Study on teaching effect and optimization strategy of Clinical Epidemiology and Evidence-based Medicine from cognitive-behavioral integration

Abstract:Objective To assess the knowledge,attitudes,and teaching needs of the course Clinical Epidemiology and Evidence-based Medicine(CEEBM)in medical undergraduate students,and to provide evidence for curriculum reform.Methods A cross-sectional online survey was conducted among clinical medicine undergraduates using Wenjuanxing from the Clinical College of Anhui Medical University.The questionnaire covered pre-course knowledge,perceived learning outcomes,self-directed behaviors,and course evaluation,multivariate ordered Logistic regression analysis was used to analyze the influencing factors of the utility value of the course.Results A total of 714 valid questionnaires were collected,56.30%were male(402 students)and 43.70%were female(312 students).Before the course,31.65%reported understanding clinical epidemiology,while only 16.53%had knowledge of Evidence-based Medicine(EBM).After completing the course,63.17%felt they basically or completely mastered the course content,and 68.49%of students have a basic or complete understanding the relationship between EBM and clinical epidemiology.Multivariate ordinal Logistic regression analysis showed that self-assessed learning ability,prior knowledge of clinical epidemiology and EBM,and course mastery were significant factors influencing the evaluation of the course's effectiveness(P<0.05).Regarding learning behaviors,48.60%of students actively used library resources to retrieve and read literature,and 65.55%indicated they would integrate evidence-based knowledge with clinical experience in their future practice.However,only 15.97%of students expressed an intention to participate in related research projects.Course evaluation showed that 95.66%of students recognized the necessity of offering this course within the clinical medicine curriculum,with a preference for flexible assessment methods such as open-book exams(57.98%)and review writing(14.71%).Conclusion The CEEBM course has a significant effect on improving students'professional cognition and evidence-based thinking.While students generally appreciated the course,their willingness to engage in research and active learning behaviors remains limited.This highlights the need to further enhance the course's practical orientation and research engagement components,as well as adjust the assessment methods to better promote students'autonomous learning and critical thinking.

The impact of predictive nursing intervention based on modified early warning score on the prognosis of patients with acute myocardial infarction:a Meta-analysis

Abstract:Objective To systematically evaluate the impact of predictive nursing intervention based on Modified Early Warning Score(MEWS)on the prognosis of patients with acute myocardial infarction(AMI).Methods PubMed,Embase,Web of Science,CNKI,Wanfang Data,and VIP Database were electronically searched to collect relevant randomized controlled trials(RCTs)on the effect of MEWS-based predictive nursing intervention in AMI patients published from inception to December 2024.Meta-analysis was performed using RevMan 5.3 software.Results A total of 25 RCTs involving 2,430 patients were included.The Meta-analysis results showed that the triage time,emergency rescue time,and length of hospital stay in the MEWS intervention group were all shortened compared with conventional nursing.The patients'heart rate,left ventricular ejection fraction,left ventricular end-systolic diameter,and left ventricular end-diastolic diameter improved significantly.The incidence of adverse events was low and nursing satisfaction was high,but no significant difference was observed between the two groups in terms of anxiety and depression indicators.Conclusion MEWS-based predictive nursing has clear advantages in improving cardiac function,clinical diagnostic and therapeutic indicators,incidence of adverse events,and nursing satisfaction in AMI patients.However,its effect on improving patients'psychological status is not significant.Future studies should focus on developing intelligent early warning systems integrated with biomarker monitoring and embedding psychological nursing modules into the MEWS framework to achieve precise intervention in both physical and psychological dimensions.

