Effect of Procyanidin on Permeability of Blood - brain Barrier and Free Radical Content in Rats with Cerebral Ischemic Reperfusion Injury
Abstract:Objective To investigate the effect of procyanidin on the permeability of the blood - brain barrier and brain water con-tent and the contents of free radical in rats with cerebral ischemia reperfusion(I/R) injury. Methods Rats were randomly divided into four groups:Sham operated group,I/R gronp, procyanidin low dosage group and proeyanidin high dosage group. The focal middle cerebral artery occlusion(MCAO) model was made by suture- occluded method. After 90 rain MCAO following 24 hours of reperfusion, brain water content, the contents of Evens blue (EB), superoxide dismutase (SOD), malondialdehyde (MDA) and triphenyltetrazolium chloride(TTC)staining were observed. Results Compared with sham operated group, brain water content and the contents of EB were in-creased in I/R group. Compared with I/R group, brain water content and the contents of EB were decreased dose -dependently in pro-cyanidin low dosage group and procyanidin high dosage group(P<0.05). Compared with I/R group, the contents of MDA were de-creased dose- dependently in procyanidin low dosage group and procyanidin high dosage group, while the contents of SOD were in-creased(P<0.05). Compared with I/R group, cerebral infarction volume in procyanidin low dosage group and procyanidin high dosage group was decreased dose -dependently (P<0.05). Conclusion Procyanidin had protective effect on cerebral ischemia injury in rats, which might be associated with alleviating the permeability of the blood - brain barrier and oxidative damage.
