Effects of Gastric Bypass Surgery on Plasm Ghrelin,Glucagon Peptide-1 and Peptide YY in Rats with High-fat Diet-induced Obesity

Abstract:Objective To the effect of Roux-en-Y gastric bypass surgery(RYGB on plasm ghrelin,glucagon peptide-1 (GLP-1) and peptide YY (PYY) in rats with high-fat diet-induced obesity.Methods Thirty Wistar rats were randomly divided into three groups:normal diet group(ND group) fed with normal diet,high-fat diet group(HFD group)fed with high-fat diet,and high-fat diet plus RYGB group(HFD plus RYGB group)fed with high-fat diet for 12 weeks.After 12 weeks,rats underwent sham operation in ND group and HFD group,and RYGB in HFD plus RYGB group.Rats were sacrificed after 4 weeks of operation.The concentrations of plasm ghrelin,glucagon peptide-1 (GLP-1) and peptide YY (PYY) were detected by ELISA.Results The body weights were higher in HFD group than that in ND group(P<0.05).Compared with HFD group,body weights decreased in HFD plus RYGB group (P<0.05).The concentrations of plasm ghrelin,peptide YY were higher in HFD group than that in ND group(P<0.05).Compared with HFD group,the concentrations of plasm ghrelin,GLP-1 increased while peptide YY concentration decreased in HFD plus RYGB group (P<0.05).Conclusion RYGB can reduce body weights,regulate the gut hormone ghrelin,GLP-1 and peptide YY levels in rats with high-fat diet-induced obestiy.