Project characteristics and correlation analysis of out-of-hospital nursing service demand of patients with enterostomy

Abstract:Objective To analyze the project characteristics and correlation of out-of-hospital nursing service demand of enterostomy patients,and provide reference for optimizing continuous nursing service of enterostomy patients.Methods From January 2022 to March 2023,enterostomy patients who were treated in Northern Jiangsu People's Hospital were selected as the research object.The quantitative research adopts questionnaire survey to collect patients'general information,the current situation of ostomy use and their needs for continuous nursing services,and uses Apriori correlation analysis method to analyze the relevance of various nursing service needs,and calculate the support,confidence and promotion of needs or demand sets;Qualitative research selected 21 patients with enterostomy for semi-structured interview according to stratified sampling method,based on the principle of theoretical saturation of data.Results The demand for out-of-hospital nursing service of enterostomy patients includes five dimensions:physiological function,psychological function,ostomy care,daily activities and social support.Enterostomy patients have a high demand for out-of-hospital nursing,among which the demand for ostomy care accounts for the highest proportion(79.92%),and also with highest average item score(4.00±0.47),with the support degree of 0.397 and the confidence degree of 0.917.The higher the education level,the higher the score of nursing service demand outside the hospital(P<0.05).In the end,three themes were condensed:Satisfaction with out-of-hospital nursing service;The opinions of enterostomy patients on out-of-hospital nursing service are mostly found in the problems of ostomy nursing and the use of ostomy bags;Understanding the potential demand of enterostomy patients of out-of-hospital service.Conclusion Patients with enterostomy have different nursing service needs after discharge.Clinicians should set up more optimized continuous nursing service scheme according to the main needs of enterostomy patients for out-of-hospital nursing service and the related characteristics of service needs,so as to improve the level of out-of-hospital nursing service for enterostomy patients.

The distribution of RhCE phenotypes and molecular genetic background of D variants in preliminary screening of RhD-negative blood donors

Abstract:Objective To explore the distribution of RhCE phenotypes and the molecular genetic background of D variants among preliminary screening RhD-negative blood donors.Methods Blood samples sent for testing to the Linyi Central Blood Station from July 2022 to June 2023 were collected.The Rh phenotype was detected using the test tube method,and RhD blood type confirmation was performed using a broad-spectrum anti-human globulin card(microcolumn method).For samples serologically confirmed as RhD variant,the RHD gene was tested using PCR-SSP.Sequencing analysis was performed on samples with positive amplification of all or part of the exons.Results A total of 564 cases were initially screened as RhD negative.The ABO blood type distribution was as follows:O type(32.98%),A type(27.48%),B type(26.06%),and AB type(13.48%).The confirmation results for RhD negative status showed 548 cases(97.16%)as RhD negative and 16 cases(2.84%)as RhD variants.The distribution of Rh phenotypes was ccee,Ccee,ccEe,CCee,CcEe and CCEe.The RhCE phenotypes of RHD variants all contained C or E antigens and there were several alleles which were previously reported.To be specific,DEL RHD1227A homozygous,weak D15,weak D15/DEL RHD1227A,DVI III,RHD-CE(5)-D and RHD-CE(5)-D/711delC were detected in 2 cases,6 cases,3 cases,1 case,3 cases and 1 case respectively.Conclusion The most common RhCE phenotype of RhD negative donors were ccee and Ccee.There was polymorphism in RHD gene with D variant,weak D15 and DEL RHD1227A were the most common.The RhCE phenotype of D variant was associated with RHD genotype.

Investigation and implementation exploration on knowledge,attitude and practice of influenza vaccination among college students

Abstract:Objective The purpose of this study is to understand the knowledge,attitude,and practice(KAP)of influenza vaccine inoculation and their influencing factors,among Chinese college students and further support the formulation of implementation plans to promote influenza vaccination.Methods From April 7th to May 31st,2024,a convenience sampling method was used to select college students in Beijing Jiaotong Unversity to complete a questionnaire survey.The content included basic information,cognition,attitude,behavior towards influenza virus and vaccine,and expected channels for science popularization,etc.Logistic regression analysis were used to explore factors influencing college students'KAP towards influenza virus vaccination.Results A total of 295 valid questionnaires from college students were collected.Female students and vaccination history were associated with a higher correct rate of knowledge,while infection history was associated with a lower correct rate of knowledge.Female students were more likely to recognize the severity of influenza virus infection,higher grade students and those with infection history believed they were more likely to be infected with influenza.Female students,students with vaccination history and third-year students were more willing to actively search and share health information about vaccination in daily life,students majoring in science were more likely to share health information about vaccination.Conclusion Numerous factors influenced college students'KAP level.The vaccine efficacy rate can be increased through effective measures such as improving the education and publicity of healthcare providers,establishing centralized and convenient vaccination sites,providing convenient vaccination times,and implementing vaccine cost reimbursement policies.

