Study on the Rat Animal Model of Calcified Aortic Valve Stenosis Induced by Guidewire Injury of the Aortic Valve Under Ultrasound Guidance

Abstract:Objective:To establish the rat model with aortic valve stenosis by direct mechanical injury on the aortic valve of rats.Methods:After one week of adaptive rearing,the rats were randomly divided into the sham operation group,the mild model group,the moderate model group and the severe model group,with 10 rats in each group.Under ultrasound guidance,the guide wire was inserted into the left ventricle of SD rats through the right common carotid artery,causing aortic valve injury.The left ventricular cardiac function and aortic valve stenosis of rats in each group were detected by ultrasound.Hematoxylin-eosin(HE)staining was used to observe the changes in valve thickness.Alizarin red staining was used to detect calcium salt deposition in valves.The expression of macrophages,osteopontin(OPN),and α-smooth muscle actin(α-SMA)positive cells in specimens of each group was detected by immunohistochemical staining method.The expression of bone morphogenetic protein-2(BMP-2)in each group was detected by Western Blot.Results:Eight weeks after modeling,in the mild model group,moderate model group and severe model group,the left ventricular ejection fraction(LVEF),left ventricular short-axis shortening rate(LVFS),and effective aortic valve orifal area(AVA)decreased,while the peak aortic valve flow velocity and aortic valve cross-valve pressure gradient(AVPG)increased,with severe injury(P<0.05).Compared with sham operation,the aortic valves of rats in the mild model group,moderate model group,and severe model group,thickness,calcium salt deposition increased,CD68 infiltration increased,the expressions of OPN and α-SMA positive cells increased,and the expression of BMP2 protein increased,and the changes in severe injury were the most significant(P<0.05).Conclusion:The rat model of aortic valve calcification by guide wire injury simulated the characteristics of human aortic valve stenosis,including increased peak velocity and AVPG of the arterial valve,reduced effective orifitate area of the aortic valve,thickening of the aortic valve,with fibrosis,macrophage infiltration,and calcium salt deposition.
