Optimum Concentration of Folate Inducing Mesenchymal Stem Cells into Neurons in Rats
Abstract:Objective To probe the optimum concentration of folate inducing mesenchymal stem cells into neurons in rats.Methods Human mesenchymal stems cells were isolated primarily from bone marrow of adult volunteers and purified sterilely with gradient centrifugation by Ficoll.The low-density cells including MSCs were cultivated.The 5th passage of MSCs were planted into plastic Petri dishes and induced with BME,DMSO,BHA and edaravone.The control group did not receive any induced treatment.Neuron-specific enolase (NSE) and glial fibrillary acidic protein (GFAP) were detected by means of MTT.Results After induction for 24h,the differentiated cells showed typical neural morphology in all groups,but the numbers of neuron and astrocyte in medum concentration group was more than that in other groups.The rate of neuron in medum concentration group was more than that in the other groups.Conclusion The result suggested that 40 mg/L folate was the best concentration for MSDCs differentiated into neurons.
