The Effect of Kangxian Yixin Formula on lncRNA-mRNA-ceRNA Regulatory Network in Patients with Dilated Cardiomyopathy

Abstract:Objective:To observe the effect of the Kangxian Yixin Formula on the ternary regulatory network of long non-coding RNA(lncRNA)-messenger RNA(mRNA)-competitive endogenous RNA(ceRNA)in patients with dilated cardiomyopathy,and to explore its mechanism of action on ventricular remodeling.Methods:Based on the prospective research method,peripheral blood samples were collected from 30 patients with dilated cardiomyopathy in the Western medicine group and the Kangxian Yixin Formula group after treatment for 6 months.Five samples in each group were randomly selected for ceRNA chip analysis to screen for differential lncRNA and mRNA before and after treatment to establish the ceRNA regulatory network.Venn analysis of differentially expressed lncRNA between groups was conducted to obtain the expression profiles of the co-acting lncRNA and the lncRNA acting exclusively on the Kangxian Yixin Formula.The key mRNA in the ceRNA regulatory network of the Kangxian Yixin Formula group were screened out,and the ceRNA regulatory networks were established respectively.Results:The results of differential gene screening revealed that the Western medicine group identified 802 differentially expressed lncRNA and 186 differentially expressed mRNA,while the Kangxian Yixin Formula group identified 395 differentially expressed lncRNAs and 89 differentially expressed mRNA.Venn analysis revealed 25 lncRNA acting together,and 370 lncRNA acting independently on the Kangxian Yixin Formula.Ultimately,five key lncRNA were involved(MSTRG.66929.5,NONHSAT187420.1,ENST00000382313.2,NONHSAT187211.1,lnc-FOXL2-5:4),potentially acting through pathways including dilated cardiomyopathy,Rap1 signalling,actin cytoskeleton regulation,and phosphoinositide 3-kinase(PI3K)/protein kinase B(AKT).Conclusion:The Kangxian Yixin Formula might improve myocardial fibrosis and inhibit ventricular remodeling in patients with dilated cardiomyopathy by regulating the ceRNA network mediated by MSTRG.66929.5,NONHSAT187420.1,ENST00000382313.2,NONHSAT187211.1,and lnc-FOXL2-5:4.
