin,as well as the serum levels of SOD,GSH-Px,and TNF-α,remained no significant change(P>0.05).Conclusions Blue light combined with gel can alleviate oxidation, reduce inflammation, and increase collagen in photoaged skin, providing experimental evidence for its clinical application in photoaging management.

[Journal Article]YAN Ruotian, HU Honghua, ZHU Chengyao-Journal of Diagnosis and Therapy on Dermato-venereology2026, No.01

Abstract:We report two cases of misdiagnosed subungual squamous cell carcinoma(SSCC).Case 1:A 50-year-old man presented with recurrent swelling and purulent discharge under the left middle fingernail for two years. The patient had been diagnosed with paronychia and pyogenic granuloma.Dermatological examination revealed a red, nodular, granulomatous lesion under the left middle fingernail, approximately 0.5 cm×0.3 cm×0.3 cm in size, with well-defined bor-ders.A partial nail unit excision was performed, and postoperative histopathology showed multiple tumor cell clusters in the dermis that were connected to the epidermis, accompanied by the forma-tion of squamous eddies and horn pearls. The tumor cells exhibited marked atypia and disordered arrangement. The diagnosis was subungual squamous cell carcinoma.Case 2:A 66-year-old woman presented with a painful subungual nodule on the left index finger for several months. She had been diagnosed with verruca vulgaris and subungual fibroma.Dermatological examination showed a red,soybean-sized nodule under the left index fingernail that protruded from the nail plate and the fingertip, with a rough and crusted surface.A partial nail unit excision was per-formed,and postoperative histopathology showed atypical tumor cell clusters connected to the epi-dermis,with the presence of mitosis and dyskeratosis.Postoperative diagnosis:Subungual squa-mous cell carcinoma. Both patients have been followed up after surgical excision and have not shown any signs of recurrence to date.

ANKRD22 recombinant protein attenuates psoriasis in mice through inhibiting noncanonical NF-κB signaling pathway
[Journal Article]LIANG Kaiyu, JI Suyun, XIA Xichun-Journal of Diagnosis and Therapy on Dermato-venereology2025, No.12

Abstract:Objective To elucidate the anti-inflammatory effects and underlying molecular mechanisms of recombinant ANKRD22 protein in dendritic cells, and to assess its therapeutic effi-cacy in mitigating the progression of psoriasis. Methods In vitro, mouse bone marrow cells isola-ted from femurs and tibias were induced into mature bone marrow-derived dendritic cells(BMDC)using GM-CSF.The BMDC were divided into four groups:PBS,IMQ,PBS+recombinant ANKRD22,and IMQ+recombinant ANKRD22.Each group was treated accordingly with PBS,IMQ,or recombinant ANKRD22 protein for 24 h.Quantitative reverse transcription PCR(qRT-PCR)was used to measure the expression levels of mRNA for IL23a,IL1 b,and ANKRD22,while Western blot was performed to detect the protein levels of key members of the noncanonical NF-κB signaling pathway. In vivo,an IMQ-induced mouse model of psoriasis was established(n=10)and divided into a PBS control group(n=5)and a recombinant ANKRD22 group(n=5).The changes in body weight and PASI scores were recorded daily.Histological changes and acanthosis area were evaluated by hematoxylin and eosin(H&E)staining.Flow cytometry was used to determine the proportions of neutrophils,Th17,and γδT17 cells, and qRT-PCR was used to quantify the expression levels of mRNA for IL1 b,IL23a,IL17a,IL17f,IL6 and TNF-α in the skin. Results In vitro, compared with the PBS group, expression levels of IL23a,IL1 b and ANKRD22 mRNA in the IMQ group significantly increased to 1.97±0.33(P=0.007),1.80±0.32(P=0.014)and 2.34±0.52(P=0.014)fold,accompanied by enhanced NIK accumulation and downstream p52/RelB nuclear translocation. In contrast, compared with the IMQ group,IL23a expression in the IMQ+recombinant ANKRD22 group was significantly reduced, from 1.97±0.33 to 1.17±0.19-fold(P=0.023),without significant differences in the expression levels of IL1 b and ANKRD22 mRNA.Concurrently,NIK accumulation and downstream p52/RelB nuclear translocation were markedly attenuated. In vivo, recombinant ANKRD22 significantly alle-viated psoriasis-like symptoms. On day 5,the PASI score was significantly lower in the recombi-nant ANKRD22 group than in the PBS group(6.10±0.65 vs.7.80±0.83,P=0.007),acan-thosis area was reduced from 0.08±0.01 to 0.04±0.01 mm2(P=0.001),and splenomegaly was mitigated with an increased body weight(20.24±0.51 g vs.19.06±0.45 g,P=0.005).The proportions of neutrophils,Th17,and γδT17 cells in the skin decreased from(21.00±3.07)%,(38.16±3.66)%,and(9.74±3.02)%to(6.48±1.96)%(P=0.001),(24.48±3.37)%(P=0.001),(6.42±1.55)%(P=0.030),respectively.The expression levels of mRNA for IL1 b,IL23a,IL17a,IL17f,IL6 and TNF-α in the skin were also reduced from(32.67±17.34)%to(67.37±25.43)%.Conclusions Recombinant ANKRD22 protein sup-presses IL-23 production by selectively inhibiting the noncanonical NF-κB signaling pathway in dendritic cells, thereby blocking the IL-23-Th17/γδT17 immune axis, promoting inflammatory resolution, and effectively ameliorating psoriasis progression.

