Effects of Sinoaortic Denervation on Mitochondrial Ca2+ and Membrane Potential of Nerve Cel in Rats

Abstract:Objective To explore the effects of sinoaortic denervation (SAD)on mitochondrial Ca2+ and membrane potential of nerve cel in rats. Methods Fifty Sprague Dawley (SD)rats were randomly divided into control group,sham operation group,30 day SAD group,60 day SAD group and 90 day SAD group. Mitochondrial membrane potential (by Laser scanning confocal micro-scope)and level of Ca2+(by flame atomic absorption spectrometry)were measured and compared between the five groups.Results There were no differences in blood pressure between the sham operation group and the control group. Compared with the control group,both systolic blood pressure and diastolic blood pressure in three SAD groups fluctuated obviously within 24 hours.The level of Ca2+ of mitochondrion was elevated significantly(P<0.01)and mitochondrial membrane potential was decreased significantly(P<0.01)in the three SAD groups compared with that in the control group and sham operation group. And in the three SAD groups,the level of Ca2+ of mitochondrionwas gradual y increased and mitochondrial membrane potential was gradual y decreased(P<0.01). Conclusion The fluctuation of blood pressure caused by sinoaortic denervation,which is a significant damage to the mitochondria of brain cel s,and is becoming more and more serious with time.It is very important to maintain the stability of blood pressure.