Prediction and analysis of red blood cells clinical demand based on the ARIMA model

Abstract:Objective To construct an ARIMA multiplicative seasonal model suitable for predicting the clinical demand of red blood cells in Wuhu City,and provide a scientific basis for blood collection organizations to formulate red blood cell collection and supply balance programs and recruitment plans.Methods The ARIMA model was constructed based on the clinical use of red blood cells in Wuhu City Central Blood Station from January 2012 to December 2021.The data were processed through time series stabilization,model identification,and parameter verification to determine the optimal model.The red blood cell clinical use demand from January 2022 to August 2022 was predicted using the optimal model,and the prediction effect was verified using actual values.Results The optimal model was ARIMA(0,1,0)(1,1,0)12,with BIC=12.162.The ACF and PACF of the residual sequence were basically within the 95%confidence interval,and the Ljung-Box Q value was 15.265,with P=0.576>0.05,which met the requirements of white noise sequence,and the model fitting was validated.Except for April and May,the actual values of each month were within the 95%confidence interval of the predicted value,and the average relative error between the ARIMA model prediction value and the actual value was-0.00375,and the mean absolute percentage error(MAPE)was 7.087%,with good prediction effect.Conclusion The ARIMA(0,1,0)(1,1,0)12 model can be used to predict the clinical demand of red blood cells,and it can provide a reference for blood collection,non-remunerated blood donation recruitment and inventory management.

Efficacy of dexmedetomidine in mechanical ventilation for ARDS:a Meta-analysis

Abstract:Objective Systematic evaluation of the efficacy of dexmedetomidine for mechanical ventilation in acute respiratory distress syndrome(ARDS).Methods PubMed,Embase,Cochrane Library and CNKI were searched from the time of inception to June 25,2025.Two researchers independently performed literature screening,data extraction,and quality assessment according to predefined inclusion and exclusion criteria.Meta-analysis was conducted using Stata 12.0 software.Results A total of 9 randomized controlled trials were included.The results showed that dexmedetomidine alone or in combination with other drugs significantly reduced the level of inflammatory factors IL-6[WMD=-1.13,95%CI(-1.92,-0.35)]and TNF-α[WMD=-0.87,95%CI(-1.34,-0.4)],improved oxygenation index[WMD=1.83,95%CI(0.64,3.03)],shortened ICU length of stay[WMD=-1.44,95%CI(-1.76,-1.12)]and mechanical ventilation duration[WMD=-0.93,95%CI(-1.26,-0.95)],the differences were statistically significant(P<0.05).However,there was no significant difference in MAP levels[WMD=0.39,95%CI(-0.20,0.98)].Conclusion In mechanically ventilated ARDS patients,dexmedetomidine reduces acute stress-induced inflammation,improves oxygenation index,shortens ICU stay,decrease mechanical ventilation duration.These findings provide clinical evidence supporting the use of dexmedetomidine in ARDS patients undergoing mechanical ventilation.

