Establishment and preliminary application of NanoPCR detection method for hepatitis A virus in shellfish aquatic products

Abstract:HAV is one of the important pathogens causing acute hepatitis in humans.Shellfish become the carrier of most foodborne viruses by accumulating viruses in aquatic environments.In order to understand the pollution of HAV in shellfish,effectively prevent the spread of HAV,and ensure consumer food safety,it is necessary to develop a simple,highly sensitive,and specific HAV detection method.This study combines nanomaterials and PCR technology to establish a novel HAV NanoPCR detection method by optimizing the reaction system and conditions.The results showed that compared with traditional PCR,the NanoPCR method established in this study has higher sensitivity,detecting at least 9.0 copies/μL,which is 10,000 times more sensitive than traditional PCR;This method has no cross reactivity to other shellfish pathogens and has good specificity.Further testing was conducted on 264 shellfish samples,and the detection rate of NanoPCR positive samples was 9.47%,while the detection rate of PCR detection method was 7.58%,indicating that the sensitivity of NanoPCR detection method is higher than that of traditional PCR detection method.Therefore,the HAV NanoPCR detection method is suitable for detecting samples of food products,which can be widely used for rapid detection of HAV in aquatic products,providing technical support for protecting human health.
