Effects of Xiaoqinglong Decoction,Maxinfu Decoction Combined with Yupingfeng Powder on TNF-α and IL-1β in Rats with Allergic Rhinitis Based on the NF-κB Signaling Pathway

Abstract:Objective To investigate the mechanism of Xiaoqinglong Decoction,Maxinfu Decoction combined with Yupingfeng Powder in treating allergic rhinitis(AR)by observing their effects on serum levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in AR rats based on the nuclear factor-κB(NF-κB)signaling pathway.Methods Ninety clean-grade healthy male Wistar rats(weight 200-240 g)were purchased from the Experimental Animal Center of Liaoning University of Traditional Chinese Medicine.The rats were divided into nine groups(10 rats per group)using a randomized block design according to body weight:blank control group,AR model control group,pyrrolidine dithiocarbamate ammonium(PDTC)intervention group,Forskolin intervention group,H89 dihydrochloride intervention group,loratadine treatment group,and low-,medium-,and high-dose groups of Xiaoqinglong Decoction,Maxinfu Decoction combined with Yupingfeng Powder.The AR model was established by systemic sensitization with ovalbumin followed by local challenge.The levels of TNF-α and IL-1β in the nasal mucosa of each group were measured.Experimental data were analyzed using one-way ANOVA with IBM SPSS Statistics 26 software.Results Serum levels of TNF-α and IL-1β were significantly elevated in the AR model control group.Compared with the model control group,the Forskolin intervention group,PDTC intervention group,and all dose groups of Xiaoqinglong Decoction,Maxinfu Decoction combined with Yupingfeng Powder significantly reduced TNF-α and IL-1β levels(P<0.05).In contrast,the H89 intervention group significantly increased the levels of these inflammatory factors(P<0.05).Compared with the low-dose group,the medium-and high-dose groups showed more significant reductions in TNF-α and IL-1β levels,with statistically significant differences(P<0.05).Conclusion Xiaoqinglong Decoction,Maxinfu Decoction combined with Yupingfeng Powder may exert therapeutic effects on AR by inhibiting the NF-κB signaling pathway,activating the cAMP-PKA signaling pathway,and reducing TNF-α and IL-1β levels in the nasal mucosa.The efficacy is dose-dependent,with medium-and high-dose groups showing superior effects to the low-dose group,reflecting the dose-response relationship in traditional Chinese medicine treatment.