Assessment of current status and influencing factors of financial toxicity among elderly hypertensive patients in rural Zhuzhou

Abstract:Objective To explore the status of financial toxicity and its influencing factors among elderly hypertensive patients in rural areas,and to provide a reference for implementing effective interventions.Methods Using a random number table method,registered elderly hypertensive patients from a rural area of Zhuzhou,Hunan Province,were selected as the study subjects.The Incharge Financial Distress/Financial Well-being Scale,the Hypertensive Patients Self-management Behavior Scale,the Morisky Medication Adherence Scale,and the EuroQol Five Dimensions(EQ-5D)Health Questionnaire were used for assessment.A random forest model was employed to identify the importance of influencing factors,combined with Lasso regression and multiple linear regression to determine the main influencing factors.Results A total of 240 valid questionnaires were collected.The mean total financial toxicity score was(4.96±1.46)points,with 64.6%of patients experiencing moderate financial toxicity and 22.5%experiencing high financial toxicity.The random forest model indicated that variables such as self-rated health status,medication adherence,and self-management behaviors significantly influenced financial toxicity.Multiple linear regression analysis further identified self-rated health status,medication adherence,self-management behaviors,hypertensive complications,daily exercise time,duration of having a family-contracted doctor,and serum total cholesterol as the main influencing factors for financial toxicity(P<0.05),collectively explaining 51.3%of the total variance.Conclusion The level of financial toxicity is relatively high among elderly hypertensive patients in rural Zhuzhou.Healthcare professionals should develop targeted intervention strategies based on the related influencing factors to improve patients'financial toxicity levels.

Impact of assisted reproductive technology on fetal development

Abstract:Assisted reproductive technology(ART)offers infertile patients and couples carrying hereditary diseases the possibility of achieving normal conception and delivering healthy offspring.However,ART is also associated with certain risks of complications.This paper primarily elucidates the potential factors and mechanisms through which ART may induce epigenetic alterations,particularly abnormalities in DNA methylation.It further explores the adverse effects of DNA methylation abnormalities at different genomic loci on fetal development,including an increased incidence of placental abnormalities,a higher risk of preeclampsia,and elevated rates of adverse perinatal outcomes.In addition,several improvement strategies are proposed,such as supplementation with moderate doses of folic acid and the implementation of single-embryo transfer,aiming to reduce ART-related epigenetic abnormalities and the risk of unfavorable pregnancy outcomes.

Abstract:The kynurenine pathway(KP)is the primary route of tryptophan metabolism in most mammalian cells,with approximately 95%of dietary tryptophan being converted into kynurenine and its downstream metabolites.Kynurenine is a central metabolite of tryptophan and possesses anti-inflammatory,antioxidant,and neuroprotective properties.Current research has shown that the KP plays a role in various diseases such as keratitis,cataract,glaucoma,and diabetic retinopathy.It was deeply involved in ocular pathophysiology through multiple mechanisms,including immunomodulation,antioxidant activity,neuroprotection,and mediation of photodamage.This paper presents a review of the role of KP in the eye and their related mechanisms in ocular diseases,including keratitis,cataract,glaucoma,and diabetic retinopathy,along with an outlook on future research.

Abstract:Virtual simulation(VS)is a hot issue in the field of higher education.At present,VS has been applied to experimental teaching in universities.In order to promote the progress of VS teaching of experimental animal science,this paper summarizes the characteristics,defects and deficiencies of existing software courses in the market,and introduces a variety of simulation objects of VS courses of medical experimental animal science.It includes the audience of the course and job roles,model animals and disease models,and laboratory facilities and equipment and instrument parameters.At the same time,this paper also discusses the specific content of VS used for teaching curriculum,such as animal breeding and feeding,infected and non-infected animal experiments,facility and equipment overhaul and maintenance,animal experiment safety and admission training,etc.,and gives professional advice on updating and optimizing the VS curriculum.

