Effects of miRNA-183 Overexpression on Apoptosis and Immune Response in LPS-induced H9c2 Cardiomyocytes

Abstract:Objective:To explore the effects of miRNA-183 on apoptosis and immune response in lipopolysaccharide(LPS)-induced H9c2 cells.Methods:The apoptosis of H9c2 cells was induced by LPS.On this basis,a miRNA-183 overexpression H9c2 cell line(LPS+miRNA-183)and its negative control(LPS+mimic-NC)were constructed.The levels of myocardial injury markers,as well as the levels of cell proliferation,apoptosis,and inflammation-related proteins and mRNA were detected by enzyme-linked immunosorbent assay(ELISA),CCK8,flow cytometry,Hoechst staining,Western Blot,and reverse transcription-polymerase chain reaction(RT-PCR),respectively.Results:The miRNA-183 lowly expressed in H9c2 cells induced by LPS.Overexpression of miRNA-183 could inhibit the increase of cardiac troponin Ⅰ(cTnⅠ),creatine kinase-MB(CK-MB),and myohemoglobin(Mb),promote cell proliferation,and reduce the apoptosis rate and the formation of apoptotic bodies.Overexpression of miRNA-183 could inhibit the increase of LPS-induced Cleaved Caspase 9,Cleaved Caspase 3,Bax/Bcl-2 ratio,interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),TNF-α mRNA,and IL-6 mRNA levels,and increase the level of interleukin-10(IL-10)mRNA.Conclusion:The overexpression of miRNA-183 can inhibit the apoptosis of LPS-induced H9c2 cells,and improve LPS-induced inflammatory response.
