The Neuroprotection of Compound Tongluo Decoction on Cerebral Infarction in Rats and its Angiogenesis Mechanism

Abstract:Objective To evaluate the protective effect of compound Tongluo decoction (CTD) on cerebral infarction in rats,and to explore its possible mechanism.Methods The male Sprague-Dawley (SD) rats were randomly divided into 3 groups:sham-operation group,permanent middle cerebral artery occlusion (pMCAO) group,and pMCAO + CTD group.Neurological deficit score and brain infarct volume were assessed.Real time polymerase chain reaction (PCR) and Western blot were used to evaluate the level of vascular growth factors.lmmunofluorescence staining was used to determine the capillaries density in ischemic penumbra.Results Compared with pMCAO group,neurological deficit scores improved,and cerebral infarction volume reduced in pMCAO + CTD group (P <0.05).The expression of vascular endothelial growth factor (VEGF),angiopoietin 1 (Ang-1).and their related receptors were upregulated,while the density of CD31 labeled microvessels in ischemic cortex increased in pMCAO + CTD group (P < 0.05).Conclusion CTD provides the neuroprotection against cerebral ischemic injury through up-regulating the expression of VEGF,Ang-1,and its related receptors and promoting angiogenesis after cerebral infarction.
