Effects of Neural Stem Cells Transplantation and Renshen Chuanxiongqin Injection on the Expression of BDNF Protein in Rats after Cerebral Infarction

Abstract:Objective To observe the effects of neural stem cells(NSCs)transplantation and Renshen Chuanxiongqin injection(RCI) on brain-derived neurotrophic factor(BDNF)expression in rats after cerebral infarction.Methods NSCs were cultured in vitro,sixty healthy Sprague-Dawley(SD)rats were selected,and the model rats of cerebral infarction was established.The rats were randomly divided into cerebral infarction group,NSCs transplantation group and combination group.The number and migration of bromode-oxyuridine(BrdU)labeled NSCs in cerebral infarction area in each group were detected by immunofluorescence assay.The changes of neural function were observed in rats before transplantation and after transplantation 1 week,2 weeks,3 weeks.After two weeks of transplantation,the expression level of BDNF gene was detected by reverse transcription PCR(RT-PCR)method,and the expres-sion level of BDNF protein was detected by western blot method.Results Detection results by immunofluorescence assay showed that the number of NSCs labeled by BrdU in combined group was significantly higher than that in NSCs group,the difference was sta-tistically significant (P<0.05).The neurological function of the combined group was significantly better than that in the cerebral in-farction group and the NSCs group by Basso,Beattie,and Bresnahan Locmotor Rating Scale(BBB)Score,and the difference was statistically significant(P<0.05).RT-PCR test showed that the expression level of BDNF gene in combination group was signifi-cantly higher than that in cerebral infarction group and NSCs group.Western blot test results showed that the expression levels of BDNF protein in combination group was significantly higher than that in cerebral infarction group and NSCs transplantation group(P<0.05). Conclusion NSCs transplantation and RCI can promote the expression of BDNF mRNA and protein in rats with cerebral infarction and improve neurological function recovery after cerebral infarction.
