Experimental Research on Local Inflammatory Reaction of Hypertensive Heart and Intervention with Qingda Granule

Abstract:Objective The aim of this study was to investigate the anti-inflammatory effect of Qingda granules(QDG)on the heart of spontaneously hypertensive rats(SHR),and to provide a theoretical basis for antihypertensive and prevention of target organ damage. Methods Twelve 17-week-old male SHR were randomly divided into model group(SHR-control group,n=6)and Qingda granule group (SHR+QDG group,n =6).Six Wistar-Kyoto(WKY)rats were used as blank control group(WKY group).The blood pressure of the rats was monitored weekly using a non-invasive blood pressure monitor for 8 weeks.Serum interleukin(IL)- 1β,IL-6,and tumor necrosis factor-α(TNF-α)levels were measured by enzyme-linked immunosorbent assay(ELISA).The cardiac pathological changes were observed by Hematoxylin-Eosin(HE)staining.Masson staining was used to detect cardiac collagen expression.Results There was a significant difference in systolic blood pressure(SBP)and diastolic blood pressure(DBP)between the SHR control group and SHR+ QDG group after treatment(P <0.05).The levels of serum IL-1βand IL-6,TNF-αin the SHR+QDG group were significantly lower than those in the SHR-control group(P <0.05).Compared with SHR-control group,the degree of myocardial inflammation and fibrosis in SHR+QDG group was significantly lower(P <0.05).Conclusion QDG can significantly reduce the blood pressure in SHR,and reduce the level of inflammatory and fibrosis of hypertension,which may be related to its mechanism of effectively reducing blood pressure to prevent target organ damage.The specific anti-inflammatory mechanism remains to be further studied.