The role of specialized pro-resolving mediators in skin inflammation
[Journal Article]WANG Yuying, LIN Simin, WANG Xiaoying et al.-Journal of Diagnosis and Therapy on Dermato-venereology2025, No.12

Abstract:Specialized pro-resolving mediators(SPMs)are endogenous lipid molecules that are biosynthesized from ω-3/ω-6 polyunsaturated fatty acids via key enzymes such as lipoxygenases and cyclooxygenases.By targeting specific receptors on the surfaces of immune cells,SPMs pre-cisely regulate inflammation resolution and play a crucial role in maintaining skin homeostasis. This review delineates the dynamic changes and regulatory roles of SPMs in various physiological and pathological processes, including psoriasis, atopic dermatitis, skin wound healing, and skin aging.Specifically, certain SPMs,such as RvD5 and PDx,are detectable in psoriatic lesions, while an imbalance between pro-inflammatory mediators and SPMs is observed in the skin of pa-tients with atopic dermatitis.During skin wound healing, the content of the resolvin D series in-creases,promoting inflammation resolution and tissue repair. In the process of skin aging, the downstream metabolism of EPA/DHA in aged skin fails to effectively produce active SPMs,lead-ing to a decline in the skin's ability to resolve inflammation.Furthermore, this article elucidates the role of SPMs in alleviating skin inflammatory symptoms such as pain, pruritus, edema, and er-ythema through mechanisms that include regulating transient receptor potential(TRP)channels, inhibiting the release of pro-inflammatory factors, and modulating vascular permeability. It pro-vides a theoretical basis for the clinical application of novel skin anti-inflammatory strategies based on SPMs or their analogs.

[Journal Article]YANG Yuying, FANG Yali, LI Yuan-Journal of Diagnosis and Therapy on Dermato-venereology2025, No.12

Abstract:Acne is a common chronic, inflammatory disease of the pilosebaceous glands. Its pathogenesis is complex and affected by many factors. In recent years, the research on the patho-genesis of acne has been deepened. As a pleiotropic growth factor,insulin-like growth factor-1(IGF-1)plays an important role in the pathogenesis of acne.Studies have shown that IGF-1 can promote the proliferation and lipid synthesis of sebaceous gland cells by activating PI3K/Akt/mTORC1 signaling pathway, while also increasing androgen receptor sensitivity and aggravating se-baceous hypersecretion. This article reviews the current research progress on the role of IGF-1-me-diated inflammation in the pathogenesis of acne.

Research progress on the role of ferroptosis in the pathogenesis of vitiligo
[Journal Article]ZHUO Xiujuan, HUANG Xiuyun, SHI Xiaowei et al.-Journal of Diagnosis and Therapy on Dermato-venereology2025, No.12

Abstract:Vitiligo is a common depigmenting skin disorder with a complex pathogenesis,pri-marily resulting from the destruction, death,or dysfunction of melanocytes. In recent years,fer-roptosis,a novel iron-dependent form of cell death driven by lipid peroxidation, has been recog-nized as playing a significant role in the onset and progression of vitiligo.Factors such as iron ho-meostasis imbalance, lipid peroxidation accumulation, weakened antioxidant defenses, and activa-tion of immune inflammation collectively contribute to melanocyte damage. This review systemati-cally examines the physiological processes of iron metabolism, explores the molecular mechanisms of ferroptosis in melanocyte injury and its role in vitiligo pathogenesis, and summarizes potential therapeutic targets and drug interventions related to ferroptosis. An in-depth exploration of the in-teractions between ferroptosis and immune regulation provides novel insights and a strong theoreti-cal foundation for the precise prevention and treatment of vitiligo.

Abstract:The invasion of Treponema pallidum can affect multiple structures of the eyes, thereby leading to the development of ocular syphilis. The pathogenesis of ocular syphilis is closely related to the activity of mucopolysaccharide enzymes on the surface of Treponema pallidum. The clinical manifestations are complex and diverse, and often lack specificity. It may present as con-junctivitis,keratitis,uveitis,optic neuritis, etc.Diagnostic methods include ophthalmic function-al examinations, ophthalmic morphological examinations, ophthalmic imaging studies, and labora-tory tests.Treatment primarily relies on the use of antibiotics, and corticosteroids may occasionally be added. There is a close connection between ocular syphilis and neurosyphilis, and in cases of co-infection with HIV,the prognosis of patients is usually poor. This article reviews the progress in the diagnosis and treatment of ocular syphilis, providing reference for clinical diagnosis and treatment.

Abstract:Objective To observe the clinical efficacy of glycolic acid combined with skin bar-rier function repair in patients with facial melasma. Methods Sixty-four patients with facial melasma treated at the Department of Dermatology,Yantian District People's Hospital,Shenzhen from January 2024 to June 2024 were randomly divided into two groups(32 patients per group). The control group was treated with glycolic acid once every four weeks. The treatment group re-ceived a combined therapy of glycolic acid and skin barrier function repair, with glycolic acid treatment once every 4 weeks and desensitization treatment once every two weeks. After 16 weeks of treatment, the changes in the melasma area and severity index(MASI)scores were evaluated, and the effective rate and patient satisfaction were compared between the two groups. Results Af-ter 16 weeks of treatment, the MASI score in both groups was decreased, with a more prominent decrease in the treatment group(P=0.035 vs.the control group). The effective rate in the treat-ment group was higher than that in the control group(87.50%vs.71.88%,P<0.05),and pa-tient satisfaction rate was also higher in the treatment group than in the control group(87.50%vs.75.00%,P<0.05).Conclusion The combination of skin barrier function repair and glycolic acid has a good therapeutic effect on melasma, along with a higher patient satisfaction rate.
