Effects of Naloxone,Epinephrine and Vasopressin on Cytokines in Resuscitation Rats

Abstract:Objective To investigate the effects of naloxone,epinephrine and vasopressin on cytokines in resuscitation rats,and to explore the possible mechanism.Methods One hundred healthy wistar rats,weight in 280 g to 320 g,were randomly divided into five groups:sham operation group (group A,n=20), epinephrine group (group B,n=20),epinephrine plus vasopressin group (group C,n=20),epinephrine,vasopressin plus naloxone group (group D,n=20) and control group (group E, n=20).Rats were treated with anesthesia and tracheal intubation in group A.Rats in group B, groupC, group D and group E were successfully established to cardiac arrest model,adrenaline 0.2 mg/kg,vasopressin 0.8 U/kg and naloxone 0.4 mg/kg and normal saline in tail vein were injected, respectively.The changes of mean arterial pressure (MAP) in 30 min after spontaneous circulation recovery in group B,group C and group D were recorded.The levels of tumor necrosis factor alpha (TNF-α), interleukin (IL)-6 and IL-10 were detected by immunohistochemical method.Results Compared with group E,there were no significant difference in asphyxia time between group B, group C and group D (P>0.05).Compared with group C and group B,there was a significant difference in the success rate of spontaneous circulation in group D (P<0.05).When 30 min after return of spontaneous circulation (ROSC), MAP had an upward trend in grup B,group C,group D.Compared with group B, MAP increased more significantly in group C and group D (P<0.05).At the same time, there was a significant difference in the levels of TNF-α,IL-6 and IL-10 between group E and group B,or group C,or group D (P<0.05).Conclusion During cardiopulmonary resuscitation (CPR),naloxone,epinephrine and vasopressin can significantly increase the rate of spontaneous circulation restoration.At the same time, arterial pressure maintained at a relatively high level in rats after the recovery.Naloxone,epinephrine and vasopressin can reduce the levels of serum proinflammatory cytokines,improve the levels of serum anti inflammatory cytokines in rats.
