Medication Rule and Mechanism of Traditional Chinese Medicine Compound Formula for the Treatment of Tachyarrhythmia

Abstract：Objective:To analyze medication rule of traditional Chinese medicine compound formula for the treatment of tachyarrhythmia in the national patent database,and to explore the potential mechanism of action of core drugs.Methods:The patents with traditional Chinese medicine compound formula for the treatment of tachyarrhythmia were collected from the National Patent Database,and the data were unified and standardized for drug efficacy,frequency of traditional Chinese medicine,nature,taste,and channel entry.The association rules,cluster analysis and factor analysis were performed.Drug components,targets and disease targets were obtained from TCMSP and OMIM databases,respectively.After the intersection targets were selected,the protein-protein interaction(PPI)network map was constructed by STRING and Cytoscape.The core targets were screened by topological analysis.The Kyoto Encyclopedia of Genes and Genomics(KEGG)pathway enrichment analysis was performed by DAVID.Results:A total of 177 prescriptions and 473 traditional Chinese drugs were included.The top drugs in terms of efficacy frequency were tonifying deficiency drugs,invigorating blood and dissolving stasis drugs,clearing heat drugs and calming the mind drugs.The most frequent drugs were licorice,salvia miltiorrhiza,and ophiopogon.The medicinal nature was mainly warm,cold and flat.The medicinal taste was mainly sweet,bitter,and Xin.The main channel entry were heart,liver,lung,and spleen.A total of 14 drug combinations were obtained by association rule analysis.The five kinds of results were obtained by clustering.Six common factors were obtained by factor analysis.A total of 229 core drug active ingredients,393 targets,2 262 disease targets and,122 intersection targets were obtained through network pharmacological analysis.PPI network topology were used to screen 7 core targets,involving cell senescence,advanced glycosylated end-product receptor(AGE-RAGE)signaling pathway,tumor necrosis factor(TNF)signaling pathway,and FoxO signaling pathway.Conclusion:The traditional Chinese medicine compound formula for the treatment of tachyarrhythmia in the national patent database was formulated according to the principles of boosting Qi and enriching Yin,clearing heat and invigorating blood,and nourishing the heart and calming the mind.The combination drugs such as licorice,salvia miltiorrhiza,ophiopogon,sour jujube kernel and Coptis coptis were often used in clinic.Drugs focus on core targets such as cylic adenosine monophosphate(cAMP)-dependent protein kinase catalytic subunit α(PRKACA),RAC-α serine/threonine protein kinase(AKT1),FoxO signaling pathway,cell aging pathway,and other mechanisms for treating tacharrhythmia by inhibiting reentry,reducing abnormal automaticity,reducing post-depolarization,inhibiting cardiac remodeling and inflammation.
