The Mechanism of Yiqi Yangyin Formula in Anti-atherosclerosis through SDF-1α/CXCR4 Axis

Abstract：Objective:To observe the effect of Yiqi Yangyin Formula on the expression of stromal cell-derived factor 1α(SDF-1α)and CXC chemokine receptor 4(CXCR4)in rats with atherosclerosis(AS).Methods:Forty-six Sprague Dawley(SD)rats were randomly divided into the normal group,the model group,the rosuvastatin calcium group,and the Yiqi Yangyin Formula group.The normal group was fed with basal diet,while the rats in the other groups were fed with high-fat diet combined with aortic intimal balloon strain to establish the AS model.After modeling at 56 days,the normal group and the model group were gavaged with normal saline,while the Yiqi Yangyin Formula group and the rosuvastatin calcium group were gavaged respectively with Yiqi Yangyin Formula and rosuvastatin calcium suspension.After 42 days of administration,atherosclerotic plaques in the abdominal aorta were collected as specimens.The pathological structural changes of the aortic tissue were observed by hematoxylin-eosin(HE)staining.Masson staining was used to observe the collagen fibrin condition of plaques.The expressions of SDF-1α and CXCR4 mRNA in plaque tissues were detected by reverse transcription polymerase chain reaction(RT-PCR).The levels of SDF-1α and CXCR4 protein in plaques were detected by Western Blot.The expression of serum monocyte chemoattractant protein-1(MCP-1)was detected by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the normal group,the expressions of SDF-1α and CXCR4 mRNA,and protein in the plaque tissue of the model group increased(P＜0.05).Compared with the model group,the levels of SDF-1α,CXCR4 mRNA and protein,and the expression of MCP-1 in the rosuvastatin calcium group and the Yiqi Yangyin Formula group were significantly improved(P＜0.05).Conclusion:Yiqi Yangyin Formula might play an anti-AS effect by regulating the expression of SDF-1α and CXCR4 in rats with AS.
