The Mechanism of Modified Gualou Xiebai Banxia Decoction for Treating Coronary Heart Disease Based on Network Pharmacology and Molecular Docking Technology

Abstract:Objective:To explore the mechanism of Modified Gualou Xiebai Banxia Decoction(MGXBD)for treating coronary heart disease(CHD)by network pharmacology and molecular docking techniques.Methods:The target and active ingredient of MGXBD were selected from the Traditional Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP),and CHD targets were searched from PharmGkb,GeneCards,OMIM,TTD,and DrugBank databases.The molecular mechanism of MGXBD was analyzed by potential target analysis,protein-protein interaction(PPI)network construction,enrichment analysis and other network pharmacological methods.Molecular docking technology was used to verify the binding of active ingredients and targets.Results:There were 145 active components of MGXBD and 163 targets related to CHD.The key active components of MGXBD included luteolin,quercetin,tanshinone ⅡA,and so on.The core targets included mitogen-activated protein kinase 3(MAPK3),mitogen-activated protein kinase 1(MAPK1)and heat shock protein 90α(HSP90AA1).Conclusion:Network pharmacology and molecular docking techniques revealed the mechanism of MGXBD for treating CHD.MGXBD could treat CHD through signaling pathways such as interleukin-17(IL-17),advanced glycosylated end-product receptor(AGE-RAGE),and tumor necrosis factor(TNF).
