Effect of Salidroside on TLR4/NF-κB Signaling Pathway in Atherosclerotic Mice

Abstract:Objective:To explore the effects of Salidroside(SAL)on the Toll-like receptor 4(TLR4)/nuclear factor-κB(NF-κB)pathway and inflammatory factors in atherosclerotic(AS)mice.Methods:C57BL/6J mice were used as the control group.After modeling,ApoE-/-mice were randomly divided into model group,low-dose SAL(SAL-L,50 mg/kg)group,high-dose SAL(SAL-H,80 mg/kg)group,and the atorvastatin(10 mg/kg)group.The mice in control group and model group were given the same volume of normal saline by gavage.Hematoxylin-eosin(HE)staining,Oil Red O staining,and immunohistochemical staining were used to observe the atherosclerotic plaque in mice.The contents of serum lipids and inflammatory factors in mice,as well as the expressions of TLR4 and NF-κB in aortic tissue were detected.Results:Compared with the control group,the model group showed increased aortic sinus plaque area,lipid content and inflammatory infiltration;the levels of serum total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),interleukin-1β(IL-1β),and tumor necrosis factor-α(TNF-α)significantly elevated,while the level of high-density lipoprotein cholesterol(HDL-C)significantly decreased;the expressions of TLR4 and NF-κB in aortic protein significantly increased(all P<0.01).Compared with the model group,the area of atherosclerotic plaques,lipid content and inflammatory infiltration in the intervention group reduced,and the contents of serum TC,TG,IL-1 β,and TNF-α decreased,while the content of HDL-C increased.The levels of TLR4 and NF-κB in aortic tissue significantly decreased(all P<0.01).The above indicators in the SAL-H group and the atorvastatin group were better than those in the SAL-L group,but there was no statistically significant difference between the two groups(P>0.05).Conclusion:SAL improve blood lipid levels and reduce inflammation in AS mice,with more pronounced effect in the SAL-H group.SAL may attenuate the inflammatory response in AS mice by inhibiting the TLR4/NF-κB pathway.
