Correlation of SERCA2a mRNA with Myocardial Injury,Arrhythmia,Syndrome Differentiation of Deficiency and Excess in Rats with Acute Myocardial Infarction

Abstract:Objective To investigate the correlation of sarco-endoplasmic reticulum calcium ATPase 2a (SERCA2a) mRNA with myocardial injury,arrhythmia,syndrome differentiation of deficiency and excess in rats with acute myocardial infarction (AMI).Methods Forty Sprague-Dawley (SD) rats were randomly divided into sham operation group,model group,Xintongfang group and Diltiazem control group.The myocardial infarction model was established by using ligation method.Expression of SERCA2a mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR) method.The arrhythmia was tested by electrocardiogram.The morphological changes were observed by Hematoxylin-Eosin (HE) staining.The syndrome differentiation of deficiency and excess was made by scoring criteria.Results The degree of myocardial injury was negatively correlated with SERCA2a mRNA expression(r=-0.780,P=0.00).Expression of SERCA2a mRNA in non-arrhythmia group was significantly increased (P<0.01).The expression of SERCA2a mRNA was positively correlated with syndrome differentiation score (r=0.969,P=0.01).Conclusion SERCA2a mRNA has protective effect on myocardial tissue,and SERCA2a mRNA expression is associated with arrhythmia and syndrome differentiation of deficiency and excess.
