The Effect of Leech Injection and Ischemic Postconditioning on the Expression of Phosphoinositide-3-kinase against Ischemia-reperfusion Injury in Rats

Abstract:Objective To observe the effect of leech injection and ischemic postconditioning (IPOC) on the expression of phosphoinositide-3-kinase (PI3K) mRNA and protein against ischemia-reperfusion (I/R) injury in rats.Methods Forty male Sprague-Dawly (SD) rats were randomly divided into four groups.:sham-operation (SO) group,middle cerebral artery occlusion (MCAO) group,IPOC group,and leech injection postconditioning (P-L) group.The IPOC and MCAO rat models were established by suture occlusion method.The volume of cerebral infarction was calculated by 2,3,5-Triphenyltetrazolium chloride (TrC) staining.The ex pression of PI3K mRNA and protein was detected by real-time fluorescence quantitative polymerase chain reaction (PCR) and western blot.Results The infarct volume of brain tissue was significantly lower in IPOC group than that in MCAO group after 24 h following I/R,which was lower in P-L group than that in IPOC group (P <0.05).The expression of PI3K mRNA and protein increased at 24 h following reperfusion in MCAO group (P <0.01).The expression of PI3K increased in IPOC group and P-L group (P <0.05 or P <0.01),which was higher in P-L group than that in IPOC group (P <0.05).Conclusion IPOC could protect neurons through upregulating the expression of PI3K.Leech injection postconditioning could augment the neuroprotection induced by IPOC.
