Experimental Study on the Detection of Cardiomyocytes Apoptosis Induced by H2O2Based on High-content Screening

Abstract:Objective To investigate the advantages and disadvantages of high-content screening(HCS)for detecting cardiomyocyte apoptosis,and to provide reference for experimental methods for apoptosis detection.Methods The cardiomyocyte apoptosis rat model was induced by different concentrations of H2O2.FITC Annexin V Apoptosis Detection Kit and Hoechst probe were used for staining. The HCS instrument Opetta system was used to detect and analyze cardiomyocyte apoptosis related indicators.The cell viability was determined by T Cell Counting Kit-8(CCK-8)method to determine the degree of cardiomyocyte apoptosis.Results The model of apoptosis induced by H2O2at 300μmol/L was the most ideal.The early apoptotic cells accounted for 61.4% and the late apoptotic cells accounted for 38.2%.The microscopic images showed obvious apoptosis.Conclusion The method of H2O2-induced cardiomyocyte apoptosis is universal and practical.HCS method is simple and effective.The data obtained by single detection is rich,and it can be used for high-throughput screening.It can be used for apoptosis detection of a large number of cell samples.
