STM2457 Reverses Cisplatin Resistance in Gastric Cancer Organoids by Demethylation of m6A

Abstract:Objective:To explore the relationship between m6A methylation and Cisplatin resistance in gastric cancer,and to find ways to reverse its resistance.Method:In this study,gastric cancer tissues sensitive and resistant to Cisplatin treatment were collected,single cells were isolated and made into organoids.Different concentrations of Cisplatin or different concentrations of STM2457 were used to incubate organoids with Cisplatin to observe the growth and vitality of organoids;the methylation level of m6A in different tissues and organs were detected,and the expression levels of methyltransferase-like 3(METTL3)in different organs were detected by quantitative real-time polymerase chain reaction(qRT-PCR)and Western blot.Result:Compared with normal gastric cancer cells and organoids,the sensitivity of the drug-resistant group to Cisplatin was significantly reduced,and the m6A methylation level and METTL3 expression level were significantly increased.STM2457 can restore the sensitivity of drug-resistant organs of gastric cancer to Cisplatin,and reduce the level of m6A methylation and the expression level of METTL3.Conclusion:STM2457 reversed Cisplatin resistance in gastric cancer organoids by demethylation of m6A.