Exploring the causal association of immunophenotypes on gestational diabetes mellitus based on Mendelian randomization

Abstract:Objective To explore the causal relationship between immune cell phenotypes and the risk of developing gestational diabetes mellitus(GDM)by using Mendelian randomization(MR).Methods Using genome-wide association study data from European populations,we extracted 731 immune cell traits as instrumental variables,and obtained GDM summary statistics from the Finnish database(FinnGen).The inverse-variance weighted(IVW)method served as the primary estimator,supplemented by the weighted median,MR-Egger regression,simple mode,and weighted mode to evaluate the causal relationship between immune cell phenotypes and GDM,with additional tests for heterogeneity,horizontal pleiotropy,and sensitivity analyses.Reverse MR was further conducted to assess the potential causal impact of GDM on immune phenotypes.Results A total of 40 immune phenotypes showed potential causal associations with GDM risk.After false discovery rate correction(FDR<0.05),two phenotypes remained significant:HLA DR on CD33-HLA DR+[OR=1.107,95%CI(1.050,1.166)]and HLA DR on DC[OR=1.098,95%CI(1.048,1.150)].Reverse MR revealed no significant causal impact of GDM on any of the 40 immune phenotypes(P>0.05).Conclusion Specific immune cell phenotypes(HLA DR on CD33-HLA DR+,HLA DR on DC)may be potential causal risk factors for the development of GDM and may provide targets for future risk prediction and immune intervention strategies.

Analysis of the disease burden and future trends of appendicitis in China from 1990 to 2021

Abstract:Objective To analyze the disease burden of appendicitis in China from 1990 to 2021 and to predict the future trends.Methods The incidence,mortality,disability-adjusted life-years(DALY)and corresponding age-standardized rates of appendicitis in China from 1990 to 2021 were extracted from the 2021 Global Burden of Disease Study database.The annual percentage change and average annual percentage change(AAPC)were calculated using Joinpoint software to analyze the changing trend of the disease burden of appendicitis in China from 1990 to 2021.A Bayesian age-period-cohort(BAPC)model was constructed to predict the changes in the disease burden of appendicitis in China from 2022 to 2040.Results The age-standardized incidence rate(ASIR),age-standardized mortality rate(ASMR),and age-standardized DALY rate(ASDR)of appendicitis in China in 2021 were 311.67/100 000,0.09/100 000,and 5.81/per 100 000 person years,respectively.From 1990 to 2021,the ASIR[AAPC=0.75%,95%CI(0.71%,0.80%)]for appendicitis in China showed an increasing trend,while the ASMR[AAPC=-5.32%,95%CI(-5.38%,-5.26%)]and ASDR[AAPC=-3.51%,95%CI(-3.55%,-3.48%)]both showed a decreasing trend.Women had a higher incidence rate than men,but lower mortality and DALY rates.The incidence rate of appendicitis in China initially increased and then decreased with age,with the peak age of onset always between 15~24 years.Deaths were primarily among the elderly,with the peak age of death being over 85 years.The peak age of the DALY rate of appendicitis in China was concentrated between the ages of 90 and 94.A BAPC model predicted that the ASIR of appendicitis in China would increase from 2022 to 2040,while the ASMR and ASDR showed a decrease trend.Conclusion From 1990 to 2021,the ASIR of appendicitis in China increased,while the ASMR and ASDR decreased,and the overall disease burden showed a slow decline in the future.Currently,efforts should be made to strengthen prevention and treatment efforts among the elderly,vigorously carry out scientific education,and promote healthy lifestyles and dietary patterns to further reduce the disease burden of appendicitis.

Identification of potential drug targets for restless legs syndrome by Mendelian randomization analysis based on plasma proteomics

