Expression and Clinical Significance of OASL Gene in Low-grade Gliomas

Abstract:Objective:To search for new prognostic markers and therapeutic targets for low-grade glioma(LGG),and to study the expression of 2'-5'oligoadenylate synthetase-L(OASL)gene in LGG and its influence on the prognosis of LGG.Methods:Transcriptional and clinical data of LGG were downloaded from the cancer genome atlas database(TCGA)and analyzed for survival and clinical relevance in the microenvironment.Gene function were studied by gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis.The core genes related to prognosis were screened and combined with the genotype-tissue expression database(GTEx),single gene clinical correlation analysis of OASL gene was performed,and then gene set enrichment analysis was performed.The relationship between OASL gene and major immune cells was analyzed by immune cell infiltration.Spearmen correlation analysis was used to study the relationship between OASL gene and DNA methylation,tumor protein p53(TP53),isocitrate dehydrogenase 1(IDH1),O6-methylguanine-DNA methyltransferase(MGMT),serine/threoninekinase(BRAF)genes.The relationship between human protein atlas database(HPA)and glioma was verified by cell experiments and human protein atlas database.Results:Microenvironment analysis showed that tumor microenvironment was significantly correlated with LGG prognosis(P<0.05).OASL gene expression analysis showed that OASL gene was highly expressed in LGG and was significantly correlated with the prognosis of LGG(P<0.05).Immunoinfiltration analysis showed that OASL gene was positively correlated with B cells,natural killer(NK)cells,and T cells(P<0.05).Correlation analysis with common clinical prognostic indicators showed that OASL gene was positively correlated with TP53,IDH1,and MGMT,and negatively correlated with DNA methylation(P<0.05).HPA verification showed that OASL gene did not express in normal glial cells,but express in glioma cells.Cell experiments showed that knocking down OASL gene could inhibit tumor cell migration and promote tumor cell apoptosis.Conclusion:OASL gene is an important molecular marker of unfavorable prognosis of LGG and provides a new therapeutic target for the treatment of LGG.
