The Influence on Rapamycin Management Against Myocardial Ischemia/Reperfusion Injury in Rats

Abstract:Objective To investigate the protective effects of rapamycin (RPM)on myocardial ischemia/reperfusion (MI/R)injury in rats.Methods Rat model of MI/R injury was prepared by coronary artery ligation for 30 min fol owed by 120 min reperfusion.Then rats were divided into 3 groups at random (n=10),namely sham,model,RPM group (5 mg/kg).RPM was administered by intragastric gavage once a day for 3 days pre operation.The creatine phosphokinase (CK),lactate dehydrogenase (LDH)and superoxide dis-mutase (SOD)activities,and the contents of malondialdehyde (MDA)in sera were examined,and the area of myocardial infarction was calculated after the reperfusion.Results RPM significantly reduced the area of myocardial infarction,compared to that of model group (P<0.05).The CK and LDH activities in RPM group were significantly lower than those of model group (P<0.05 or P<0.01). RPM evidently elevated the SOD activity (P<0.01),and reduced the MDA contents in sera (P<0.01).Conclusion The effective-ness of RPM pretreatment in MI/R injury appears to be associated with the inhibition of lipid peroxidation.
