The Molecular Mechanism of Danshen-Andrographis in Treating Coronary Heart Disease Angina Pectoris Based on Network Pharmacology

Abstract:Objective:The molecular mechanism of action of Danshen-andrographis combination in treating coronary heart disease angina pectoris was explored based on network pharmacology.Methods:The active compounds of Danshen and andrographis paniculata were screened by the traditional Chinese medicine(TCM)system pharmacology database(TCMSP)database.The drug targets were obtained by UniProt database.The related targets of angina pectoris were searched in GeneCards database with"angina pectoris"as the key word.The drug targets and angina-related targets were input into Venny 2.1 software to obtain drug-disease common targets.STRING11.5 database was used to construct a protein-protein interaction(PPI)network diagram.The results of gene ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)signaling pathway enrichment analysis were obtained through the Metascape platform.The GO functional enrichment analysis histogram and the KEGG enrichment analysis bubble chart were drawn using microarray information.The drug-compound-target gene and drug-common target-pathway network diagrams were constructed using Cytoscape 3.9.1 software.Results:Finally,82 active chemical components,153 target genes,1 707 disease-related targets,and 91 common targets for drugs and diseases were identified.The key targets of Danshen-andrographis in treating angina pectoris were threonine protein kinase 1(AKT1),interleukin 6(IL6),tumor necrosis factor(TNF),transcription factor TP53(TP53),vascular endothelial growth factor A(VEGFA),estrogen receptor(ESR1),signal transducer and activator of transcription 3(STAT3),cysteine-aspartic acid protease 3(CASP3),and FOS oncogene(FOS).The intersection target gene enrichment analysis obtained 833 biological processes and 149 signaling pathways.Danshen-andrographis mainly acted on the phosphatidylinositol-3-kinase(PI3K)-protein kinase B(AKT),glycation end products and their receptors(AGE-RAGE),and interleukin-17(IL-17)signaling pathways.These involved regulatory processes such as cell proliferation and apoptosis,inflammatory responses,platelet activation,and vasodilation and vasoconstriction.Conclusion:The mechanism of Danshen-andrographis in the treatment of coronary heart disease angina pectoris may involve inhibiting vascular inflammatory responses and platelet activation,regulating the proliferation and apoptosis of vascular endothelial and smooth muscle cells,and dilating the coronary arteries,thereby exerting the therapeutic effect on coronary heart disease angina pectoris.
