The Mechanism of Tanshinone ⅡA in Intervening Angiogenesis Based on Network Pharmacology and Molecular Docking Technology

Abstract:Objective:To explore the mechanism of tanshinone Ⅱ A in intervening angiogenesis by network pharmacology and molecular docking techniques.Methods:The Target of tanshinone Ⅱ A was predicted through the Swiss Target Prediction database.Primary targets for angiogenesis were obtained from Human Mendelian Genetics Online Database(OMIM),Therapeutic Target Database(TTD),GeneCards Database,and Pharmacopharmacology and Pharmacogenomics Database(PharmGKB).The protein-protein interaction(PPI)network was constructed using Cytoscape 3.9.1 software to search for core targets.The gene ontology(GO)function and the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment were analyzed using R4.3.0 software and metagenomic platform.Molecular docking and results analysis were performed using AutoDock Vina and PyMOL 2.5.5.Results:A total of 54 intersection targets of tanshinone Ⅱ A for angiogenesis intervention were obtained.The topological analysis of PPI network showed that interleukin 6(IL-6),tumor necrosis factor(TNF),hypoxia-inducing factor-1A(HIF-1A),tumor protein P53(TP53),and vascular endothelial growth factor A(VEGFA)were the key targets.GO functional analysis suggested that tanshinone Ⅱ A was mainly involved in the positive regulation of protein serine/threonine kinase activity,the regulation of MAP kinase activity,and mesenchymal development.KEGG enrichment analysis included phosphatidylinositol 3-kinase-protein kinase B(PI3K-AKT)pathway,mitogen activated protein kinase(MAPK)pathway,HIF-1 pathway,TNF pathway,interleukin-17(IL-17)pathway,etc.Molecular docking showed that the binding activity between the core target and the active ingredient was better.Conclusion:Tanshinone Ⅱ A interferes with angiogenesis through multiple targets and pathways,among which the targets VEGFA,TNF,HIF-1A,PI3K-AKT pathway,and HIF-1 pathway may be the focus of research in the mechanism of tanshinone Ⅱ A promoting angiogenesis.
