Gastrodin Inhibits Pyroptosis in MCAO Rats by Regulating the lncRNA Meg3/miR-485/AIM2 Pathway Via Exosomes

Abstract:Objective:To investigate the mechanism by which gastrodin inhibits pyroptosis in middle cerebral artery occlusion(MCAO)rats through the long non-coding RNA(lncRNA)maternally expressed gene 3(Meg3)/miR-485/absent in melanoma 2(AIM2)pathway via exosomes,thereby providing an experimental basis for early intervention with traditional Chinese medicine during the acute stage of cerebral infarction.Methods:SD rats were divided into control group,sham operation group,model group,and gastrodin group.The model group and the gastrodin group were subjected with MCAO surgery.Gastrodin at a dose of 100 mg/(kg·d)was administered intraperitoneally.SD rats were sacrificed on days 1,3,and 7.The ischemic hemisphere and serum were collected.Exosomes were extracted from the serum.The levels of NOD-like receptor protein 3(NLRP3)and AIM2 in the ischemic hemispheres were detected by Western blotting and immunohistochemistry.The levels of NLRP3 and AIM2 mRNA,as well as lncRNA Meg3 and miR-485,in the ischemic hemispheres of rats in each group were detected by quantitative real-time polymerase chain reaction(qPCR).Exosomes were isolated from peripheral plasma,and the levels of lncRNA Meg3 and miR-485 were measured by qPCR.Results:Western Blot results showed that the contents of NLRP3 and AIM2 in the ischemic hemisphere of gastrodin group were lower than those in the model group(P<0.05).The immunohistochemical results showed the same trend.The qPCR results showed that the mRNA levels of AIM2 in the ischemic hemisphere of rats in the gastrodin group on the 1st,3rd,and 7th days after cerebral infarction were all lower than those in the model group(P<0.05 or P<0.01).The lncRNA Meg3 level was also lower than that in the model group(P<0.05 or P<0.01),while the miR-485 level was higher than that in the model group(P<0.05 or P<0.01).The content of lncRNA Meg3 in serum exosomes of the ginsenoside group was lower than that of the model group on the 1st,3rd,and 7th days after cerebral infarction(P<0.05 or P<0.01),while the content of miR-485 was higher than that of the model group(P<0.01).Conclusion:Gastrodin probably inhibits pyroptosis in MCAO rats by regulating the lncRNA Meg3/miR-485/AIM2 pathway via exosomes.
