The Effect of Lidocaine on Cardiomyocytes Injury and Autophagy Induced by Ischaemia-reperfusion through Regulating the Pink1/Parkin Signaling Pathway

Abstract:Objective:To observe the effect of lidocaine on cardiomyocytes injury and autophagy induced by ischemia-reperfusion(I/R)through regulating the Pink1/Parkin signaling pathway.Methods:The logarithmic H9c2 cells were divided into the NC group(normal culture),I/R group,low-lidocaine group(0.5 mmol/L lidocaine before I/R treatment),medium-lidocaine group(2.5 mmoVL lidocaine before I/R treatment),high-lidocaine group(5.0 mmoVL lidocaine before I/R treatment),and high-lidocaine+Pink1/Parkin signaling pathway activator(3-MA)group(high-lidocaine group+5 mmoVL 3-MA).The proliferation ability of H9c2 cells,the levels of malondialdehyde(MDA),superoxide dismutase(SOD),lactate dehydrogenase(LDH),mitochondrial autophagy and mitochondrial membrane potential(MMP)in each group were detected.The expression of Pink1/Parkin pathway proteins and autophagy-related proteins were detected by Western Blot.Results:Compared with the NC group,the cell proliferation inhibition rate,LDH,MDA levels,rabbit anti-microtubule-associated protein light chain 3 Ⅱ/Ⅰ(LC-3 Ⅱ/Ⅰ),Pink1,Parkin protein expressions in the I/R group increased,while the MMP,SOD levels and silkworm isolate protein 1(p62)protein expression decreased(P<0.05).Compared with the I/R group,the cell proliferation inhibition rate,LDH,MDA levels,LC-3 Ⅱ/Ⅰ,Pink1,Parkin protein expressions in each lidocaine groups decreased,while the MMP,SOD levels and p62 protein expression all increased(P<0.05),and lidocaine effect showed a dose-dependent(P<0.05).Conclusion:Lidocaine could inhibit mitochondrial autophagy under VR conditions and prevent H9c2 cells from being damaged by oxidative stress.The mechanism might be related to the inhibition of the activation of the Pink1/Parkin signaling pathway.
