Comparison on the Effect and Safety of Regimens Based on Different Treatment Courses of Daratumumab in the Treatment of Patients with Relapsed and Refractory Multiple Myeloma

Abstract:Objective:To explore and compare the efficacy and safety of different courses of daratumab-based regimens in the treatment of patients with relapsed/refractory multiple myeloma.Method:A total of 92 patients with relapsed/refractory multiple myeloma treated with daratumumab in Shijingshan Teaching Hospital of Capital Medical University from January 2023 to February 2025 were selected as the research subjects.They were divided into a short-course group and a long-course group according to different treatment courses,with 46 cases in each group.The short-course group was treated for 8 courses or less,while the long-course group was treated for 9 courses or more.The total effective rate of treatment,incidence of adverse reactions,progression-free survival(PFS),tumor markers before and after treatment,and prognosis-related indicators were compared between the two groups.Result:The total effective rate of treatment in the long-course group was significantly higher than that in the short-course group,and the PFS was longer than that in the short-course group,with statistically significant differences(P<0.05).There was no statistically significant difference in the incidence of adverse reactions between the two groups(P>0.05).There was no statistically significant difference in the comparison of tumor markers and prognosis-related indicators between the two groups before treatment(P>0.05).After treatment,the tumor markers and prognosis-related indicators in the long-course group were significantly lower than those in the short-course group,and the difference was statistically significant(P<0.05).Conclusion:The long-course daratumab-based regimen has a better therapeutic effect on patients with relapsed/refractory multiple myeloma,its safety is worthy of recognition,and its application value is relatively higher.