Abstract:Objective To evaluate the causal relationship between plasma proteins and restless legs syndrome(RLS)by proteome-wide Mendelian randomization(MR)and to explore new therapeutic targets for RLS.Methods Genome-wide association studies(GWAS)data of plasma proteins were obtained from the UK Biobank,and GWAS data of RLS were extracted from the FinnGen R11 database.Colocalization analysis was performed to identify the common causal variants between plasma proteins and RLS.Results MR analysis showed that plasma proteins ASPSCR1[OR=0.678,95%CI(0.544,0.847)]and C1QTNF5[OR=0.526,95%CI(0.367,0.752)]had a protective effect on RLS;plasma proteins KRT19[OR=1.769,95%CI(1.283,2.439)]and LAT2[OR=5.536,95%CI(2.206,13.893)]were associated with the increased risk of RLS.Colocalization analysis showed that among the 4 plasma proteins,LAT2 had a strong causal variation relationship,while ASPSCR1,C1QTNF5,and KRT19 had a moderate causal variation relationship.The protein functional network suggested that protein-associated genes related to RLS were significantly enriched in sugar transmembrane transporter activity,carbohydrate transmembrane transport activity,and Fc receptor signaling pathway.Conclusion This study identifies the causal relationship between 4 plasma proteins and RLS,which provides potential therapeutic targets for the treatment of RLS,but further studies are needed to confirm it.

Meta-analysis of the prevalence and influencing factors of medication discrepancies among cardiovascular disease patients in China

Abstract:Objective To systematically evaluate the incidence and influencing factors of medication discrepancies in patients with cardiovascular disease(CVD)in China.Methods CNKI,Wanfang,VIP,SinoMed,PubMed,Embase,Web of Science,CINAHL,and the Cochrane Library databases were searched from their inception to December 2024.Literature on the incidence and influencing factors of medication discrepancies in patients with CVD in Chinese was included.Meta-analysis was performed using R 4.3.2 software.Results A total of 22 articles were included,involving 6,278 patients.Meta-analysis results showed that the prevalence of medication discrepancies among Chinese CVD patients was 67%[95%CI(56%,76%)].High monthly household income,high education level,good family functioning,living with others,fewer combined chronic diseases,a low Charlson comorbidity index,and male sex were protective factors for medication discrepancies,while a high number of medications used and the presence of adverse drug reactions were risk factors.Conclusion The prevalence of medication discrepancies among Chinese patients with CVD is high.Clinicians should identify high-risk patients based on these factors and conduct early screening and intervention to prevent medication discrepancies.

The incidence and influencing factors of combined pulmonary infections in patients with tumor chemotherapy:a Meta-analysis

Abstract:Objective To systematically evaluate the incidence and influencing factors of pulmonary infections in chemotherapy patients.Methods CNKI,Wan Fang,VIP,CBM,PubMed,Embase,The Cochrane Library,and Web of Science were searched to collect the literature on influencing factors for lung infections in chemotherapy patients,with a timeframe from the inception of the database to July 10,2025.Two investigators independently screened the literature according to the inclusion and exclusion criteria,and the Meta-analysis was performed using Stata 16.0 software.Results A total of 23 studies covering 8,800 patients were selected,with high literature quality scores.Meta-analysis showed that the incidence of combined pulmonary infection in patients with tumor chemotherapy was 24.4%,with 19.6%for lung cancer,14.1%for esophageal cancer,and 44.6%for hematologic cancer.Age,age≥60 years,history of smoking,comorbid diabetes,comorbid chronic obstructive pulmonary disease(COPD),tumor location in the mid-thoracic region,late tumor staging,chemotherapy cycles≥2,combination of chemotherapeutic agents,presence of invasive operation,duration of radiotherapy≥30 days,neutrophil count reduction,hypoproteinemia,and severe anemia were independent risk factors for pulmonary infection in chemotherapy patients.Conclusion This systematic review identified 14 significant risk factors that are common clinical phenomena in chemotherapy patients.These factors should be monitored by clinical workers for early identification of high-risk individuals and enhanced assessment and care to reduce the incidence of pulmonary infection.

