Study on the Effect of Kudzu Root Enzyme Complex on Improving Alcoholic Hepatitis

Abstract:Objective Based on the established optimal gavage dose,this study further investigated the pharmacological effects of the kudzu enzyme complex on alcoholic hepatitis in mice.Methods(1)Experimental grouping and administration:Fifty mice were randomly divided into a control group(Con),a model group(Mod),a low-dose kudzu enzyme complex group[JS(L)],a medium-dose kudzu enzyme complex group[JS(M)],and a high-dose kudzu enzyme complex group[JS(H)],with 10 mice per group.Dosing began on the fourth day of modeling:JS(L)15 mL/(kg·d),JS(M)30 mL/(kg·d),JS(H)60 mL/(kg·d),while the Con group received an equal volume of physiological saline,continuously for 14 days.After the experiment,mice were euthanized,and liver tissue and orbital venous plexus blood were collected.Detection indicators:calculation of liver index;observation of hematoxylin-eosin(HE)staining;Western blot detection of transforming growth factor-β(TGF-β),hypoxia-inducible factor-1α(HIF-1α),3-hydroxy-3-methylglutaryl-CoA reductase(HMG-CR),and sterol regulatory element-binding protein-1C(SREBP-1C)protein levels;measurement of alanine aminotransferase(ALT),aspartate aminotransferase(AST),albumin(ALB),and total bilirubin(TBIL)using assay kits.(2)Experimental grouping and administration:Forty mice were randomly divided into the Con group,the kudzu enzyme complex group(JS),the Mod group,and the treatment group(Mod+JS),with 10 mice per group,using the same modeling method as in(1).After the experiment,mice were euthanized,and liver tissue and orbital venous plexus blood were collected.Detection indicators:observation of HE staining;Western blot detection of cyclooxygenase(COX),catalase(CAT),Bcl-2-associated X protein(BAX),B-cell lymphoma-2(BCL-2),caspase-3,caspase-9,interleukin-6(IL-6),interleukin-1(IL-1),tumor necrosis factor-α(TNF-α),TGF-β,HIF-1α,HMG-CR,SREBP-1C,phosphorylated calcium/calmodulin-dependent protein kinase 2(P-CaMK2),and phosphorylated AMP-activated protein kinase(P-AMPK)protein levels;measurement of ALT,AST,ALB,and TBIL using assay kits.Results(1)Compared with the Mod group,the JS(L),JS(M),and JS(H)groups showed concentration-dependent changes in liver index,fibrosis,and steatosis;HE staining results indicated concentration-dependent improvement in the JS(L),JS(M),and JS(H)groups compared to the Mod group.(2)Compared with the Mod group,the Mod+JS group showed significant improvements in liver indicators,fibrosis,and steatosis;HE staining results revealed disordered liver tissue structure,significant inflammatory cell infiltration,and lipid vacuoles in the Mod group compared to the Con group,while the Mod+JS group exhibited marked improvement compared to the Mod group;Western blot results demonstrated that the Mod+JS group effectively reduced the expression levels of inflammatory factors(IL-1,IL-6,TNF-α)and apoptosis-related factors(Caspase-9,Caspase-3,TGF-β)while enhancing antioxidant levels(COX,CAT)in the livers of mice with alcoholic hepatitis;further detection showed significantly increased expression of P-AMPK and P-CaMK2 in the Mod+JS group compared to the Mod group.Conclusion The kudzu enzyme complex can ameliorate alcoholic hepatitis in mice by modulating the expression of oxidative stress markers,inflammatory factors,and apoptosis-related factors in the liver.
