Research Progress of Indoleamine 2,3-Dioxygenase 1 in Gliomas
Abstract:Indoleamine 2,3-dioxygenase 1 (IDO1) has become a key driver in the occurrence and development of gliomas by depleting tryptophan in the tumor microenvironment, inhibiting effector T cell proliferation, and weakening natural killer (NK) cell function, thereby inducing tumor immune evasion. At the same time, IDO1 is also an important biomarker in glioma immunotherapy. In-depth exploration of the mechanisms of IDO1 and the development of novel IDO1 inhibitors have significant clinical value for the treatment of gliomas and improvement of patient prognosis. This article comprehensively elaborates the multiple roles of IDO1 in gliomas, including its function as a rate-limiting enzyme in tryptophan metabolism, and how IDO1 activation promotes tumor immune evasion and is associated with poor prognosis. Meanwhile, this article also reviews the research progress of IDO1 inhibitors in glioma treatment, especially the potential advantages of IDO1/DNA dual-target inhibitors in reducing cytotoxicity caused by alkylating agents. This review aims to provide new research directions and treatment strategies for glioma therapy.
