Abstract:This review summarizes the role and prospects of SIRT1 (Sirtuin 1) in traumatic brain injury (TBI). Traumatic brain injury refers to brain dysfunction caused by direct or indirect external violence to the head. Under the combined effects of primary and secondary injuries, it may lead to severe neurological dysfunction, with a high mortality and disability rate. Therefore, the protection and recovery of brain function in TBI patients are particularly important. As a nicotinamide adenine dinucleotide-dependent histone deacetylase, SIRT1 can deacetylate histones and non-histone substrates, thereby regulating various cellular biological processes, such as gene transcription, inflammatory responses, metabolism, apoptosis, oxidative stress, and autoimmunity. SIRT1 plays an important role in preventing secondary brain injury induced by TBI and may become a new therapeutic target to reduce secondary injury after cranial trauma.
