Abstract:Objective To investigate the effect of paeonol pretreatment on nuclear factor κB (NF κB)in rats with cerebral ischemia/reperfusion (CI/R)injury.Methods Rat model with CI/R was prepared by four vessel occlusion for 10 min fol owed by 12 hour reperfusion.Rats were then randomly divided into 5 groups (n=10):sham,model,paeonol high, medium and low dose groups(20 mg/kg,10 mg/kg and 5 mg/kg,respectively).Paeonol was injected intraperitoneal y once a day for 7 days before the operation and once 1 h after the operation, respectively. The NF κB activities in brain tissue of CI/R gerbils were detected by immunohistochemis-try after 12 hour reperfusion.Resluts Paeonol (10 mg/kg and 20 mg/kg)evidently improved the neurological behavior scores in rats with CI/R injury, compared with that of model group (P<0.05).The IOD values of NF κB in paeonol groups (20 mg/kg,10 mg/kg and 5 mg/kg)were significantly lower than that of model group (P<0.05).Conclusion The effectiveness of paeonol pretreatment in rats with CI/R injury appears to be associated with the inhibition of NF κB in brain tissue.
