Abstract:This review summarizes the biological characteristics of high mobility group box 1 (HMGB1) and its role in the pathophysiological mechanisms of traumatic brain injury (TBI), aiming to provide insights for research and treatment of TBI. TBI is a serious disease that poses a major threat to human health and social economy, characterized by high incidence, high disability rate, high mortality, and poor prognosis. HMGB1 is a non-histone DNA-binding protein that participates in various life activities such as intracellular DNA recombination, repair, and gene transcription regulation. In recent years, increasing studies have found that HMGB1 plays an important role in the pathogenesis of TBI. As an early damage-associated molecular pattern (DAMP), HMGB1 participates in multiple pathological processes after TBI, including ischemic brain injury, inflammatory response, coagulation disorders, formation of cerebral edema, and secondary brain injury.
