The Effect of Cinnamic Acid on Pathological Pulmonary Remodeling in Left Heart Failure Mice

Abstract:Objective:To explore the intervention effect of cinnamic acid(CA)on pathological pulmonary remodeling secondary to left heart failure(LHF).Methods:The chronic LHF model in mice was established by aortic constriction(TAC)surgery.The C57BL/6J male mice were randomly divided into the sham operation(Sham)group,the model(TAC)group,the low-dose cinnamic acid(CA-L)group,and the high-dose cinnamic acid(CA-H)group.The degree of LHF was evaluated through transthoracic echocardiography analysis,cardiac mass index,lung mass index,and wheat germ agglutinin(WGA)staining.Hematoxylin-eosin(HE)staining was used to evaluate the pathological morphological changes of the lungs.The immunohistochemical analysis of α-smooth muscle actin(α-SMA)was used to evaluate the degree of muscleization of the pulmonary arterioles.Masson's trichrome staining was used to assess the degree of lung fibrosis.CD11b,F4/80,and Ly6G immunohistochemical analysis were used to evaluate the accumulation of inflammatory cells in the lungs.Results:CA can significantly improve the left ventricular function and left ventricular hypertrophy induced by TAC(P<0.05).CA can inhibit the thickening of alveolar septa,the focal collapse of lung tissue and the deposition of fibrous substances,the hypertrophy of pulmonary small arteries and the accumulation of inflammatory cells induced by TAC(P<0.05).Conclusion:CA can significantly improve the pathological pulmonary remodeling of heart failure mice,and suggests that it may improve the clinical prognosis of patients with end-stage HF.
