The Effects of Mfn-2 Expression and PI3K-Akt Signaling Pathway in Reduction of Myocardial Ischemia-reperfusion Injury by Sevoflurane Postconditioning in Rats

Abstract:Objective To research the protection function of myocardial ischemia-reperfusion injury by sevoflurane postconditioning in rats and the role of Mfn-2 expression and PI3K-Akt signaling pathway.Methods Thirty Sprague-Dawley rats were randomly divided into 5 groups (n=6 earch):group S,group MIRI,group SP,group SP+W,and group W.After anesthesia and fix the rats,except group S,the rest of the four groups would be ligation tight the left anterior descending branches for 30 minutes,then renew blood flow for 120 minutes.Group SP should inhale 2.3% sevoflurane for 15 minutes,when it is 10 minutes before the blood flow renewed.Group SP + W should be given a 15 μg/kg diluented Wortmannin for 1 mL by vein injection before inhalation.Group W was given the diluented Wortmannin only before the blood flow renewed.Then cTn-I was measured by ELISA.The total Akt,P-Akt and Mfn-2 expression were measured by western blot.The ratio of the P-Akt and total Akt was calculated.The morphology changes of myocardial cell were observed under electron microscopy.Results The total Akt was closed in 5 groups,there was no statistically significant difference(P>0.05).Compared with group S,cTn-I and P-Akt/total Akt were increased statistically significantly in group MIRI,group SP,group SP+W,and group W(P<0.05).cTn-I in group SP was less than that in group MIRI,group SP+W,and group W(P<0.05).P-Akt and P-Akt/total Akt in group SP was more than that in group MIRI,group SP+W,and group W(P<0.05).There was no significant difference in aspects of cTn-I,P-Akt/total Akt among group SP+W,group MIRI and group W(P>0.05).Compared with group S,Mfn-2 were increased in the others(P<0.05).There was no significant difference in Mfn-2 between group SP and group SP+W(P>0.05).Neither the difference between group MIRI and group W,but the former two groups were smaller than the latter ones,there was a significant difference(P<0.05).It showed that there was no signs of damage in group S,while it shows a significantly damage lighter in group SP than other three groups with the electron microscope.Conclusion Sevoflurane postconditioning can alleviate myocardial ischemia-reperfusion injury in rats,probably via with the reduction of Mfn-2 expression and the activation of PI3K-Akt pathway,also,the former factor effects the latter one.