Abstract:Objective To investigate the potential associations between lncRNAs and the risk of oral squamous cell carcinoma(OSCC)and oral precancerous lesions(oral leukoplakia and oral lichen planus)using a two-sample Mendelian randomization(MR)approach.Methods Expression profiles were analyzed from the Gene Expression Omnibus database(GSE23558 and GSE85195)to identify differentially expressed lncRNAs.Using the eQTLGen Consortium's cis-eQTLs dataset and genome-wide association analysis data for OSCC and oral precancerous lesions,five two-sample MR models,including inverse variance weighting and MR-Egger regression,were used to assess the causal effects of lncRNAs on OSCC and oral precancerous lesions,using single-nucleotide polymorphisms as instrumental variables.Heterogeneity tests,pleiotropy tests,and sensitivity analyses were performed to assess the reliability and stability of the results.False discovery rate(FDR)and Bonferroni correction were applied to adjust the results.Results MR analysis indicated that HCG22[OR=0.999,95%CI(0.999,1.000)]may reduce the risk of OSCC,FAM30A[OR=1.001,95%CI(1.000,1.002)]may increase the risk of OSCC,and FAM182B[OR=0.738,95%CI(0.581,0.937)]may reduce the risk of oral leukoplakia.However,FDR and Bonferroni analysis confirmed that these results may be false positives.Sensitivity analysis revealed no significant heterogeneity or pleiotropy bias,suggesting that the results are robust.Conclusion Current genetic evidence does not support a robust causal relationship between the selected lncRNAs and OSCC or its precancerous lesions.This study provides preliminary genetic insights into the role of lncRNAs in the development of oral precancerous lesions.However,future studies with larger sample sizes,functional validation,and multi-omics integration are needed to further explore their biological mechanisms and clinical potential.

Effect of regulating miR-29b-3p/FoxO3 axis by asiaticoside on alleviating liver injury in rats with non-alcoholic fatty liver disease

Abstract:Objective To investigate the effect of Asiaticoside(ASI)on liver injury in non-alcoholic fatty liver disease(NAFLD)and its impacts on the microRNA-29b-3p(miR-29b-3p)/forkhead box protein O3(FoxO3)axis.Methods Rats were stochastically grouped into a Control group,a Model group,a low-dose ASI-L group(ASI-L),a high-dose ASI-H group(ASI-H),a high-dose ASI+blank vector group(ASI-H+Vector),and a high-dose ASI+miR-29b-3p over expression group(ASI-H+OV-miR-29b-3p),with 12 rats in each group.The changes in rat liver index were analyzed.HE staining method was applied to observe the pathological changes of liver tissue.Biochemical analyzer was used to detect lipid metabolism indicators and indicators reflecting liver parenchymal damage in rats.The levels of oxidative stress and inflammatory cytokines were detected by the ELISA method.The RT-qPCR method was applied to detect the mRNA levels of miR-29b-3p and FoxO3 in rat liver tissue.The Western blot method was applied to detect the expression levels of FoxO3 and apoptosis-related proteins in rat liver tissue.Dual luciferase activity was applied to validate the targeting relationship between miR-29b-3p and FoxO3.Results Compared with the Control group,rats in the Model group showed severe liver tissue damage,liver cell rupture,and extensive infiltration of inflammatory cells,the liver weight and liver index increased,the levels of TC,TG,AST,ALT,MDA,TNF-α,IL-6 obviously increased,the levels of SOD and CAT obviously reduced,the expression level of miR-29b-3p and expression level of Caspase 3 protein in liver tissue obviously increased,the expression level of FoxO3 mRNA and expression levels of FoxO3,Bcl-2 proteins decreased(P<0.05).Compared with the model group,as the dose of ASI increased,the degree of liver tissue damage,liver cell rupture,and inflammatory cell infiltration of rats in the ASI-L and ASI-H groups decreased,the liver weight and liver index gradually decreased,the levels of TC,TG,AST,ALT,MDA,TNF-α,IL-6 in serum gradually decreased,the levels of SOD and CAT gradually increased,the expression level of miR-29b-3p and expression level of Caspase 3 protein in liver tissue gradually decreased,the expression level of FoxO3 mRNA and the expression levels of FoxO3 and Bcl-2 proteins gradually increased.miR-29b-3p overexpression reversed the improvement effect of ASI on liver tissue in NAFLD rats.Dual luciferase activity confirmed the targeting relationship between miR-29b-3p and FoxO3.Conclusion ASI can enhance the antioxidant function,regulate lipid metabolism,alleviate inflammation and liver tissue damage in NAFLD rats,which may be related to the down-regulation of miR-29b-3p and the up-regulation of FoxO3 expression.

