Influence of Ditan Decoction on Inflammatory Cytokine in Rabbits with Atherosclerosis

Abstract：Objective To observe the effects of Ditan decoction (DTD)on nuclear factor κB (NF κB),E selectin,tumor necrosis factor α(TNF α)and interleukin 6 (IL 6)in rabbit with atherosclerosis,and to explore its mechanism of DTD on atherosclerosis. Methods Forty eight healthy male New Zealand white rabbits were randomLy divided into blank group,atherosclerosis model group,low,medium and high dose DTD group,and atorvastatin group.The rabbits in blank group were fed with common forage.The rabbits in other groups were fed with high lipid forage.After twelve weeks,rabbits were sacrificed to measure serum TNF αand IL 6 by ELISA.Morphological changes in aorta were observed under the light microscope.The mRNA expressions of TNF αand IL 6 were detected by PCR,while the protein expressions of NF κB and E selectin were detected by western blot.Results Compared with the control group,the TNF α,IL 6,NF κB and E selectin decreased significal y in DTD groups.Conclusion DTD can reduce the expressions of inflammatory factors TNF α,IL 6,NF κB and E selectin,and it may be one of the mechanisms to prevent and treat atherosclerosis.
