The Influence of Gandou Fumu Decoction on TGF-β1/Smad Signaling Pathway in TX Mice with Hepatic Fibrosis

Abstract:Objective To observe the effect of Gandou Fumu decoction (GFD)on transforming growth factor-β1(TGF-β1)/Smad signaling pathway in TX mice with hepatic fibrosis,and to explore its mechanism of anti-hepatic fibrosis.Methods Twenty-four male TX mice were randomly divided into model group,penicillamine group and GFD group.Eight DL mice were as normal control group.The levels of serum hyaluronic acid(HA),laminin(LN),precollagen typeⅢ(PCⅢ)and collagen type Ⅳ(C-Ⅳ)were detected by competitive radioimmunoassay.The expression of TGF-β1,Smad2 and Smad7 were observed by immunohistochemistry.Results Compared with the normal control group,the serological indexes of hepatic fibrosis in the model group were significantly increased (P<0.01).Compared with the model group,the serological indexes of hepatic fibrosis in each treatment group were decreased(P<0.05 or P<0.01).Compared with penicillamine group,the serological indexes of hepatic fibrosis in GDFMT group were reduced(P<0.05).Compared with the normal control group,the expression of TGF-β1 and Smad2 in the model group were significantly increased and the expression of Smad7 protein was significantly decreased (P<0.01).Compared with the model group,the expression of TGF-β1 and Smad2 in each administration group were significantly decreased and the expression of Smad7 protein was significantly in-creased (P<0.01).Compared with penicillamine group,the expression of TGF-β1 and Smad2 in GDFMT group were decreased and the expression of Smad7 protein was significantly increased(P<0.05).Conclusion GFD can reduce the serological indexes of he-patic fibrosis in TX mice,which has a good anti-liver fibrosis.The mechanism may be related to the inhibition of TGF-β1 and Smad2 protein expression,up-regulation of Smad7 protein expression.