Analysis of the predictive value of event-related potential contingent negative variation combined with cognitive function indicators for chronic schizophrenia

Abstract:Objective To analyze the predictive value of event-related potential(ERP)contingent negative variation(CNV)induced by dot pattern expectancy(DPX)and cognitive function indicators in chronic schizophrenia(CS).Methods Behavioral data of all subjects were collected using DPX,CNV waves based on DPX task-induced events were performed on the patients,and the cognitive function of all subjects was assessed using MATRICS set of recognized neurocognitive function tests.Pearson correlation was used to analyze the correlation among behavioral data,CNV amplitude and cognitive function.The receiver operating characteristic(ROC)curve and the area under the curve(AUC)to evaluate the predictive value of CNV index combined with cognitive function indicators for CS.Results A total of 110 subjects were included,comprising 50 CS patients as the study group and 60 participants from health checkups as the control group.The error rate of ERP task EAX,EAY and EBX in the study group was higher than that in the control group,and EBX-AY was lower than that in the control group.The continuous operation 2D,continuous operation 3D,continuous operation 4D,continuous mean,connection test,maze test,symbol coding score and the absolute value of CNVA,CNVB and CNVB-A amplitude of CP3,CP4,CPZ sites,and the absolute value of CNVB-A amplitude of CP3 sites in the study group were lower than those in the control group,the differences were statistically significant(P<0.05).In the study group,CP3 site CNVA and EBX-AY,CNVB-A and EAX,EBX,EBX-AY and continuous operation 2D were positively correlated,EAY and continuous operation 2D,continuous mean,EBX and continuous operation 3D,line test,symbol coding were negatively correlated,CP3 site CNVA was positively correlated with EAY and EBY,CPZ site CNVA and EBY,CPZ site CNVA and continuous operation 2D,CPZ site CNVB and wired test were negatively correlated in the study group,and the above comparisons were statistically significant(P<0.05).ROC curve analysis showed that the AUC values of continuous operation 2D,continuous operation 3D,continuous operation 4D,continuous mean,maze test and symbol coding score amplitude diagnostic CS were greater than 0.7,which were 0.826,0.838,0.773,0.875,0.870 and 0.933,respectively.Conclusion The CNV wave amplitude induced by the DPX task is correlated with the cognitive function of CS patients.It can be used as an objective neurophysiological index to evaluate the degree of cognitive function impairment in CS patients,providing a theoretical basis for the guidance and treatment of CS in the future.

Sinicization and reliability test of the Family Caregiver Care Stress Management Scale

Abstract:Objective To translate the Care Stress Management Scale(CSMS)in Family Caregiver into Chinese,and to test its reliability and validity among the family caregivers of patients with dementia.Methods The scale was translated into Chinese according to the Brislin model and cross-cultural debugging was carried out.A convenience sampling method was used to administer the questionnaire to 110 family caregivers of dementia patients in Shiyan City from April 2023 to May 2024 to evaluate the reliability and validity of the scale.Results The Chinese version of the CSMS comprises 2 dimensions with a total of 8 items.The Cronbach's α coefficient of the overall scale was 0.862,the spilt-half reliability was 0.680,and the intraclass correlation coefficient value for test-retest reliability was 0.842[95%CI(0.647,0.934)].The item-level content validity index was 0.833~1.000,and the scale-level content validity index was 0.875.Exploratory factor analysis extracted two common factors that could explain 69.473%of the total variation,namely emotional-focused coping(4 items)and problem-focused coping(4 items).Conclusion The Chinese version of CSMS has good reliability and validity,which can be used to assess the ability of family caregivers of dementia patients to manage stress in caregiving.
