The Potential Clinical Application of Vericiguat through the Research Progress of sGC Agonists

Abstract:The nitric oxide (NO)-soluble guanylyl cyclase (sGC)-cyclic guanosine monophosphate (cGMP) signaling pathway plays an indispensable role in the cardiovascular system, participating in various physiological and pathological processes. sGC is a key enzyme in the NO-sGC-cGMP signaling pathway, and a series of drugs targeting sGC have emerged in recent years. In 2009, the World Health Organization (WHO) added the suffix '-ciguat' to this class of compounds. Various clinical applications of sGC agonists have been discovered, such as improving pulmonary arterial hypertension, renal function, metabolic syndrome, coronary artery spasm, non-alcoholic steatohepatitis, treating osteoporosis, and attenuating platelet activation. However, some other potential clinical applications are still under experimental exploration. Currently, the novel oral sGC stimulator Vericiguat has been approved for the treatment of heart failure, but there are still other biological functions that remain to be explored. As an sGC agonist, it is speculated that Vericiguat's functions go beyond the treatment of heart failure. By summarizing the research progress of sGC agonists, we aim to explore the potential clinical applications of Vericiguat, thereby expanding its drug indications.
