Study on colloidal gold test strip for rapid detection of melamine residues in milk

Abstract:In order to develop a fast immunochromatography assay for melamine detection in milk, colloidal gold coupled with specific melamine antibody, the concentration of coating antigens, the amount of antibody la-beled colloidal gold and goat anti-mouse antibody were optimized in our study. The results show that the strip presented a best overcome when the concentration of coating antigen was 0.25 μg/mL, the amount of antibody labeled colloidal gold was 9.0 μL/strip, the dilution rate of goat anti-mouse antibody was 1:100. Under the optimal conditions, the limit of detection is 0.25 μg/mL, and the results of the test could be read by naked eyes within 5 min. Milk sample could be analyzed directly without any treatment. The developed methods in this study suit for onsite testing of melamine in milk.
