The Effect of Remifentanil on PI3K/AKT/mTOR Signaling Pathway Regulation of ox-LDL-induced Vascular Smooth Muscle Cell Proliferation,Apoptosis and Migration

Abstract:Objective:To explore the effect of remifentanil on the phosphatidylinositol 3 kinase(PI3K)/protein kinase B(AKT)/mammalian target of rapamycin(mTOR)signaling pathway in regulating the proliferation,apoptosis and migration of vascular smooth muscle cells(VSMCs)induced by oxidized low-density lipoprotein(ox-LDL).Methods:VSMCs were divided into the normal control group(untreated),ox-LDL group(50 mg/L ox-LDL),L-remifentanil group(0.5 ng/mL remifentanil),M-remifentanil group(5.0 ng/mL remifentanil),H-remifentanil group(50.0 ng/mL remifentanil),and 740Y-P group(50.0 ng/mL remifentanil+10 μmol/L PI3K activator 740Y-P).The Cell Counting Kit 8(CCK-8)kit was used to detect the proliferation of VSMCs.The flow cytometer was used to detect the apoptosis of VSMCs.The scratch assay was used to detect the migration of VSMCs.The Transwell method was used to detect the invasion of VSMCs.The protein immunoblotting method(Western Blot)was used to detect the protein expression levels of PI3K,phosphorylated phosphatidylinositol 3-kinase(p-PI3K),AKT,phosphorylated protein kinase B(p-AKT),mTOR,and phosphorylated mammalian target of rapamycin(p-mTOR).Results:Compared with the control group,the OD value,scratch healing rate,invasion number,p-PI3K/PI3K,p-AKT/AKT,and p-mTOR/mTOR protein expressions in the ox-LDL group increased,while the apoptosis rate decreased(P<0.05).Compared with the ox-LDL group,the OD values,scratch healing rate,invasion number,protein expressions of p-PI3K/PI3K,p-AKT/AKT,and p-mTOR/mTOR in the L-refrinixin group,M-refrinixin group,and H-refrinixin group decreased,while the apoptosis rate increased(P<0.05).Compared with the H-refrinaxin group,the OD value,scratch healing rate,invasion number,expression of p-PI3K/PI3K,p-AKT/AKT,and p-mTOR/mTOR proteins in the 740Y-P group increased,while the apoptosis rate decreased(P<0.05).Conclusion:Remifentanil can further inhibit the proliferation and migration of smooth muscle cells and promote their apoptosis by suppressing the PI3K/AKT/mTOR signaling pathway.
